GL261 Syngeneic Model Service for Glioblastoma

Fig 1: GL261 Syngeneic Model for Glioblastoma preclinical research.

The GL261 syngeneic model is the most extensively characterized immunocompetent glioblastoma platform in existence, engrafting a methylcholanthrene-induced murine glioma into the brains of syngeneic C57BL/6 mice to recreate the invasive, angiogenic, and immunosuppressive hallmarks of human high-grade glioma. Alfa Cytology delivers this benchmark model as a complete preclinical service — authenticated cell banking from the NCI reference line, stereotactic intracranial implantation, scheduled bioluminescence and MRI monitoring, checkpoint-inhibitor and temozolomide-combination testing, and full survival-based histopathology — giving sponsors the field's standard-setting GBM system without building neurosurgical, imaging, and neuro-statistical infrastructure in-house.

Overview of GL261 Syngeneic Model for Glioblastoma

GL261 was born in the era of classical chemical carcinogenesis: the tumor was induced by intracranial 3-methylcholanthrene in C57BL/6 mice and carried for decades by serial intracranial and subcutaneous transplantation of tumor fragments, until stable in vitro-growing cultures were established in the mid-1990s and Ausman and colleagues formalized the model with intracranial implantation studies in 1970, reporting a median survival of 24–25 days at 105 cells per mouse. Genetically, the line carries activating K-ras and p53 mutations that drive high c-myc expression, shows EGFR upregulation with active PI3K/AKT and Ras/MAPK signaling, and is intrinsically PTEN-deficient — a constellation overlapping human glioma biology. Histologically the tumors pass through four instructive stages over about four weeks: perivascular organization, proliferation near vasculature, hypoxia as vessels degenerate, and neovascularization toward necrotic regions, yielding the pleomorphism, pseudopalisading necrosis, and angiogenesis that define GBM pathology. Owing to decades of serial passaging, the line has lost canonical glial markers such as GFAP while retaining vimentin — a documented quirk that in no way diminishes its phenotype.

Intracranial GL261 syngeneic model workflow and survival — C57BL/6 mice are inoculated intracranially with GL261 cells, vaccinated with ferroptotic GL261 cell-armed dendritic cells on days 7 and 14, and followed for survival and neurological status. What truly distinguishes GL261 is its immune dimension. The line is the most immunogenic of the murine glioma panel — very high mutational load, abundant MHC class I with limited MHC II and B7 co-stimulators, PD-L1 present and PD-L2 absent — and it has yielded some of the most consequential immunotherapy findings in glioma research. Combined PD-1 and CTLA-4 blockade produces long-term survival in over half of treated animals, dendritic-cell vaccines have achieved 60% hundred-day survival with protection against rechallenge, and triple blockade of IDO, CTLA-4, and PD-L1 reduces tumor-infiltrating Tregs in parallel with durable cures. The model also carries hard-won lessons about combination logic: standard-dose temozolomide (50 mg/kg × 5 days) raises LAG-3 and TIM-3 exhaustion markers and actually abrogates the survival benefit of PD-1 blockade, whereas metronomic dosing (25 mg/kg × 10 days) preserves it — a scheduling insight that has shaped real clinical trial design. Tumor burden in GL261 mice drives thymic and splenic involution and peripheral CD4/CD8 depletion mirroring human disease, the microenvironment is densely infiltrated by MDSCs, macrophages, and Tregs, and luciferase-tagged GL261-Luc2 enables weekly quantitative bioluminescence tracking from day 7 onward, with MRI providing anatomical correlation. A glioma stem-cell derivative, GL261-GSC, forms lethal tumors from as few as 5,000 cells and has been mapped at single-nucleus resolution, further extending the platform's utility.

Cell Line Information: GL261

Our Services

Engaging Alfa Cytology for the GL261 syngeneic model places the field's reference GBM workflow under one accountable team: authenticated GL261 or GL261-Luc2 banks are maintained at low passage under documented provenance, syngeneic C57BL/6 hosts are prepared and acclimated, tumor cells are implanted stereotactically into the striatum by neurosurgeons experienced with the coordinate system, intracranial burden is tracked by scheduled bioluminescence imaging from day 7 with MRI correlation, and treatment arms — from checkpoint combinations and DC-vaccine formats to temozolomide scheduling questions — are executed through survival endpoints and terminal brain immunohistochemistry, all under IACUC-compliant conditions and returned as an audit-ready data package.

Workflow of GL261 Syngeneic Model Construction

GL261 studies follow the definitive intracranial format in which survival is the primary endpoint, supported by quantitative imaging and deep immune profiling; the model's published benchmark windows make randomization and interim reads highly standardized. A standard program proceeds as follows.

  1. Cell preparation and quality control: GL261 or GL261-Luc2 cells are expanded at low passage, confirmed mycoplasma-free, and luciferase activity validated in vitro; single-cell suspensions at defined concentrations are prepared fresh immediately before implantation.
  2. Host selection and acclimation: Male or female C57BL/6 mice, typically 6–10 weeks of age, are quarantined and acclimated in a specific-pathogen-free barrier facility, with cohort sizing matched to the survival-powered design.
  3. Stereotactic setup and craniotomy: Under isoflurane anesthesia with temperature support, the head is fixed in a stereotactic frame, the scalp is disinfected and incised, and a burr hole is drilled over the striatal target coordinates.
  4. Intracranial implantation. A Hamilton microsyringe is lowered to the striatum, and the cell suspension: classically 105 cells in 10 μL — is infused slowly with a dwell time before withdrawal to prevent reflux; lower-inoculum or GL261-GSC formats follow the same geometry.
  5. Recovery and perioperative care: The burr hole is sealed, the scalp is closed, and mice recover on warming with analgesia and twice-daily neurological observation against predefined humane endpoints.
  6. Imaging-guided monitoring and randomization: Bioluminescence imaging after intraperitoneal D-luciferin injection is performed weekly from day 7, with T2-weighted MRI as needed; mice are randomized into arms on the basis of day-7 signal intensity to balance tumor burden.
  7. Treatment administration: Test articles, checkpoint inhibitors, vaccine formats, or temozolomide regimens are dosed per protocol — with attention to the documented interaction between temozolomide dose intensity and immunotherapy efficacy — and body weight and neurological status are scored at defined intervals.
  8. Survival analysis and terminal histopathology: Kaplan–Meier survival is the primary endpoint with log-rank analysis; endpoint brains undergo H&E and immunohistochemistry (CD31, CD8, FoxP3, F4/80, PD-L1), tumor-infiltrating leukocyte profiling by flow cytometry where specified, and rechallenge studies for cured animals, with all data consolidated into the final report.

Case Study-GL261 Syngeneic Model Development

Alfa Cytology has applied the GL261 syngeneic platform to preclinical programs spanning checkpoint blockade combinations, dendritic-cell and tumor-cell vaccine formats, temozolomide scheduling optimization, and tumor microenvironment interrogation in glioblastoma. In these engagements, weekly bioluminescence imaging quantified intracranial tumor burden while Kaplan–Meier survival served as the primary endpoint, and terminal brain immunohistochemistry with flow-cytometric tumor-infiltrating-leukocyte profiling completed the pharmacological and immunological assessment. Study parameters — inoculum format, imaging cadence, randomization intervals, and endpoint panels — are tailored to each program. Case materials can be shared under confidentiality agreements upon request.
