LM8 / LM8-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: LM8 / LM8-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The LM8 / LM8-luc orthotopic mouse model of sarcoma places a highly metastatic murine osteosarcoma line directly into the bone of syngeneic C3H hosts, where it forms primary tumors within the marrow cavity and disseminates to the lung with near-complete fidelity. From authenticated cell banking and intratibial implantation surgery to serial luciferase imaging of both the primary bone lesion and pulmonary metastases, Alfa Cytology delivers complete LM8 / LM8-luc orthotopic study programs tailored to each sponsor’s mechanism, dosing schedule and endpoint needs—exclusively within preclinical research.

Overview of LM8 / LM8-luc Orthotopic Mouse Model for Sarcoma

LM8 traces its pedigree to the Dunn osteosarcoma, itself established in 1963 from a spontaneous tumor of the tail in a C3H mouse. Seeking a more aggressive derivative, investigators applied Fidler’s classic in vivo selection scheme—injecting Dunn cells into the tail vein, harvesting the resulting lung metastases, re-expanding them in vitro and repeating the cycle eight times—until a variant emerged that produced lung metastases in 100% of recipients (Asai and colleagues, International Journal of Cancer, 1998). Side-by-side characterization showed that LM8 outgrows its parent and carries a molecular arsenal suited to dissemination: markedly higher VEGF expression, elevated matrix metalloproteinase-2 and -9 activity, greater invasiveness through mesothelial monolayers, enhanced alkaline phosphatase activity reflecting its osteogenic identity, and increased β-catenin. Its surface is ruffled with filopodia and lamellipodia, the physical signature of a motile cell.

In the orthotopic format, LM8 or luciferase-tagged LM8-luc cells are introduced into the medullary cavity of the proximal tibia, where the tumor grows within genuine bone microenvironment, incites osteolysis and neovascularization, and seeds the lung—the dominant cause of mortality in the human disease. Because donor and host are both C3H-derived, the model runs in immunocompetent animals, an asset for immuno-oncology and tumor-associated macrophage studies. Bioluminescence imaging of LM8-luc tumors is remarkably informative: the primary lesion is macroscopically evident within five days of implantation, the first pulmonary signals appear around day twelve, and by day nineteen every animal shows lung involvement, with metastases confirmed at necropsy by ex vivo imaging and H&E staining. One methodological nuance deserves attention—whether early lung signals arise purely from spontaneous dissemination or partly from procedure-related cell seeding is actively debated, a question longitudinal imaging and primary-tumor ablation designs can help resolve. Even with this caveat, the model has repeatedly proven its worth in preclinical screens of anti-metastatic agents, from gemcitabine and eribulin to CSF-1R inhibitors that reprogram macrophages and stimulate T-cell infiltration.

Cell Line Information: LM8 / LM8-luc

LM8 and its luciferase-labeled derivative LM8-luc share the same C3H genetic background and aggressive metastatic behavior; the reporter line additionally expresses firefly luciferase introduced by lentiviral transduction. Key characteristics are summarized below.

Reporter variant (LM8-luc) Lentiviral firefly luciferase; primary tumors macroscopically evident by day 5, first lung signals around day 12, all mice lung-positive by day 19 in the intratibial model
Typical orthotopic inoculum 1 × 106 cells per site in 20–100 µL PBS, injected into the medullary cavity of the proximal tibia, with or without Matrigel
Metastasis formats Spontaneous lung metastasis from the orthotopic primary tumor; experimental lung colonization via tail-vein injection; subcutaneous flank tumors also metastatic
Representative applications Anti-metastatic agent screening (gemcitabine, eribulin), CSF-1R / tumor-associated macrophage reprogramming studies, immuno-oncology combinations, radiation research, osteosarcoma metastasis biology

Workflow of LM8 / LM8-luc Orthotopic Mouse Model Construction

Orthotopic construction of the LM8 / LM8-luc model is a survival-surgery workflow carried out under inhalation anesthesia, with aseptic technique, scheduled analgesia and daily post-operative welfare checks consistent with institutional animal-care standards. The procedure typically proceeds in the following stages:

  1. Cell line qualification: LM8 or LM8-luc vials are mycoplasma-tested and morphology-checked against the parental profile; luciferase-positive cells are confirmed in vitro by luminometer before expansion.
  2. Animal preparation: C3H/HeJ or C3H/HeN mice (typically 6–8 weeks old) are acclimatized, health-screened and randomized by body weight ahead of surgery.
  3. Anesthesia and asepsis: mice are anesthetized with isoflurane, the surgical leg is clipped and disinfected, and a sterile field with sterile instruments is maintained throughout.
  4. Surgical approach: a small incision over the knee exposes the proximal tibia, providing access to the medullary cavity under magnification (a refined percutaneous approach is equally established).
  5. Intratibial inoculation: LM8 / LM8-luc cells (commonly 1 × 106 in 20–100 µL PBS, with or without Matrigel) are delivered into the proximal tibial marrow cavity with a fine needle, with care to avoid cortical breach and extraosseous spill.
  6. Closure and analgesia: the soft tissue and skin are closed with absorbable sutures and wound clips, and buprenorphine or meloxicam is dosed on schedule; gait and body condition are monitored daily until full recovery.
  7. Longitudinal monitoring: primary tumor growth and lung dissemination are tracked in vivo by weekly bioluminescence imaging after intraperitoneal D-luciferin for LM8-luc, complemented by body-weight measurement and clinical scoring; plain X-ray or micro-CT can be added to document cortical destruction and reactive bone formation.
  8. Endpoint and analysis: at termination the tibial tumor is excised and weighed, tumor dimensions are recorded, lungs are removed for ex vivo bioluminescence and metastasis counting, and tissues are processed for histopathology, immunohistochemistry, flow-cytometric immune profiling and downstream molecular assays.
