SMMC-7721 Metastatic Tumor Model Service for Liver Cancer

Fig 1: SMMC-7721 Metastatic Tumor Model for Liver Cancer preclinical research.

The SMMC-7721 metastatic tumor model studies hepatic tumor growth and its spread beyond the liver — principally to the lung — through orthotopic implantation in immunodeficient mice. Alfa Cytology delivers this model with full scientific rigor: STR-authenticated and transparently documented SMMC-7721 cell banks, luciferase-labeled derivatives for in vivo imaging, standardized intrahepatic surgical inoculation, and metastasis-scored endpoint packages — executed entirely within preclinical, IACUC-approved workflows.

Overview of SMMC-7721 Metastatic Tumor Model for Liver Cancer

SMMC-7721 was established in 1977 from the liver tumor of a Chinese male patient with hepatocellular carcinoma, and for more than four decades it has been one of the most widely used human liver cancer lines in the Chinese research literature — grown as an epithelial-like, adherent monolayer in RPMI-1640 with 10% FBS, reported to express EGFR and VEGFR, and applied to thousands of studies of tumor growth, angiogenesis, and drug response. Any credible discussion of this line today, however, must include a well-documented authentication finding: independent STR-based studies published in 2015 and 2017 demonstrated that SMMC-7721 is in fact a HeLa derivative, and it now appears on the ICLAC Register of Misidentified Cell Lines (ICLAC-00554). The practical consequence is twofold. Data generated in SMMC-7721 reflect a HeLa-derived biology and should not be over-interpreted as liver-cancer-specific, yet the line remains a legitimate tool where continuity matters — sponsors frequently hold legacy efficacy, angiogenesis, and mechanism datasets on SMMC-7721, and comparability with that history has real value.

Dual in vivo validation in the SMMC-7721 model: subcutaneous xenograft growth of Lv-Lnc-Myd88 versus Lv-NC SMMC-7721 cells (5×10⁶ cells/50 μL PBS, tumor photographs, tumor weight, and longitudinal growth curves) and lung metastasis assessment via tail-vein injection (2×10⁶ cells/0.2 mL PBS, 6 weeks, pulmonary metastatic nodule counts with H&E histology). (Xu, Xiaoliang, et al., 2017)

As a metastasis platform, SMMC-7721 teaches an instructive lesson in model design: when inoculated subcutaneously, the cells form flank tumors reliably but do not spontaneously metastasize to distant organs, which is precisely why orthotopic delivery became the standard approach for dissemination studies. When SMMC-7721 cells or tumor fragments are implanted directly into the liver parenchyma of nude mice, the tumor grows in its native organ context and gives rise to intrahepatic spread and pulmonary metastases — a pattern repeatedly confirmed by H&E histopathology across the literature — while luciferase-tagged derivatives (SMMC-7721-Luc) permit whole-body bioluminescence tracking of tumor burden at the group level and, in cell-suspension formats, multiple hepatic nodules form that better simulate multifocal human disease. Surgical technique matters in this model: small injection volumes and careful sealing of the injection track minimize leakage into the peritoneal cavity, which otherwise seeds abdominal metastases and confounds the pulmonary readout. For sponsors, the resulting study format pairs a robust primary efficacy endpoint (orthotopic liver tumor weight) with a genuine metastatic endpoint (lung nodule counts), within a five-to-six-week timeline.

Cell Line Information: SMMC-7721

SMMC-7721 (synonyms SMMC7721, H7721; Cellosaurus CVCL_0534; RRID:CVCL_0534) is distributed by Chinese repositories and commercial suppliers alike (e.g., Ubigene YC-C056), and omics resources including transcriptome microarrays, SNP arrays, proteome datasets, and COSMIC/GEO entries are associated with the line. Cells display epithelial-like adherent morphology with a recommended 1:2 to 1:4 passaging ratio, express EGFR and VEGFR, and grow robustly for xenograft production. Because of its documented history, Alfa Cytology treats authentication as a first-class deliverable rather than a footnote: STR profiles are verified at study initiation and reported transparently in the final study documentation, and authenticated hepatocellular carcinoma comparators (such as Huh7 or HepG2) can be run in parallel when liver-cancer specificity is essential. Key characteristics are listed below.

Our Services

Few CROs will openly discuss what is known about SMMC-7721; Alfa Cytology prefers to put the facts on the table and build the study around them. You receive STR-authenticated cells, a documented chain of custody, orthotopic surgical technique refined to minimize leakage artifacts, luciferase-enabled burden tracking, and endpoint packages that score lung metastasis by both imaging and histopathology — with the option to run authenticated HCC lines side by side whenever liver-specific conclusions are required, all strictly within preclinical research.

Workflow of SMMC-7721 Metastatic Tumor Model Construction

Constructing an SMMC-7721 metastatic study is an exercise in surgical precision and honest readouts: the model only reveals its metastatic phenotype when cells are placed in the liver, and only when peritoneal contamination is controlled. The sequence below reflects the standard operating flow, configurable to either cell-suspension or tumor-fragment orthotopic formats plus a subcutaneous cohort where primary efficacy is the priority.

  1. Cell line authentication and banking: SMMC-7721 is expanded in RPMI-1640 + 10% FBS from low-passage stocks; identity is confirmed by STR profiling, results are documented, and cultures are verified mycoplasma-free before entering the animal phase.
  2. Reporter labeling where imaging is planned: For imaging cohorts, luciferase (or GFP) expression is established and validated in vitro — including the linearity of photon output versus cell number — so that bioluminescence measured later is a quantitative proxy for viable tumor burden.
  3. Inoculum preparation: For cell-suspension orthotopic models, a concentrated single-cell suspension is prepared in a minimal volume (on the order of 10–20 µL per site); for fragment models, donor subcutaneous tumors are excised under aseptic conditions and trimmed into standardized pieces.
  4. Nude mouse preparation and anesthesia: BALB/c nude mice are acclimatized, health-screened, and anesthetized; the abdominal wall is prepared aseptically, and a small left-lateral incision exposes the left hepatic lobe for inoculation.
  5. Intrahepatic implantation and wound closure: The suspension is injected slowly into the liver parenchyma, the needle track is compressed (and sealed, e.g., with a sterile swab or bioadhesive) to prevent leakage, the abdominal wall and skin are closed in layers, and the animal recovers under warming and monitoring per IACUC protocol.
  6. Tumor growth monitoring and randomization: Animals are imaged on a fixed schedule (for Luc formats, e.g., days 7, 14, and 35) and weighed regularly; cohorts are randomized at matched burden into vehicle and treatment arms, with sorafenib or other reference agents available as positive controls for liver cancer studies.
  7. Treatment administration: Test articles — kinase inhibitors, anti-angiogenic agents, natural-product derivatives, or combinations — are dosed per schedule, with body weight, clinical condition, and interim imaging capturing tolerability and early response.
  8. Necropsy, metastasis scoring, and reporting: At endpoint (~weeks 5–6), the liver is excised and weighed, the abdominal cavity is inspected for leakage artifacts, lungs are fixed for surface nodule counting, and liver/lung tissues undergo H&E and IHC confirmation; bioluminescence kinetics, liver weights, and lung metastasis data are analyzed and compiled into the final report with full authentication documentation.
