LS174T Orthotopic Mouse Model Service for Colon Cancer

Fig 1: LS174T Orthotopic Mouse Model for Colon Cancer preclinical research.

The LS174T orthotopic mouse model replicates a mucin-producing, KRAS^G12D-driven human colorectal adenocarcinoma within the murine cecal wall, preserving the anatomical site, stromal architecture, and secretory phenotype of intestinal goblet cell-derived malignancy for rigorous preclinical therapeutic evaluation. Alfa Cytology specializes in constructing this mucinous orthotopic xenograft system with authenticated cell line stewardship, precision surgical fragment implantation onto the cecal serosa, and terminal multimodal endpoint analysis, enabling robust assessment of Wnt/β-catenin pathway inhibitors, mucin-targeting strategies, and standard-of-care chemotherapeutic regimens in a physiologically faithful tumor microenvironment.

Overview of LS174T Orthotopic Mouse Model for Colon Cancer

The LS174T cell line was originally established by Tom and colleagues in 1976 from a primary colorectal adenocarcinoma classified as Dukes type B, arising in a 58-year-old female patient. It represents a trypsinized variant of the parental LS180 line, selected for enhanced passage stability while retaining the mucin-secreting, intestinal epithelial phenotype of its progenitor. Genetically, LS174T is characterized by an activating KRAS^G12D mutation that drives constitutive MAPK signaling, a BRAF^D211G alteration, an activating PIK3CA^H1047R mutation, biallelic APC inactivation that unleashes canonical Wnt/β-catenin pathway hyperactivity, and wild-type TP53—a molecular profile that aligns with microsatellite instability (MSI) status. The cells exhibit a partially differentiated epithelial phenotype with abundant microvilli, intracytoplasmic mucin vacuoles, and robust constitutive expression of MUC2 secretory mucin, carcinoembryonic antigen (CEA), and cytokeratins, faithfully recapitulating the biology of mucinous colorectal carcinoma.

When established as an orthotopic xenograft within the cecal wall of immunodeficient mice, LS174T forms moderately growing, mucin-rich adenocarcinomas that maintain their secretory properties in vivo. The surgical orthotopic implantation technique—wherein subcutaneous donor tumor fragments are sutured onto the serosa of the cecal lesser curve—establishes a tumor-stromal interface that preserves the serosal, muscular, and mucosal architecture of the colonic wall. This anatomical fidelity fosters physiologic angiogenesis, lymphatic drainage toward the mesenteric lymph nodes, and peritoneal seeding patterns that subcutaneous models cannot replicate. The pronounced CEA and MUC2 expression further render LS174T particularly valuable for evaluating antibody-drug conjugates targeting CEA, agents that disrupt the mucin-associated extracellular matrix barrier, and Wnt/β-catenin pathway modulators—offering a preclinical platform where mucinous tumor biology, KRAS-driven proliferation, and Wnt-dependent stemness converge within a clinically relevant anatomical context.

Workflow of LS174T Orthotopic Mouse Model Construction

Construction of the LS174T orthotopic colon cancer model integrates cell culture quality assurance, sterile surgical technique, and standardized endpoint harvesting. Our protocol is optimized for consistent engraftment of mucin-producing tumors, minimal perioperative mortality, and robust therapeutic readouts suitable for Wnt pathway and mucin-targeting therapeutic assessment. The workflow proceeds as follows:

