ENU-Induced Glioma Model Service

Fig 1: ENU-Induced Glioma Model preclinical research.

The ENU-induced glioma model achieves something no other chemically induced brain tumor system can claim, a near-100% tumor incidence from a single transplacental exposure, combined with a finely resolved progression profile from hyperplasia to invasive glioma. Alfa Cytology delivers this neuro-oncology workhorse as a full-service program, managing timed-pregnancy breeding, carcinogen administration, longitudinal MRI surveillance, and stage-graded neuropathology so that sponsors can study glioma biology and intervention across every developmental stage of the disease.

Overview of ENU-Induced Glioma Model

The story of the ENU glioma model begins in the late 1960s with the demonstration that a single intravenous injection of N-ethyl-N-nitrosourea into a pregnant rat could sentence every one of her offspring to a brain tumor. ENU is a direct-acting, short-lived alkylating agent whose hallmark lesion, O6-ethylguanine, is removed from DNA far more slowly in nervous tissue than elsewhere in the body; this persistence, first correlated with neuro-specific carcinogenesis in classic experiments, explains why a compound delivered to the dam expresses its mutagenicity almost exclusively in the fetal central nervous system. Affected pathways include p53, Ha-ras, neu/erbB-2, and Braf, and the target cell is the proliferating neuroepithelium of the subventricular zone, whose daughters form glial-lineage tumors that begin as microscopic clusters and mature over months.

What makes the model uniquely valuable is the precision with which its natural history has been documented. Tumorigenesis passes through discrete, measurable stages: hyperplastic foci in young animals, expanding cellular clusters of under 500 µm by roughly ten to twelve weeks of age, microtumors below 1 mm, medium tumors of 1–2 mm, and finally gross tumors exceeding 2 mm, all arising with multiple lesions per brain and essentially complete penetrance. Histologically the tumors traverse the human spectrum from oligodendroglioma and mixed oligoastrocytoma through anaplastic glioma toward glioblastoma-like histology in aged animals, and extraneural lesions such as malignant schwannomas bearing neu/erbB-2 mutations appear in parallel. Adult animals, tellingly, do not develop tumors from ENU, and mice require additional genetic manipulation, underscoring that the model's power lies in capturing the vulnerability of the developing brain. Because the tumor burden can be followed non-invasively by MRI from about four months of age, and because interventions from calorie restriction to anti-angiogenic agents have been benchmarked against its staged growth, the ENU model remains the reference platform for preclinical neuro-oncology carried out in vivo.

Principles of ENU-Induced Glioma Model Construction

The ENU model's reliability rests on four foundations: the developmental timing of exposure, the DNA-repair asymmetry that concentrates damage in the brain, the staged natural history of lesions, and disciplined longitudinal monitoring. The governing elements are tabulated below.

Element Agent / Factor Function in Model Construction Representative Conditions
Carcinogen N-ethyl-N-nitrosourea (ENU), single intravenous injection to pregnant dams A direct-acting alkylating agent with a ~20-minute half-life; alkylates O6-guanine, O2-thymine, and O4-thymine, with the O6-ethylguanine adduct persisting in brain DNA due to weak nervous-system repair, driving mutation of p53, Ha-ras, neu/erbB-2, and Braf. 50 mg/kg i.v. to the dam on gestation day 15 (offspring tumor incidence approaching 100%); 20–50 mg/kg i.v. in related protocols
Developmental window Gestational or neonatal timing Sensitivity is confined to development: exposure in the last week of gestation or the first postnatal week induces tumors in the majority or entirety of offspring, whereas adult exposure is ineffective; tumor multiplicity peaks with day-20 exposure, while per-cell sensitivity is greatest earliest. Gestation day 15–21 transplacental protocols; neonatal day 1–7 intraperitoneal alternatives (~10 mg/kg single dose)
Species and strain Rats (highly susceptible) vs mice (refractory alone) Rats, especially BD IX, Sprague-Dawley, Wistar, and F344 backgrounds, develop gliomas and schwannomas readily; mice do not respond to ENU alone and require cooperating genetic lesions (for example Trp53 or Nf1 defects), a critical design consideration. BD IX historical standard; F344 and Sprague-Dawley for contemporary MRI-tracked studies
Staged tumor progression Hyperplasia → microtumor → medium → gross glioma Lesions advance through a documented continuum from hyperplastic foci, to sub-500 µm clusters at ~10–12 weeks of age, microtumors (<1 mm), medium tumors (1–2 mm), and gross tumors (>2 mm), with multiple tumors per brain; histology spans oligodendroglioma through anaplastic glioma. Hyperplasia at 4–6 weeks of age; MRI-visible tumors from ~4 months; malignant evolution toward glioblastoma-like histology by ~1 year
Parallel lesions & monitoring Schwannomas/MPNSTs and MRI surveillance Extraneural malignant schwannomas with characteristic neu/erbB-2 mutations arise along cranial nerves; longitudinal T2-weighted MRI detects intracranial tumors from ~4 months of age with well-characterized doubling times (~0.5 months), enabling in-life therapeutic evaluation. MRI at 4, 6, and 8 months or per protocol; histological confirmation and immunohistochemistry at endpoint

Our Services

Because the ENU model begins before its subjects are born, it punishes improvisation and rewards planning, which is why Alfa Cytology pairs every engagement with timed-mating logistics, written dosing calendars for the dams, and pre-registered MRI and sacrifice schedules that keep months-long studies perfectly synchronized.

Workflow of ENU-Induced Glioma Model Construction

An ENU glioma study is a two-generation project: carcinogen delivery to timed-pregnant dams, followed by a year or more of observation in the offspring, punctuated by imaging and staged sacrifices. The operational sequence is as follows.

  1. Study design and strain selection: The sponsor's objectives determine the rat strain, typically F344 or Sprague-Dawley for MRI compatibility, the gestational day of exposure, group sizes, and the therapeutic or chemoprevention arms to be tested during tumor progression.
  2. Timed mating program: Breeder pairs are assembled and monitored so that gestational age is known precisely, since the carcinogen must be delivered within a narrow developmental window; pregnancy is confirmed and dams assigned to dosing cohorts.
  3. Baseline assessment of dams: Pregnant dams are weighed and clinically assessed, and baseline condition is documented to normalize any pregnancy-related variables before carcinogen exposure.
  4. ENU administration: A freshly prepared ENU solution in distilled water or saline is administered to the dam as a single intravenous injection at the protocol dose on the designated gestation day, with prepared solution used promptly given the compound's ~20-minute half-life.
  5. Parturition and offspring rearing: Dams deliver normally, litters are standardized for size, and offspring are reared under controlled conditions with regular weighing; general health, neurological signs, and behavioral changes are monitored throughout development.
  6. Randomization and intervention scheduling: Offspring are weaned, genotyped or ear-marked as required, and randomized to control and treatment groups; test articles, dietary regimens such as calorie restriction, or candidate therapeutics are administered on the sponsor-defined schedule aligned to tumor stage.
  7. Longitudinal MRI surveillance: Beginning at approximately four months of age, cohorts undergo T2-weighted MRI at protocol intervals to detect intracranial tumors, measure their volumes, and calculate individual growth rates for in-life efficacy assessment.
  8. Endpoint euthanasia and neuropathological analysis: At the final time point or upon predefined neurological endpoints, animals are euthanized, brains are harvested and fixed, and lesions are staged histologically from hyperplasia through glioma subtypes with confirmation by GFAP, Ki-67, and lineage-marker immunohistochemistry, alongside neu/erbB-2 analysis of any schwannomas.
