NCI-H929 / NCI-H929-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: NCI-H929 / NCI-H929-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The NCI-H929 / NCI-H929-luc orthotopic mouse model establishes the CD38-high reference human myeloma line — the substrate on which daratumumab-class antibodies, SLAMF7-directed agents, and BCMA bispecifics were preclinically validated — in the bone marrow and tissues of immunodeficient mice. Alfa Cytology delivers this Gazdar-curated, ATCC-patented line with authenticated banking, difficult-culture-aware expansion SOPs, subcutaneous or systemic luciferase-imaging configurations, and benchmark arms anchored to the published antibody and bispecific record, so your CD38, SLAMF7, BCMA, and proteasome programs are tested on the line the immunotherapy field itself trusts.

Overview of NCI-H929 / NCI-H929-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

NCI-H929 was established by Gazdar, Oie, Kirsch, and Hollis at the NCI from the malignant pleural effusion of a 62-year-old Caucasian woman with relapsed IgA-kappa myeloma, published in Blood in 1986 — and it arrived with unusual molecular baggage: a complex translocation that disrupts c-MYC regulation (mapped in detail in 1988), an activating NRAS allele (resolved by sequencing to p.Gly13Asp, heterozygous), near-tetraploid karyotype with 8q+ abnormalities, and microsatellite-stable DNA. Its immunophenotype is distinctly myelomatous: PCA-1, transferrin receptor, and CD38 positive, with HLA-DR and CALLA negative and EBV negative. CD38 expression is exceptionally high — higher than most MM lines — which is precisely why H929 became the reference substrate for the antibody era: daratumumab-class anti-CD38 agents demonstrated complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and direct apoptosis on H929 cells in vitro and tumor control in H929 xenografts in vivo, and the line features centrally in the NCI-H929 xenograft-based evaluation of immunomodulatory drugs and their combinations.

Antigen expression profiling across 28 MM patient samples and cell lines showing FcRH5 positivity (red) nearly universal (~100% plasma cells) versus heterogeneous BCMA (blue), with NCI-H929 confirmed as a dual FcRH5⁺/BCMA⁺ line in the flow cytometry panel (f, black histograms versus dashed isotype controls), establishing H929 as a reference antigen-positive MM model for CAR-T studies. (Jiang, Dongpeng, et al., 2023)

That reference status extends across modalities. H929 xenografts served in the development of SLAMF7-directed elotuzumab (whose ADCC activity was characterized against H929 cells with effector cells), and the BCMA-targeting bispecific T-cell engager AMG 420 cleared H929 tumors in disseminated NSG models, establishing the preclinical template later translated to patients. Operationally, H929 is among the most demanding myeloma lines in culture: DSMZ and curators document that cells must be maintained at no less than 1 × 106 cells/mL, show a 7–10 day growth lag after thawing, double roughly every 50–70 hours, and reach a maximum density near 1.5 × 106 cells/mL — requirements that make careless expansion a silent confounder. Placed orthotopically by systemic delivery or implanted subcutaneously for benchmarking, NCI-H929-luc adds quantitative whole-body bioluminescence in vivo for marrow-homing and disseminated designs, with monoclonal quality-controlled reporter banks. For programs targeting CD38, SLAMF7, BCMA, or the c-MYC/NRAS axis, H929 remains the line against which the field's claims were originally made.

Cell Line Information: NCI-H929 / NCI-H929-luc

NCI-H929 (ATCC CRL-9068 and the manufactured progeny CRL-3580; DSMZ ACC-163; ECACC 95050415; Cellosaurus CVCL_1600) is a patented line (US Patent 4,892,829) producing IgA-kappa paraprotein — though ATCC notes recent tests detect kappa light chain without the IgA heavy chain — and it grows as single round-to-oval cells in small clumps, doubling in roughly 50–70 hours. Its culture difficulty is documented across multiple curators and must be treated as an experimental variable: minimum density 1 × 106 cells/mL, post-thaw lag of 7–10 days, and slow early viability. The NCI-H929-luc derivative adds firefly luciferase for quantitative imaging in vivo, with monoclonal selection of high-signal clones in vitro recommended for uniform bioluminescence.

Our Services

H929 rewards operators who read the curator notes and punishes those who do not: drop below 1 × 106 cells/mL, skip the post-thaw recovery window, or over-expand and the line drifts before the study begins. Alfa Cytology runs NCI-H929 programs with density-disciplined expansion in vitro (minimum 1 × 106 cells/mL, 24–6-well recovery formats after thaw), batch-verified CD38-high status and c-MYC rearrangement, monoclonal luciferase quality control, benchmark arms anchored to the daratumumab-class and AMG 420 records, and ADCC-ready study designs with matched effector-cell formats — so the reference line arrives at the study in the condition the reference data assume.

Workflow of NCI-H929 / NCI-H929-luc Orthotopic Mouse Model Construction

NCI-H929 construction must honor the line's two signatures: culture difficulty and antibody-reference pharmacology. The workflow below covers density-disciplined preparation, the validated implantation configurations, and modality-matched endpoints.

  1. Density-Disciplined Cell Expansion and Quality Control: NCI-H929 or NCI-H929-luc cells are expanded in vitro in RPMI-1640 + 10% FBS + 0.05 mM 2-mercaptoethanol, maintained at no less than 1 × 106 cells/mL with 24-well or 6-well recovery formats after thaw; batches are confirmed mycoplasma-negative, STR-authenticated, verified for CD38-high expression, c-MYC rearrangement, and NRAS G13D status, with monoclonal luciferase populations confirmed for tagged lines.
  2. Host Selection and Acclimatization: BALB/c nude, NOD-SCID, NSG, or B-NDG mice, typically 6–8 weeks of age, are acclimatized under SPF conditions; disseminated designs may use sub-lethal irradiation to promote marrow engraftment.
  3. Configuration Selection and Inoculation: The study design selects subcutaneous flank implantation (typically 5–10 × 106 cells, with Matrigel where required) for localized benchmarking, or systemic intravenous delivery (classically 5–10 × 106 cells in ~200 µL) for marrow-homing disseminated disease, with implantation day recorded for the dosing calendar.
  4. Engraftment Verification and Randomization: NCI-H929-luc cohorts are imaged by bioluminescence in vivo after D-luciferin administration, with systemic configurations confirmed by long-bone and vertebral marrow signal; subcutaneous arms are staged by caliper, and animals are randomized at defined flux or volume thresholds.
  5. Treatment Administration with Modality Matching: Test articles are dosed on study-defined schedules — anti-CD38 or SLAMF7 antibodies (with or without effector-cell formats for ADCC arms), BCMA bispecifics, proteasome inhibitors, or IMiD combinations — with body weight and clinical condition monitored throughout.
  6. Longitudinal Monitoring: Disease burden is tracked weekly by bioluminescence quantification in vivo and caliper where applicable; ADCC-oriented studies add serial serum human immunoglobulin kappa light chain quantification as a tumor-burden correlate.
  7. Survival Follow-Up: Animals are followed to humane endpoints with Kaplan–Meier analysis, and disseminated designs monitor for hind-limb impairment from vertebral disease as a clinical signal.
  8. Endpoint Analysis and Reporting: At termination, tumors and femoral/tibial marrow are harvested; human myeloma cells are enumerated by CD38/CD138 flow cytometry, bone and soft-tissue infiltration is documented histologically, and tissues are processed for CD38 and BCMA immunohistochemistry (receptor-occupancy-style confirmation), cleaved caspase-3, and H&E; ex vivo bioluminescence maps signal distribution, and imaging, flow, pharmacodynamic, and survival data are compiled into the final report.
