Fig 1: BxPC-3 / BxPC-3-luc Orthotopic Mouse Model for Pancreas Cancer preclinical research.

The BxPC-3 / BxPC-3-luc orthotopic mouse model engrafts a KRAS wild-type human pancreatic adenocarcinoma directly into the pancreatic parenchyma, offering a genetically distinctive backdrop for preclinical pancreas cancer research. Alfa Cytology brings this model to life end to end — qualified reporter-cell supply, precision orthotopic surgery, repeated in vivo bioluminescence imaging, protocol-driven dosing, and exhaustive terminal analysis — so your therapeutic candidates can be interrogated in a biologically faithful setting.

Overview of BxPC-3 / BxPC-3-luc Orthotopic Mouse Model for Pancreas Cancer

BxPC-3 is a human pancreatic adenocarcinoma line established in 1986 from the primary tumor of the pancreatic body in a 61-year-old female patient, with moderate differentiation and no evidence of metastasis at the time of sampling. Its most cited distinguishing trait is a wild-type KRAS allele — an unusual feature among PDAC lines — alongside mutant TP53 (p.Tyr220Cys) and reported CDKN2A and SMAD4/DPC4 alterations. This genotype makes BxPC-3 an essential counterpoint to KRAS-mutant lines such as PANC-1 or MIA PaCa-2, and it is frequently described as one of the most gemcitabine-sensitive PDAC lines in comparative panels. The cells display epithelial, glandular morphology, secrete mucin, carcinoembryonic antigen (CEA), and pancreas-specific antigens, and form xenografts in nude mice that histologically and biochemically resemble the patient's original tumor — 100% take (5/5) within three weeks after subcutaneous inoculation of 1×107 cells.

The BxPC-3-luc derivative adds stable firefly luciferase expression, enabling non-invasive, longitudinal quantification of orthotopic tumor burden after D-luciferin administration — tracking primary tumor expansion in the pancreas as well as peritoneal seeding and distant metastasis in the same animal over time. Orthotopic implantation of BxPC-3 into the pancreatic tail or body has been widely adopted in in vivo programs studying KRAS-independent tumor biology, gemcitabine and combination chemotherapy, EGFR and TGF-β pathway targeting, nanoparticle-based drug delivery, and cancer stem cell–directed interventions.

Our Services

Alfa Cytology operates the BxPC-3 / BxPC-3-luc orthotopic model under a defined preclinical quality system: cells are STR-confirmed, mycoplasma-tested, and luciferase-validated before surgery; trained surgeons implant defined cell doses into the pancreatic body or tail; and the study proceeds through scheduled in vivo imaging, randomized dosing, interim observations, and terminal necropsy with full histopathology — all summarized in a structured, decision-ready report.

Workflow of BxPC-3 / BxPC-3-luc Orthotopic Mouse Model Construction

Model construction proceeds through a standardized, auditable sequence of stages, each with clear acceptance criteria to secure consistent engraftment and dependable in vivo data.

  1. Cell-line preparation and QC: BxPC-3 or BxPC-3-luc cells are expanded from authenticated, low-passage stocks in RPMI-1640 with 10% FBS; STR identity and mycoplasma status are verified, and cells are harvested in exponential phase.
  2. Reporter validation (BxPC-3-luc): Stable luciferase expression is confirmed in vitro by luminescence assay, and signal uniformity is documented prior to implantation.
  3. Animal readiness: Female immunocompromised mice (commonly BALB/c nude or NOD/SCID, 6–8 weeks old) are acclimated, health-screened, and randomized according to the approved animal-use protocol.
  4. Orthotopic implantation: Under anesthesia with sterile technique, the pancreas is exposed via a left-flank or midline laparotomy; a single bolus of 1×106–5×106 BxPC-3 / BxPC-3-luc cells (with or without Matrigel) is injected into the pancreatic body or tail under visual guidance, and the abdominal wall is sutured closed.
  5. Post-surgical care and baseline imaging: Mice are monitored for recovery, wound integrity, and body weight; baseline bioluminescence images are captured after intraperitoneal D-luciferin injection to confirm engraftment.
  6. Scheduled monitoring phase: Tumor burden is followed at defined intervals by in vivo bioluminescence imaging, alongside body-weight and clinical-condition scoring to capture early toxicity signals.
  7. Randomized treatment: When tumor signals meet pre-set thresholds, animals are randomized into vehicle and treatment arms and dosed per the client-defined regimen, with optional interim sampling for pharmacodynamic measurements.
  8. Terminal analysis and reporting: At endpoint, pancreatic tumors and any metastatic lesions are excised, weighed, and imaged ex vivo; tissues are fixed for H&E and immunohistochemistry (e.g., Ki67, cleaved caspase-3, α-SMA), and all data are compiled into a comprehensive study report.
