Pan02 (Panc02) / Pan02-luc Orthotopic Mouse Model Service for Pancreas Cancer

Fig 1: Pan02 (Panc02) / Pan02-luc Orthotopic Mouse Model for Pancreas Cancer preclinical research.

The Pan02 (Panc02) / Pan02-luc orthotopic mouse model for pancreas cancer places a chemically induced, syngeneic pancreatic ductal adenocarcinoma back into its organ of origin within an immunocompetent C57BL/6 host — an unmatched setting for studying tumor–immune interplay under treatment. Alfa Cytology offers this model as a complete preclinical solution: syngeneic cell supply with certified identity, precision surgical implantation, scheduled bioluminescence or ultrasound monitoring, dosing arms, and meticulous terminal workups — all executed to give your pancreas cancer program defensible, decision-grade in vivo data.

Overview of Pan02 (Panc02) / Pan02-luc Orthotopic Mouse Model for Pancreas Cancer

Pan02 — also written Panc02 — is the most widely used murine pancreatic ductal adenocarcinoma cell line in preclinical oncology. It was established by Corbett and colleagues in 1984, following implantation of cotton threads impregnated with the carcinogen 3-methylcholanthrene into the pancreas of C57BL/6 mice and subsequent serial in vivo transplantation of the resulting tumors. Genomic analyses have uncovered activating lesions in Kras and Smad4 in this line, although its overall mutational spectrum differs from that of human PDAC — a caveat balanced by an overwhelming practical strength: because Pan02 arose in C57BL/6 mice, it grows as a fully syngeneic tumor, leaving the host immune system intact. The cells display an aggressive, largely undifferentiated growth pattern, with rapid proliferation and invasion exceeding that of genetically engineered alternatives such as 6606PDA.

Orthotopic implantation unlocks the full value of this biology. When Pan02 cells are surgically introduced into the pancreatic parenchyma of immunocompetent C57BL/6 mice, they give rise to locally invasive tumors accompanied by the dense stromal reaction characteristic of pancreatic cancer, with progression to peritoneal seeding and distant spread in disseminated formats. The relatively measured growth of orthotopic Pan02 tumors provides a generous treatment window — a key reason the model has become a workhorse for immunotherapy evaluation, including checkpoint blockade, cancer vaccines, and oncolytic strategies. The luciferase-tagged derivative, Pan02-luc, adds quantitative bioluminescence readouts to the same framework, enabling tumor burden to be tracked serially in individual animals rather than inferred from endpoint measurements alone.

Cell Line Information: Pan02 (Panc02) / Pan02-luc

Our Services

Alfa Cytology runs the Pan02 (Panc02) / Pan02-luc orthotopic program end to end — from authenticated cell banking and surgical implantation through longitudinal imaging, dosing regimens, immune-focused endpoint analysis, and consolidated reporting — freeing your team to concentrate on candidate selection while the in vivo engine runs with proven consistency.

Workflow of Pan02 (Panc02) / Pan02-luc Orthotopic Mouse Model Construction

Constructing an orthotopic Pan02 study combines sterile microsurgery with disciplined imaging and immunologically meaningful endpoints. The sequence below reflects the platform as standardized for routine execution, with strain, inoculum, and schedule adaptable to each program.

  1. Cell Line Expansion and Release Testing: Pan02 or Pan02-luc cultures are expanded from authenticated stocks and cleared for study use only after mouse STR profiling confirms identity, mycoplasma testing proves negative, and luciferase output of the reporter line is verified in vitro.
  2. Host Animal Preparation: Immunocompetent C57BL/6 mice, typically 6–8 weeks of age, are ordered, quarantine-checked, and acclimatized for at least one week under barrier conditions, preserving the syngeneic, immune-intact character that defines this model.
  3. Surgical Orthotopic Implantation: Under aseptic conditions with a dissecting microscope, the pancreas is exteriorized through a small left-flank abdominal incision, and 1 × 106 Pan02 / Pan02-luc cells in a 25–50 µL Matrigel/PBS suspension are injected into the pancreatic tail; alternatively, a small freshly harvested Pan02 tumor fragment is sutured in place, after which the abdominal wall and skin are closed in layers.
  4. Post-Surgical Recovery and Baseline Imaging: Mice recover under warming with post-operative analgesia per approved protocols, and baseline bioluminescence imaging after D-luciferin injection — or baseline high-resolution ultrasound — confirms successful engraftment and documents starting tumor status.
  5. Cohort Randomization: Animals are allocated to control and treatment groups on the basis of baseline signal, balancing tumor burden across arms before the first dose is administered.
  6. Scheduled Monitoring During Treatment: Tumor progression is followed on a fixed schedule using bioluminescence imaging or calibrated ultrasound measurement, complemented by body weight and clinical health scoring to capture both efficacy and tolerability throughout the dosing period.
  7. Test Article Administration: Therapeutics are delivered according to the sponsor-approved regimen — intraperitoneal, intravenous, or oral routes supported — with the syngeneic host permitting meaningful evaluation of immunotherapies alongside conventional agents.
  8. Terminal Necropsy and Immune-Enriched Analysis: At endpoint, mice are euthanized and primary pancreatic tumors are excised and weighed; abdominal organs are inspected and metastatic deposits scored; tissue sections undergo H&E and immunohistochemical staining, with immune-cell markers added where immunomodulatory mechanisms are under investigation.
