AsPC-1 / AsPC-1-luc Orthotopic Mouse Model Service for Pancreas Cancer

Fig 1: AsPC-1 / AsPC-1-luc Orthotopic Mouse Model for Pancreas Cancer preclinical research.

The AsPC-1 / AsPC-1-luc orthotopic mouse model grows a KRAS-mutant, metastasis-derived human pancreatic adenocarcinoma directly within the pancreatic parenchyma, reproducing the local invasion and abdominal spread typical of advanced pancreas cancer. Alfa Cytology delivers this model as a complete preclinical package — spanning authenticated reporter-cell supply, stereoscopic surgical implantation, serial in vivo bioluminescence readouts, customized dosing regimens, and terminal histopathology — built to support robust evaluation of chemotherapies, targeted agents, and emerging therapeutic modalities.

Overview of AsPC-1 / AsPC-1-luc Orthotopic Mouse Model for Pancreas Cancer

AsPC-1 is a human pancreatic adenocarcinoma cell line derived from the ascitic fluid of a 62-year-old patient and established through nude-mouse xenografts, placing it among the most aggressive and best-documented pancreas cancer lines in the NCI collection. Genetically, it carries the canonical PDAC driver constellation — mutant KRAS (G12D), inactivated TP53 with loss of p53 protein, and disrupted CDKN2A — alongside additional alterations such as MAP2K4 and, in some databases, SMAD4, GNAS, and RNF43, making it a faithful cellular proxy for late-stage, treatment-refractory disease. Its ascitic origin and poorly differentiated epithelial phenotype correlate with strong invasive behavior: in published head-to-head orthotopic comparisons, direct injection of 1×106 AsPC-1 cells into the mouse pancreas produced tumors in 100% of animals, among the highest take rates of the commonly used PDAC panel.

The AsPC-1-luc derivative introduces stable firefly luciferase expression, converting orthotopic tumor growth into a quantifiable, non-invasive optical signal after D-luciferin administration. This enables longitudinal tracking of primary pancreatic tumor expansion, early detection of peritoneal dissemination and distant metastases, and real-time measurement of treatment response in the same cohort — advantages that have made the AsPC-1 / AsPC-1-luc orthotopic system a workhorse in in vivo studies of gemcitabine-based regimens, nab-paclitaxel combinations, stromal-targeting strategies, KRAS-pathway inhibitors, and oncolytic virotherapy.

Alfa Cytology runs the AsPC-1 / AsPC-1-luc orthotopic model as a turnkey preclinical engagement. From mycoplasma-free cell expansion and luciferase signal verification through microsurgical implantation, scheduled in vivo imaging, treatment administration, and full necropsy with immunohistochemistry, every phase is documented under a defined quality framework so your team receives decision-ready data on time.

Workflow of AsPC-1 / AsPC-1-luc Orthotopic Mouse Model Construction

Construction of the AsPC-1 / AsPC-1-luc orthotopic pancreas cancer model is organized as a series of tightly controlled stages. Each step carries defined acceptance criteria to ensure reproducible engraftment and high-quality in vivo readouts.

  1. Cell qualification and expansion: AsPC-1 or AsPC-1-luc cells are revived from low-passage, authenticated stocks; STR identity and mycoplasma negativity are confirmed, and cultures are scaled up in RPMI-1640 with 10% FBS to the exponential phase.
  2. Luciferase confirmation (AsPC-1-luc): Reporter expression is validated in vitro by luminescence assay to guarantee uniform signal intensity prior to implantation.
  3. Mouse preparation: Female immunodeficient mice (e.g., BALB/c nude or NOD/SCID, 6–8 weeks old) are acclimated, health-checked, and randomized under an approved animal-use protocol.
  4. Orthotopic surgical implantation: Under anesthesia, a small left-flank or midline laparotomy exposes the pancreas; 1×106–5×106 AsPC-1 / AsPC-1-luc cells, optionally suspended in Matrigel, are injected into the pancreatic parenchyma or pancreatic tail using a fine needle under visual guidance, and the abdominal wall is closed in layers.
  5. Recovery and baseline imaging: Animals are monitored post-operatively for wound healing and body-weight recovery; baseline bioluminescence images are then acquired after intraperitoneal D-luciferin injection to verify successful engraftment.
  6. Longitudinal monitoring: Tumor burden is tracked at scheduled intervals by in vivo bioluminescence imaging, with body-weight and condition scoring recorded in parallel to flag treatment-related toxicity early.
  7. Randomized dosing phase: Once tumor signals reach pre-specified thresholds, mice are randomized into vehicle and treatment groups and dosed per the study plan, with optional interim imaging or plasma sampling for pharmacodynamic markers.
  8. Terminal analysis: At endpoint, primary pancreatic tumors, peritoneal deposits, and metastatic organs are harvested and weighed; ex vivo luminescence is captured, tissues are processed for H&E and immunohistochemistry (e.g., Ki67, cleaved caspase-3, α-SMA), and findings are summarized in a comprehensive study report.
