U266 / U266-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: U266 / U266-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The U266 / U266-luc orthotopic mouse model for multiple myeloma engrafts a rare IgE-secreting, BCMA-positive human myeloma into the bone compartment of immunodeficient mice, where marrow disease, spinal involvement, and osteolytic bone loss can be quantified by bioluminescence and microCT. Focused exclusively on preclinical oncology, Alfa Cytology delivers this xenograft platform as a complete service: authenticated U266 / U266-luc cell supply, intra-tibial or systemic orthotopic inoculation into NOG or NSG hosts, scheduled imaging paired with serum Igλ monitoring, and terminal bone microCT and marrow flow cytometry.

Overview of U266 / U266-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

U266, also catalogued as U266B1, belongs to the pioneering generation of human myeloma lines. Established in 1968 from the peripheral blood of a 53-year-old man with refractory, terminal IgE-secreting myeloma—and reported by Nilsson and colleagues in 1970—the line is catalogued by ATCC as TIB-196 and by DSMZ as ACC-9. Its rarest distinction is its immunoglobulin: U266 secretes IgE with a λ light chain, making it the prototype IgE myeloma system and a fusion partner of choice for human hybridoma production. The cells are EBV-negative, express IL-6 without requiring it for growth, show BCL2 mRNA, and carry the plasma cell surface program: CD56, CD44, syndecan-1 (CD138), and BCMA positive, while B-lineage markers CD19 and CD20 are absent. Their HLA-A2 positivity has made them a target line for antigen-specific T-cell concepts. With a population doubling time around 55 hours and a complex hypodiploid karyotype with polyploid elements, U266 is a slow, deliberate line—and a fixture of the LL-100 blood cancer panel and MD Anderson Cell Lines Project.

Luciferase-expressing human U266, KMS-26, KMS-12-BM, and MM.1S multiple myeloma cells (stably transduced via lentivirus) were inoculated into NSG mice via the tail vein (disseminated model), and therapeutic efficacy was assessed weekly using IVIS. (Zhang, Yufei, et al., 2026)

In vivo, U266 has one of the most instructive engraftment stories in the myeloma field. Intravenously injected into NOG mice, it homes to bone marrow and produces hind-limb paralysis with cortical osteolytic lesions and trabecular bone loss—while the same cells fail to show any myeloma features in SCID or NOD/SCID hosts, a host-dependence lesson that shaped modern NSG usage. Standard formats include intravenous injection of 106 cells into 175-cGy-irradiated NSG mice, where marrow infiltration of roughly 35% is documented at six weeks, and the landmark intra-tibial protocol in NOD/SCID-γcnull mice in which 2 × 106 luciferase-tagged U266 cells in 20 µl produce a first bioluminescent signal by week three, spread to pelvis, contralateral femur, and spine, and culminate in spinal deformity and hind-limb paralysis at weeks eight to nine—with MRI confirming tumor volumes and serum Igλ rising in parallel. This model has validated bortezomib, melphalan, the aminopeptidase inhibitor tosedostat, and the HDAC inhibitor CHR-3996, and has supported microCT-based Osteolytica lesion analysis and TGF-β receptor I kinase inhibitor studies. Figure 1 outlines how this orthotopic system is assembled and monitored.

Our Services

A U266 / U266-luc engagement with Alfa Cytology is run as one continuous program: verified cell banking, host-matched orthotopic inoculation (intra-tibial for bone-confined disease or systemic for dissemination) into NOG or NSG recipients, IVIS imaging paired with serum Igλ curves, dosing per the client’s schedule, and a terminal package combining microCT bone analysis, marrow CD138+ flow quantification, and histology.

Workflow of U266 / U266-luc Orthotopic Mouse Model Construction

Building a U266 or U266-luc orthotopic myeloma model hinges on host selection and inoculation route: the intra-tibial format confines disease to bone with reproducible spinal progression, while systemic injection demands the most immunodeficient recipients. The workflow below reflects this dual-format execution.

  1. Cell Bank Verification and Expansion: U266 or U266-luc stocks are authenticated by STR profiling and mycoplasma testing, then expanded slowly in RPMI-1640-based medium at 0.2–1.0 × 106 cells/mL, with loose-adherent cells harvested by pipetting rather than trypsin.
  2. Luciferase Calibration (U266-luc Only): Signal output per cell is verified in vitro, and BCMA/CD138+ identity is confirmed by flow cytometry before in vivo use.
  3. Host Selection and Conditioning: Six-week-old NOG or NOD/SCID-γcnull mice are selected; NSG protocols apply 175 cGy total-body irradiation while the Fryer intra-tibial format uses unirradiated NOD/SCID-γcnull hosts; mice are randomized by body weight.
  4. Route-Matched Orthotopic Inoculation: For bone-confined disease, 2 × 106 cells in 20 µl RPMI-1640 are injected intra-osseously into the tibia under anesthesia; for disseminated questions, 106 cells are delivered intravenously, respecting the documented NOG/NSG requirement.
  5. Engraftment Confirmation and Longitudinal Imaging: Weekly IVIS BLI after D-luciferin (1.5 mg/mouse i.p., 9-minute delay) captures the first signal around week three, with spread to pelvis, contralateral femur, and spine charted thereafter; serial serum samples are assayed for human Igλ.
  6. Treatment Administration: Test agents—proteasome inhibitors, alkylators, epigenetic agents, or immunotherapeutics—are dosed per the client’s regimen; the landmark validation study treated from week five for four to six weeks.
  7. Surveillance and Survival Endpoint: Daily monitoring tracks body weight and hind-limb status; hind-limb paralysis, spinal deformity, or welfare criteria define the survival endpoint, typically weeks eight to nine in the intra-tibial format.
  8. Terminal Bone and Organ Analysis: At endpoint, MRI or microCT documents tumor volumes and trabecular bone loss (Osteolytica lesion analysis where applied); marrow is profiled by flow cytometry for CD138+ human cells, and spine, contralateral limbs, and organs are processed for H&E and immunohistochemistry.