  1. Cell Line Authentication and Master Bank Expansion: LS174T master stocks are revived and expanded in antibiotic-free EMEM or DMEM medium. Each production batch undergoes STR profiling against the reference database, mycoplasma PCR screening, and in vitro MUC2 and CEA expression validation by immunocytochemistry or ELISA to confirm the mucin-secreting phenotype prior to surgical use.
  2. Subcutaneous Donor Tumor Generation: LS174T cells (1 x 106 in 100 μL Matrigel) are injected subcutaneously into the bilateral flanks of donor athymic nude mice. Tumors are allowed to grow until reaching ~50–100 mm3 (typically 2–3 weeks), at which point they are excised aseptically. Necrotic tissue is trimmed away, and tumors are sectioned into 1 mm3 fragments using a sterile scalpel. Fragment size and quality are verified under a dissecting microscope to ensure uniformity.
  3. Recipient Mouse Preparation and Anesthesia: Female athymic nude mice (BALB/c nu/nu or NOD/SCID, 6–8 weeks, 18–22 g) are acclimated for 7 days under SPF barrier conditions. On the procedure day, general anesthesia is induced with 1–2% isoflurane in medical oxygen via nose cone. The ventral abdomen is shaved, cleansed sequentially with povidone-iodine and 70% ethanol, and draped sterilely.
  4. Cecal Serosa Orthotopic Implantation: A 1 cm lower midline or left paramedian abdominal incision exposes the cecum. Gentle pressure is applied to deliver the cecum through the incision. A 1 mm3 LS174T tumor fragment is positioned onto the serosa of the lesser curve of the cecum—avoiding contact with the peritoneal wall—and secured with 8-0 nylon sutures (interrupted or purse-string technique) or a biocompatible tissue adhesive (~10 μL, drying 10–15 s). The cecum is carefully returned to its anatomic position, ensuring the implant does not abut the peritoneum. The abdominal wall and skin are closed with 5-0 or 6-0 nylon sutures or surgical staples.
  5. Post-Operative Recovery and Clinical Surveillance: Mice recover on a heated pad until fully ambulatory. Post-operative analgesia (buprenorphine 0.05–0.1 mg/kg, s.c.) is administered per IACUC guidelines. Daily monitoring for 7–10 days assesses wound integrity, body weight, food intake, and abdominal distension. Humane endpoints include >20% body weight loss, wound dehiscence, or signs of severe distress. Wound clips, if used, are removed 10–14 days post-surgery once the incision has healed.
  6. Tumor Establishment Monitoring and Cohort Randomization: Tumor engraftment is confirmed by palpable abdominal mass beginning 7–14 days post-implantation. Tumor dimensions are measured twice weekly by digital caliper (length x width2 x 0.5 = mm3). Mice with confirmed tumors are enrolled and randomized into treatment cohorts once mean tumor burden reaches a predefined threshold (e.g., palpable mass ~75–125 mm3, typically Day 14–21).
  7. Therapeutic Intervention and In-Life Assessment: Test articles are administered per protocol via oral gavage, intravenous, intraperitoneal, or subcutaneous routes. Tumor dimensions and body weights are recorded twice weekly; clinical observations are documented three times weekly. Blood samples may be collected via submandibular bleed for pharmacokinetic analysis, CEA biomarker quantification, or circulating tumor DNA assessment at designated intervals.
  8. Necropsy, Tumor Harvest, and Multimodal Endpoint Analysis: At study termination, mice are humanely euthanized. The primary cecal tumor is excised, weighed, photographed, and dimensions recorded. The abdominal cavity is inspected macroscopically for peritoneal carcinomatosis and ascites. Metastatic target organs (liver, lung, spleen, mesenteric lymph nodes, diaphragm) are harvested. Tissues are fixed in 10% neutral buffered formalin for H&E histology, or snap-frozen in liquid nitrogen for molecular analysis. Immunohistochemistry for Ki-67, cleaved caspase-3, CD31, MUC2, CEA, β-catenin (nuclear vs. cytoplasmic), phospho-ERK1/2, and LGR5 provides mechanistic insight into proliferation, apoptosis, angiogenesis, mucin secretion, CEA expression, Wnt pathway activation, MAPK signaling, and stemness marker dynamics.

Case Study-LS174T Orthotopic Mouse Model Development

In a recent preclinical program, the LS174T orthotopic model was employed to evaluate the efficacy of a novel porcupine (PORCN) inhibitor—a Wnt secretion antagonist—in combination with 5-fluorouracil against mucinous colorectal cancer. LS174T tumor fragments were sutured onto the cecal serosa of athymic nude mice, with tumor establishment confirmed by palpation within 10–14 days. Upon reaching a mean tumor volume of approximately 100 mm^3, mice were randomized to receive vehicle, PORCN inhibitor monotherapy, 5-FU monotherapy, or the combination for 3 weeks. The combination arm demonstrated marked suppression of primary cecal tumor growth compared to both monotherapy and vehicle controls, as evidenced by reduced tumor weight at necropsy and diminished tumor dimensions. Immunohistochemical analysis revealed decreased nuclear β-catenin staining intensity and reduced MUC2 immunoreactivity in the combination cohort, indicating effective Wnt pathway suppression and mucin production attenuation. Additionally, Ki-67 staining was diminished while cleaved caspase-3 positivity was elevated, suggesting that dual Wnt blockade and chemotherapy not only inhibit proliferation but also enhance apoptotic cell death in mucinous tumor cells. These findings underscore the utility of the LS174T orthotopic platform for capturing both Wnt-dependent stemness modulation and chemosensitization effects in a mucin-producing colorectal cancer context.
