A549 Metastatic Tumor Model Service for Lung Cancer

Fig 1: A549 Metastatic Tumor Model for Lung Cancer preclinical research.

The A549 Metastatic Tumor Model Service for Lung Cancer delivers the single most cited human lung adenocarcinoma xenograft, defined by its signature KRAS G12S, STK11, and KEAP1 co-mutant genotype. Alfa Cytology supports this model end to end — authenticated cell banking in optimized medium, immunodeficient host preparation, subcutaneous and intrapulmonary/orthotopic implantation, scheduled efficacy monitoring, and comprehensive lung-specific endpoint analysis — giving lung cancer programs a dependable, publication-standard in vivo platform.

Overview of A549 Metastatic Tumor Model for Lung Cancer

A549 is a human lung adenocarcinoma cell line established in 1973 by Giard and colleagues from the tumor tissue of a 58-year-old male patient, and it remains the most widely utilized lung cancer cell line in existence. The line is defined by a mutational combination now recognized as one of the most clinically ominous in lung adenocarcinoma: homozygous KRAS G12S driving MAPK/ERK signaling, together with homozygous STK11 (LKB1) nonsense mutation and KEAP1 mutation — a double tumor-suppressor loss that hyperactivates NRF2-dependent antioxidant, glutathione, and ferroptosis-protective programs and confers broad therapeutic resistance. Additional alterations including SMARCA4 truncation and TP53-wild-type status complete a molecular portrait that is intensively studied in vitro for redox metabolism, LKB1/AMPK biology, and drug-resistance mechanisms.

In immunodeficient mice, A549 forms epithelial, alveolar-like adenocarcinoma xenografts with exceptional reproducibility: about 5 × 106 cells in a 1:1 PBS/Matrigel suspension typically yield measurable flank tumors in roughly ten days, a format in which IAP inhibitors, MEK/CDK combinations, and nanocarrier chemotherapies are routinely benchmarked. For metastasis research the line supports a full suite of lung-relevant formats: surgical intrapleural or intraparenchymal implantation (2 × 107 luciferase-labeled cells in 0.2 mL DMEM/F-12/Matrigel) establishes orthotopic primary tumors that spread throughout the thoracic cavity, chest wall, and contralateral lung; intratracheal inoculation of as few as 3.5 × 106 cells produces tumors in 100% of animals with uniform spread to the opposite lung; tail-vein delivery models hematogenous lung colonization for anti-metastatic screening; and single-cell-resolvable engineered lineages have even been used to trace subclonal dynamics of dissemination across all five lobes, mediastinal lymph nodes, and liver. Luciferase-tagged derivatives (A549-Luc, A549-LT) provide non-invasive, whole-body tracking in vivo, cementing A549’s role as the default workhorse of NSCLC preclinical research.

Our Services

Alfa Cytology maintains authenticated, mycoplasma-free A549 stocks — including A549-Luc reporter variants — in F-12K and CO2-compatible workflows, alongside established nude, SCID, and NOD/SCID colonies with thoracic surgical capability for intrapulmonary, intrapleural, and intratracheal procedures. From rapid flank efficacy screens to imaging-guided orthotopic metastasis programs, every study is randomized, quality-controlled, and reported with rigorous statistical interpretation.

Workflow of A549 Metastatic Tumor Model Construction

Constructing an A549 study pairs routine xenograft technique with thoracic surgical expertise for orthotopic formats. The workflow below describes how the model is built and evaluated:

  1. Cell Line Verification: A549 cells are authenticated by STR profiling, confirmed mycoplasma-free, and expanded at controlled passage in F-12K or an approved alternative medium; luciferase derivatives are verified for reporter stability.
  2. Host Preparation and Allocation: Female (or male, per design) athymic nude, SCID, or NOD/SCID mice (typically 4–8 weeks old) are acclimatized and randomly assigned to experimental groups.
  3. Route-Specific Implantation: Suspensions are delivered as planned: 5 × 106 cells in 0.2 mL 1:1 PBS/Matrigel subcutaneously, 2 × 107 luciferase-labeled cells surgically into the lung parenchyma or pleural space, 3.5 × 106 cells intratracheally in Matrigel, or 5 × 106–1 × 107 cells via tail vein.
  4. Tumor Establishment Monitoring: Implant sites or surgical recovery are monitored; subcutaneous tumors typically reach 100–150 mm3 in about ten days, after which caliper measurements, body weight, and clinical status are recorded on schedule.
  5. Randomization and Dosing: Animals are re-randomized at the target tumor size, and dosing with vehicle, benchmark agents (e.g., IAP or MEK pathway inhibitors), or test articles begins on the designated regimen.
  6. In-Life Follow-Up: Tumor growth and tolerability are tracked throughout; A549-Luc cohorts undergo serial bioluminescence imaging in vivo to visualize primary growth and thoracic or systemic spread.
  7. Terminal Endpoint Analysis: At necropsy, primary tumors and lungs are excised and weighed; contralateral lung, chest wall, mediastinal lymph nodes, and liver are examined macroscopically and by hematoxylin and eosin histology, with tissues processed for immunohistochemistry or molecular assays as specified.
  8. Data Analysis and Reporting: Tumor-growth inhibition, imaging signals, metastatic incidence and distribution, and molecular data are statistically evaluated and consolidated into a final study report.

Case Study-A549 Metastatic Tumor Model Development

In a representative preclinical engagement, Alfa Cytology implemented the A549 metastatic lung cancer model for a sponsor advancing a therapeutic candidate in KRAS-mutant non-small cell lung cancer. The scope included expansion and authentication of cells, establishment of tumors in immunodeficient hosts by the route selected for the program, balanced allocation of vehicle and treatment arms, scheduled monitoring of tumor growth and animal condition, and terminal examination of primary tumors and thoracic tissues with histological confirmation — producing an integrated dataset that supported the client’s preclinical development planning. Formats can be configured around subcutaneous efficacy, orthotopic growth, experimental metastasis, or redox/ferroptosis mechanism-of-action endpoints.
