Hep G2 / Hep G2-luc Orthotopic Mouse Model Service for Liver Cancer

Fig 1: Hep G2 / Hep G2-luc Orthotopic Mouse Model for Liver Cancer preclinical research.

The Hep G2 / Hep G2-luc orthotopic mouse model of liver cancer implants a well-differentiated, hepatocyte-like human tumor directly into the hepatic lobe, where it recreates organ-authentic growth, vascular interaction, and locoregional spread while remaining fully quantifiable through luciferase bioluminescence and serum AFP. Alfa Cytology supports this model as a complete preclinical service — spanning authenticated Hep G2 banking, luciferase line release testing, subcapsular hepatic implantation in nude or NSG recipients, scheduled in vivo imaging, dosing regimens, and endpoint analysis combining tumor metrics, biomarkers, and histopathology.

Overview of Hep G2 / Hep G2-luc Orthotopic Mouse Model for Liver Cancer

Hep G2 is a human liver cancer cell line established in 1975 at the Wistar Institute from the tumor of a 15-year-old male patient and deposited with ATCC as HB-8065. Although originally recorded as a well-differentiated hepatocellular carcinoma, genomic re-examination led to its reclassification as hepatoblastoma-derived, a distinction supported by chromosome 4q losses, trisomies 2 and 20, a deletion of exon 3 of CTNNB1 activating the Wnt/β-catenin pathway, and a gene-expression profile matching pediatric-type liver tumors. What sets Hep G2 apart from most liver cancer lines is how much normal hepatocyte function it retains: the cells secrete alpha-fetoprotein, albumin, transferrin, fibrinogen, and a broad panel of other plasma proteins, display polarized membrane organization, and carry wild-type TP53 with an intact p53 pathway — making the line equally useful for hepatocyte-biology studies and for cancer research.

Orthotopically, Hep G2 cells are introduced beneath the capsule of a hepatic lobe, where they form discrete intrahepatic tumors that grow within the organ’s native perfusion, stromal, and matrix environment and can progress to abdominal dissemination in advanced stages. The luciferase-tagged variant, Hep G2-luc, converts tumor mass into a serially measurable photon flux after D-luciferin administration, enabling non-invasive in vivo quantification of tumor growth kinetics and response to intervention. Because Hep G2 tumors continue to secrete alpha-fetoprotein, circulating AFP can be tracked as a complementary serum biomarker alongside imaging — a dual-readout advantage that few liver models offer. This combination has made the Hep G2-luc orthotopic platform a workhorse for evaluating targeted agents, nanoparticle and antibody-drug conjugate delivery, oncolytic and gene therapies, and locoregional treatment concepts in a differentiated human liver tumor context.

Our Services

Our ServicesOur ServicesHep G2 rewards careful handling — its hepatocyte-like differentiation, wild-type p53 background, and AFP secretion are precisely the properties that studies depend on, and they are also the properties that sloppy cell management erodes. Alfa Cytology protects them by working from authenticated HB-8065 master banks at restrained passage numbers, qualifying Hep G2-luc for uniform reporter expression before release, and pairing subcapsular implantation with scheduled imaging plus serum AFP collection. Through every phase of the Hep G2 / Hep G2-luc orthotopic workflow, Alfa Cytology keeps the biology intact and the data traceable, so sponsors can trust what the model is telling them.

Workflow of Hep G2 / Hep G2-luc Orthotopic Mouse Model Construction

Building the Hep G2-luc orthotopic model joins hepatic microsurgery with imaging discipline and biomarker collection. The sequence below reflects the standard flow, with acceptance criteria at each step to keep cohorts comparable and animal welfare protected.

  1. Authentication and reporter release: Hep G2 / Hep G2-luc stocks are expanded from authenticated master banks at low passage, mycoplasma-tested, and released only after confirming uniform luciferase expression by in vitro luminescence assay.
  2. Inoculum preparation: Log-phase adherent cultures are trypsinized, washed, counted, and resuspended at the working dose (commonly 2×106 to 5×106 cells in 20-50 µL of PBS, optionally with a basement-membrane matrix).
  3. Recipient preparation: Nude or NSG mice are acclimatized, stratified by body weight, and prepared in a sterile surgical field with anesthesia, analgesia, and warming support.
  4. Subcapsular hepatic implantation: A small laparotomy exposes the liver, and the suspension is injected slowly beneath the capsule of the left lateral lobe; a pale blanch at the injection site confirms subcapsular placement, the site is checked for reflux, and the abdomen is closed in layers.
  5. Recovery and baseline measurements: Animals recover under thermal support and monitoring; baseline whole-body bioluminescence is acquired after D-luciferin administration, and a pre-treatment serum sample is collected for AFP measurement.
  6. Longitudinal imaging and dosing: Photon flux is captured on a fixed schedule together with serial AFP sampling, body weight, and clinical observations; cohorts are randomized by matched baseline signals before dosing begins.
  7. Endpoint analysis: At termination, intrahepatic tumors are excised and weighed, ex vivo imaging localizes intra- and extrahepatic deposits, serum AFP is quantified, and tissues are processed for histopathology and molecular analysis.

Case Study-Hep G2 / Hep G2-luc Orthotopic Mouse Model Development

Alfa Cytology has conducted preclinical programs on the Hep G2 orthotopic platform for sponsors advancing targeted agents, delivery technologies, and locoregional concepts against liver cancer. In these studies, scheduled bioluminescence imaging traced intrahepatic tumor expansion and treatment-induced regression over time, serial serum AFP measurements provided a complementary biomarker view of tumor burden, and terminal workups combined tumor weights, ex vivo organ imaging, and histopathological confirmation of lesion identity. Where programs required it, excised tumors were additionally profiled for pathway markers or processed for pharmacokinetic correlation. Study-specific designs and results remain sponsor-specific deliverables, and representative outlines can be made available on request.
