PC-9 Metastatic Tumor Model Service for Lung Cancer

Fig 1: PC-9 Metastatic Tumor Model for Lung Cancer preclinical research.

The PC-9 Metastatic Tumor Model Service for Lung Cancer offers a well-characterized in vivo system for tracking the arrest, extravasation, and outgrowth of EGFR exon 19-deleted lung adenocarcinoma cells in immunodeficient mice. With hands-on experience across both experimental and spontaneous dissemination formats, Alfa Cytology structures every PC-9 study around the question your program needs answered — pairing authenticated cell banks, luciferase-enabled whole-body imaging, and metastasis-focused endpoints into one managed preclinical package.

Overview of PC-9 Metastatic Tumor Model for Lung Cancer

PC-9 is a human lung adenocarcinoma cell line that was established from a metastatic lymph node of a 45-year-old male patient, giving it a natural pedigree for dissemination research. The line carries an activating deletion in exon 19 of the EGFR gene (delE746-A750) and shows extreme sensitivity to first-generation EGFR tyrosine kinase inhibitors, with growth-inhibitory concentrations in the low nanomolar range. Morphologically heterogeneous — a mixture of round and spindle-shaped cells — PC-9 grows readily in standard culture and forms tumors in nude mice, and its origin from a metastatic lesion makes it particularly instructive for studying how adenocarcinoma cells survive in transit and colonize distant organs.

Fig 2: Reference figures for PC-9 cell-related literature.Fig 1. Optimized orthotopic multiple-injection (OOMM) versus traditional orthotopic model using PC9 cells: tumorosphere delivery via internal carotid artery with sustained perfusion enables robust initial colonization (day 12 hours: ~400 PC9 cells/clone) and progressively expanded brain colonies through day 14, significantly outperforming the traditional model that suffers from compromised BBB breakdown and poor clone expansion. (Liu, Zihao, et al., 2025)

As a metastasis model, PC-9 can be administered intravenously or intracardiacally to generate experimental lung, bone, or multi-organ lesions, implanted directly into the lung to reconstitute the orthotopic microenvironment, or grown subcutaneously to capture spontaneous spread from a primary mass. Stable luciferase expression converts each of these formats into a quantifiable readout: serial in vivo bioluminescence imaging reports metastatic burden over time, so researchers can measure onset of colonization, depth of response to treatment, and regrowth after withdrawal within the same cohort of animals. The model is widely applied to first-, third-, and next-generation EGFR inhibitor studies, combination regimens, and investigations of acquired resistance.

Cell Line Information: PC-9

PC-9 is one of the most frequently used EGFR-mutant NSCLC lines in preclinical oncology. Because it was recovered from a metastatic lymph node rather than a primary tumor, it combines a defined driver mutation with an intrinsic capacity for dissemination. Principal features are listed below.

Our Services

Rather than leaving clients to assemble a model piece by piece, Alfa Cytology runs the PC-9 metastatic study as a turnkey in vivo engagement: study scientists help define the dissemination route, prepare imaging-ready cell suspensions, carry out inoculation under standardized procedures, and hand back a complete evidence package covering lesion burden, histopathology, and survival outcomes.

Workflow of PC-9 Metastatic Tumor Model Construction

Building a PC-9 metastatic model is a staged process that starts with cell authentication and ends with organ-level confirmation of metastatic burden. Every stage is documented, so findings can be reproduced across cohorts and across studies.

  1. Cell authentication and release testing: PC-9 identity is confirmed by STR profiling, mycoplasma clearance is verified, and the EGFR exon 19 deletion is re-confirmed before any expansion for animal work.
  2. Working cell bank preparation: Low-passage cells are scaled up under standardized conditions and cryopreserved, ensuring that every experimental arm originates from a single qualified source.
  3. Luciferase reporter generation (optional): Where imaging readouts are required, cells are transduced and clonally selected for stable, bright expression, then validated in vitro for signal linearity against cell number.
  4. Host and route assignment: Nude, NOD-SCID, or NSG mice are selected to match the aggressiveness of the design, and the dissemination route — lateral tail vein, intracardiac, orthotopic lung, or subcutaneous-spontaneous — is assigned according to the organs under study.
  5. Systemic inoculation: A single-cell suspension with viability of at least 90% is delivered at a defined dose, typically 2 × 106 cells in 150 μL sterile PBS, with the preparation kept chilled throughout to prevent aggregation.
  6. Scheduled imaging and surveillance: Metastatic signal is captured by serial in vivo bioluminescence imaging at fixed intervals, while body weight and clinical condition are logged to trigger humane endpoints when needed.
  7. Randomized dosing and endpoint workup: Once lesions reach a pre-specified signal intensity, animals are randomized into treatment groups; at study end, lungs and target organs undergo ex vivo imaging, metastatic nodule counting, and H&E/immunohistochemical analysis.

Case Study-PC-9 Metastatic Tumor Model Development

Research groups advancing EGFR-targeted agents have relied on the PC-9 metastatic model at Alfa Cytology to characterize how disseminated lesions respond to treatment and how residual disease rebounds after therapy stops. Across these preclinical engagements, the model has supported comparisons of single-agent versus combination schedules, quantified treatment-induced changes in metastatic signal, and informed early development decisions. Outlines of representative study designs and anonymized result summaries are available on request under confidentiality, so prospective clients can evaluate how the platform would serve their own programs.

Why Choose Alfa Cytology?

What distinguishes Alfa Cytology in PC-9 metastasis work is the discipline with which each study is executed: qualified cells, pre-specified routes, scheduled imaging, and endpoints defined before dosing begins. The result is a model readout your program can defend.

  • Metastasis-native biology: PC-9 was derived from a metastatic lymph node, aligning the model closely with the disease state under study.
  • Multiple dissemination formats: experimental, orthotopic, and spontaneous designs are run side by side for direct comparison.
  • Whole-animal quantification: luciferase-based imaging turns metastatic progression into longitudinal curves rather than single observations.
  • Mutation-verified starting material: the EGFR exon 19 deletion is documented for every bank used in a study.
  • Predefined endpoints: imaging thresholds, necropsy schemes, and histology panels are fixed in the protocol before treatment starts.
  • Direct scientific communication: clients interact with the scientists running the study, not an intermediary layer.
