JIMT-1 Metastatic Tumor Model Service for Breast Cancer

Fig 1: JIMT-1 Metastatic Tumor Model for Breast Cancer preclinical research.

The JIMT-1 metastatic tumor model recreates therapy-refractory, HER2-positive breast cancer in immunodeficient hosts, with study formats that follow both primary tumor growth and spread to distant organs such as the lung. Alfa Cytology supports JIMT-1 programs from authenticated cell supply through mammary fat-pad or tail-vein implantation, longitudinal imaging, and metastasis-burden analysis, giving sponsors working on drug-resistant breast cancer a dependable in vivo platform.

Overview of JIMT-1 Metastatic Tumor Model for Breast Cancer

JIMT-1 was established in 2003 from the pleural effusion of a 62-year-old woman with grade 3 invasive ductal carcinoma (T2N1M0) who had become clinically resistant to trastuzumab (Tanner et al., 2004; DSMZ ACC 589). The line carries an amplified, overexpressed HER2 oncogene without coding-sequence mutations, yet it does not respond to trastuzumab, pertuzumab, or lapatinib, because several resistance mechanisms operate simultaneously: an activating PIK3CA mutation, low PTEN expression, high NRG1 expression, and comparatively modest HER2 protein levels despite gene amplification. Hormone receptors are absent, cytokeratins 5/14 and 8/18 confirm an epithelial progenitor phenotype, and the cells form xenograft tumors in nude mice with downstream Akt signaling that resists blockade. These properties have made JIMT-1 the reference model for studying intrinsic trastuzumab resistance.

Classification of breast cancer brain metastasis models by immunodeficiency level (xenogeneic vs. syngeneic vs. PDX), with JIMT-1-BR3 categorized in the intracardiac xenogeneic route for HER2+ brain metastasis modeling. In immunodeficient hosts—nude, SCID, NOD-SCID, or NSG/NOG strains depending on the design—JIMT-1 grows reliably as flank or mammary fat-pad xenografts and shows a natural tendency to seed the lung. Serial passage of lung deposits through SCID mice produced L-JIMT-1, a derivative that metastasizes to the lung in every recipient, more rapidly and more extensively than the parental line, and that tolerates nearly all anti-HER2 agents tested; it has since been used to benchmark next-generation antibody–drug conjugates, including T-DM1, trastuzumab deruxtecan, and disitamab vedotin. For experimental work, tumor cells are commonly implanted into the mammary fat pad to model the primary site, injected into the tail vein to generate defined lung-metastasis burdens, or labeled with luciferase so that metastatic spread can be quantified non-invasively as the study proceeds.

Cell Line Information: JIMT-1

Repository records describe JIMT-1 as slow to settle during the first one to two weeks after revival, after which it is split 1:2 to 1:8 every two to four days using trypsin/EDTA; seeding density is on the order of 1 × 106 cells per 80 cm2 and harvests reach roughly 8–10 × 106 cells per flask. The line is listed in the Cancer Cell Line Encyclopedia and has public RNA-sequencing data, and reference STR profiling against the global standard has confirmed its authenticity. Its complex, hyperdiploid karyotype carries numerous marker chromosomes and more than two dozen copy-number aberrant regions, including the HER2 amplicon at 17q12 and a second amplicon at 17q21.31—a genomic signature that helps distinguish genuine JIMT-1 derivatives from misidentified cultures.

Our Services

Built around this refractory biology, Alfa Cytology configures JIMT-1 metastasis studies to answer defined development questions—whether benchmarking a next-generation antibody–drug conjugate against standard agents, testing combinations intended to reverse PI3K-pathway resistance, or quantifying metastatic burden by imaging. Programs move from in vitro qualification through in vivo efficacy and survival arms, with histology and biomarker analysis consolidated into a single deliverable.

Workflow of JIMT-1 Metastatic Tumor Model Construction

A JIMT-1 metastasis study advances through a fixed sequence of technical stages, each of which can be tuned to the sponsor’s route, imaging, and endpoint requirements. The outline below reflects the most frequently requested configuration.

  1. Cell revival and authentication: Authenticated JIMT-1 stocks are revived, STR-profiled, and cleared by mycoplasma testing before in vitro expansion; the line’s slow early growth is anticipated and passage ratios adjusted accordingly.
  2. Study design and route selection: The metastatic format is chosen—mammary fat-pad orthotopic, subcutaneous flank, or tail-vein lung colonization—and group sizes, randomization rules, and humane endpoints are documented in the study plan.
  3. Inoculum preparation: Single-cell suspensions are prepared, typically 1 × 106 to 2 × 106 cells for site-directed implants and roughly 5 × 105 cells for tail-vein delivery, suspended in a supportive matrix where required; luciferase labeling is applied when imaging readouts are planned.
  4. Tumor establishment and baseline imaging: Palpable tumors or baseline photon flux are recorded on a fixed schedule until cohorts enter the enrollment window, commonly around 100–200 mm3 for solid implants.
  5. Randomization and dosing: Animals are balanced into vehicle, benchmark, and test-article arms, after which antibody–drug conjugates, tyrosine kinase inhibitors, antibody constructs, or combination regimens are administered on the sponsor-defined schedule.
  6. In-life monitoring: Tumor dimensions or bioluminescence signal, body weight, and clinical observations are captured at defined intervals, with imaging flags raised as soon as metastatic signal emerges.
  7. Endpoint and metastasis analysis: At termination, primary tumors are excised and weighed; lungs are inflated and scored for surface nodules with histological confirmation, IHC and molecular endpoints are completed, and the statistical report is issued.

Case Study-JIMT-1 Metastatic Tumor Model Development

Alfa Cytology has supported JIMT-1-based preclinical work that includes head-to-head benchmarking of HER2-directed antibody–drug conjugates against resistant disease, combination strategies designed to reverse PI3K-pathway-driven resistance and validated in vivo, and imaging-guided quantification of lung metastatic burden. These programs have drawn on both the parental line and its lung-tropic derivatives, with readouts spanning tumor growth, survival, metastatic lesion counts, and pathway biomarkers; representative datasets can be shared once confidentiality terms are established.

Why Choose Alfa Cytology?

A metastasis model earns its keep through disciplined inoculation, consistent imaging, and rigorous endpoint analysis rather than through the cell line alone. Alfa Cytology pairs that operational discipline with a deep working knowledge of this unusually demanding model.

  • Resistance-relevant biology: JIMT-1 mirrors the refractory HER2-positive setting that sponsors most often need to model.
  • Format range: orthotopic, flank, and experimental lung-metastasis configurations are available from a single authenticated cell source.
  • Imaging integration: bioluminescence-based burden tracking adds time-resolved data while reducing cohort sizes.
  • Endpoint rigor: standardized lung-nodule scoring, histological confirmation, and IHC biomarker readouts.
  • Authenticated inputs: STR-verified, mycoplasma-free cells with documented passage history.
  • Communication cadence: milestone updates and full raw-data access keep sponsors close to the study.
