HeyA8 / HeyA8-luc Orthotopic Mouse Model Service for Ovarian Cancer

Fig 1: HeyA8 / HeyA8-luc Orthotopic Mouse Model for Ovarian Cancer preclinical research.

The HeyA8 / HeyA8-luc orthotopic mouse model for ovarian cancer faithfully recapitulates the anatomy, ascites formation, and peritoneal dissemination pattern of human disease, enabling non-invasive, real-time monitoring of tumor progression in a physiologically relevant setting. Alfa Cytology provides this model as a fully managed, preclinical-only service — spanning cell line qualification, orthotopic implantation, serial bioluminescence imaging, dosing studies, and terminal histopathology — delivering decision-ready data packages to accelerate your ovarian cancer drug development programs.

Overview of HeyA8 / HeyA8-luc Orthotopic Mouse Model for Ovarian Cancer

HeyA8 is a human ovarian carcinoma cell line generated through serial in vivo selection: HEY cells — themselves originally established from a patient-derived xenograft of papillary cystadenocarcinoma — were passaged in the peritoneal cavity of mice, and the resulting ascites-recovered population, designated HeyA8, emerged as a markedly more aggressive variant. Molecularly, the line carries activating KRAS p.Gly12Asp and BRAF p.Gly464Glu alterations and expresses high levels of interleukin-8 (IL-8), which functions as an autocrine growth factor driving proliferation, angiogenesis, and invasiveness. These features give HeyA8 an exceptionally strong capacity for migration, colony formation, and rapid tumor take, making it one of the most widely used cell lines in ovarian cancer in vitro and in vivo research.

When HeyA8 cells are implanted into the orthotopic compartment — most commonly by injection into the ovarian bursa, or alternatively into the peritoneal cavity to model advanced disseminated disease — they reproducibly generate invasive ovarian tumors with spontaneous spread to the omentum, mesentery, diaphragm, and other peritoneal surfaces, frequently accompanied by malignant ascites. This stands in sharp contrast to subcutaneous xenografts, which cannot reproduce the tumor-stroma interactions, vascular supply, or metastatic route of clinical ovarian cancer. The luciferase-tagged derivative, HeyA8-luc, further enables longitudinal bioluminescence imaging following D-luciferin administration, allowing tumor burden, response to therapy, and metastatic progression to be quantified in the same animal over time — substantially reducing animal use while improving statistical power.

Our Services

Alfa Cytology has built its ovarian cancer portfolio around technically demanding orthotopic models such as HeyA8 / HeyA8-luc, offering end-to-end study execution — from model setup and imaging-schedule design through dosing, necropsy, and data interpretation — so your team receives a complete, audit-ready preclinical dataset without diverting internal resources to animal work.

Workflow of HeyA8 / HeyA8-luc Orthotopic Mouse Model Construction

Construction of the HeyA8 / HeyA8-luc orthotopic model follows a standardized, quality-controlled sequence. Although details such as implantation route, cell number, and mouse strain can be tailored to the scientific question, the core workflow below represents the platform as routinely executed.

  1. Cell Line Revival and Quality Control: HeyA8 or HeyA8-luc cells are revived from authenticated stocks, expanded under standardized culture conditions, and released for study only after confirming STR identity, mycoplasma negativity, and — for the luciferase line — robust reporter expression in vitro.
  2. Animal Procurement and Acclimatization: Female immunodeficient mice (nude or NSG, typically 6–8 weeks of age) are received, randomized by body weight, and allowed to acclimatize for at least one week before any procedure, with daily health observations.
  3. Orthotopic Tumor Cell Implantation: Under a dissecting microscope, 1 × 106 HeyA8 or HeyA8-luc cells in a minimal volume of PBS or Matrigel/PBS are injected into the ovarian bursa to establish a primary ovarian tumor; for studies of advanced disease, an intraperitoneal inoculation format is applied instead to model peritoneal seeding directly.
  4. Baseline Imaging and Group Randomization: Mice are administered D-luciferin intraperitoneally and imaged to verify successful engraftment and obtain a baseline bioluminescence signal; animals are then distributed across treatment groups so that starting tumor burden is statistically comparable.
  5. Longitudinal Tumor Monitoring: Bioluminescence imaging is performed on a fixed schedule (typically weekly), alongside body weight measurement, abdominal girth assessment, and clinical health scoring, generating individual animal growth curves without invasive measurement.
  6. Compound Administration and Efficacy Assessment: Test articles are dosed according to the client-approved protocol (intraperitoneal, intravenous, or oral gavage routes supported), with imaging readouts capturing treatment-induced changes in photon flux, onset of ascites, and progression pattern.
  7. Endpoint Necropsy and Tissue Collection: At study termination, mice are euthanized under IACUC-aligned procedures; ascites volume is recorded, primary ovarian tumors are excised and weighed, and peritoneal implants are counted and mapped across designated anatomical sites.
  8. Histopathology and Data Delivery: Harvested tissues are fixed, sectioned, and subjected to H&E and/or immunohistochemical staining; the final report integrates imaging data, tumor metrics, and pathology findings into a structured, study-ready data package.

Case Study-HeyA8 / HeyA8-luc Orthotopic Mouse Model Development

Alfa Cytology has applied the HeyA8 / HeyA8-luc orthotopic platform in multiple preclinical engagements for biopharma and academic clients. In one representative program, luciferase-labeled HeyA8 cells were implanted orthotopically and tumor burden was tracked weekly by bioluminescence imaging through a multi-arm treatment study; the resulting dataset — encompassing photon-flux kinetics, peritoneal implant counts, ascites measurements, and confirmatory histopathology — allowed clear differentiation between treatment arms ahead of downstream candidate selection. Because every engagement is governed by confidentiality agreements, detailed results are shared only under appropriate disclosure terms; we invite you to contact us to request a relevant case summary or to discuss a study configuration matched to your therapeutic candidate.
