A20 Syngeneic Model Service for Lymphoma

Fig 1: A20 Syngeneic Model for Lymphoma preclinical research.

The A20 syngeneic model service for lymphoma is an immune-competent BALB/c platform built on the classic spontaneous B-cell lymphoma line A20, supporting subcutaneous, systemic, and cell-therapy study formats that mimic human diffuse large B-cell lymphoma biology and checkpoint responsiveness. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated A20 / A20-luciferase expansion and banking, subcutaneous, intravenous, or intraperitoneal inoculation with validated preconditioning protocols, scheduled caliper and in vivo bioluminescence monitoring, and full lymphoma histopathology with B-cell and checkpoint immunophenotyping — all configured around each sponsor’s antibody, checkpoint, CAR-T, or combination hypothesis.

Overview of A20 Syngeneic Model for Lymphoma

A20 is among the most venerable and most immunologically characterized tumor lines in all of cancer research. Established in 1979 by Kim and colleagues at Sloan-Kettering from a spontaneous reticulum cell neoplasm found in an aged BALB/cAnN mouse, the line was deliberately selected and subcloned for B-cell properties, and it remains the canonical syngeneic model of B-cell non-Hodgkin lymphoma — pathologically mimicking human diffuse large B-cell lymphoma. Its genetics are reassuringly simple for a tumor line: a near-diploid karyotype with a modal chromosome count of 37, and molecular cytogenetic analyses that mapped its imbalances onto the human genome to suggest a profile reminiscent of sporadic Burkitt’s lymphoma. Immunologically, A20 is a fully equipped B cell — surface immunoglobulin positive (with expression that is medium-dependent, low in Click’s medium and abundant in RPMI-1640), MHC class II positive (I-A+) and MHC class I positive on the H-2d haplotype, Fc-receptor positive, and capable of presenting both alloantigens and protein antigens — features that make its interactions with T cells, antibodies, and Fc-effector pathways unusually faithful.

Optical and PET imaging of in situ CAR-T generation in the A20 syngeneic lymphoma model — aCD5 LNP-mCAR19 mRNA vector design, naive mouse IL-7 priming and LNP injection timeline, longitudinal bioluminescence of spleen/liver/lymph nodes over 24 h, ex vivo organ imaging, H&E/FLuc/CD4/CD8 histology of spleen showing CAR-T localization and T-cell infiltration after in situ engineering. (Zhang, Nisi, et al., 2025)

In vivo, A20 is remarkably versatile. Subcutaneous implantation of 2 × 105 cells into BALB/c mice produces solid, measurable tumors within about a week to ten days; intravenous infusion at 5 × 105 cells generates a disseminated lymphoma infiltrating spleen, liver, bone marrow, lymph nodes, and — pathognomonically — the meninges, where tumor growth produces hindlimb paralysis that serves as the model’s humane endpoint; intraperitoneal inoculation yields ascites and peritoneal disease. A published lymphoreplete systemic protocol uses a single 100 mg/kg cyclophosphamide dose one day before cell infusion to improve a historically variable take rate without inducing lymphodepletion. The model’s therapeutic track record is correspondingly broad: partial responsiveness to PD-1 and CTLA-4 blockade, efficacy of anti-CD20 and anti-PD-L1 antibodies, adoptive T-cell transfer and bispecific studies, CAR-T evaluation through the A20-luc-GFP system, and landmark mechanistic work in which Treg depletion at a single tumor site eradicated disease at distant sites through global anti-cancer immunity. A20-OVA-Luc, presenting defined ovalbumin epitopes on both MHC classes, rounds out the platform as a precision tool for antigen-specific immunity. For sponsors in hematologic immuno-oncology, A20 remains the first-line, fully benchmarked B-cell lymphoma system.

Our Services

A lymphoma platform this heavily benchmarked demands nothing less than benchmark-grade execution, and Alfa Cytology delivers it: authenticated A20 and reporter banks with immunophenotype re-verification, all three validated inoculation formats with the published cyclophosphamide-assisted systemic protocol, scheduled caliper and in vivo bioluminescence monitoring, and endpoint pathology tuned to B-cell lymphoma — all under one accountable preclinical operation.

Workflow of A20 Syngeneic Model Construction

Construction follows the published methodologies for each deployment format: A20 or A20-luc cells are expanded under authenticated conditions, implanted subcutaneously, intravenously, or intraperitoneally into BALB/c mice with or without cyclophosphamide preconditioning, monitored by caliper and bioluminescence, treated, and analyzed with lymphoma-appropriate endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: A20 or reporter derivatives are identity-verified, confirmed mycoplasma-negative, and re-checked for B-cell immunophenotype (surface immunoglobulin, MHC class II), then expanded into low-passage working banks under standardized RPMI-1640 culture conditions.
  2. Cell harvest and formulation: Log-phase cultures are counted and viability-checked, then resuspended at the study-specified dose: approximately 2 × 105 cells in 100 μL PBS for flank grafts, 5 × 105 cells in 100 μL for systemic models, or up to 2 × 106 cells intraperitoneally.
  3. Preconditioning and inoculation: BALB/c mice receive cyclophosphamide one day before cell infusion where the study design requires improved take (100 mg/kg lymphoreplete) or lymphodepletion (200 mg/kg); cells are implanted subcutaneously, infused intravenously via tail vein, or injected intraperitoneally.
  4. Establishment monitoring and randomization: Flank tumors are measured by caliper from first palpability; systemic arms are confirmed by in vivo bioluminescence after luciferin administration before randomization into balanced treatment groups.
  5. Treatment administration: Experimental arms — checkpoint inhibitors, CD20/CD19-directed antibodies, bispecifics, CAR-T or adoptive cell products, cytokines, and combinations — proceed per study design with scheduled body-weight and toxicity monitoring.
  6. Longitudinal monitoring: Caliper measurements or weekly bioluminescence sessions track tumor kinetics; animals are monitored closely for hindlimb paralysis and pathological burden as the model’s humane endpoints.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth inhibition or survival with ex vivo organ mapping (spleen, liver, bone marrow, lymph nodes, ovaries, meninges), flow-cytometric burden quantification in reporter arms, and histopathology (H&E confirming DLBCL-like sheets of lymphoid cells, plus CD19, CD20, MHC-II, Ki67, CD8, and PD-L1 immunohistochemistry).

Case Study-A20 Syngeneic Model Development

Alfa Cytology has conducted preclinical programs on the A20 platform spanning checkpoint combination evaluation, B-cell-directed antibody and CAR-T benchmarking, and disseminated-disease burden tracking across inoculation formats. In these studies, implanted A20 tumors were monitored through scheduled caliper and in vivo imaging measurements, treated per randomized regimens, and characterized at endpoint by histopathology with B-cell and checkpoint immunophenotyping.
