KLN 205 Syngeneic Model Service for Lung Cancer

Fig 1: KLN 205 Syngeneic Model for Lung Cancer preclinical research.

The KLN 205 syngeneic model service for lung cancer is an immune-competent DBA/2 platform built on the chemically induced Nettesheim squamous cell carcinoma, offering a PD-L1-high immunophenotype that complements low-PD-L1 models and supports both flank-efficacy and bioluminescence-tracked lung-metastatic study designs. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated KLN 205 / luciferase-engineered KLN 205 expansion and banking, subcutaneous or intravenous implantation into DBA/2 hosts, scheduled caliper and in vivo bioluminescence monitoring, and full histopathology with immune-microenvironment profiling — all configured around each sponsor’s regimen, combination strategy, and endpoint plan.

Overview of KLN 205 Syngeneic Model for Lung Cancer

KLN 205 descends from one of the classical chemically induced respiratory tract tumors: the Nettesheim carcinoma, produced in a male DBA/2 mouse by repeated intratracheal administration of 3-methylcholanthrene (3-MC), carried through serial intramuscular passage in DBA/2 mice, and finally established as a cell line by serial subculture and subcloning — all documented in the line’s foundational paper by Kaneko and LePage in 1978. Histologically it is a squamous cell carcinoma of tracheal-pulmonary origin, and the literature has deployed it flexibly — as a model of lung squamous carcinoma, non-small cell lung cancer, and head-and-neck-type SCC — a nomenclatural breadth that itself reflects how often the line has been used. Cytogenetically it is tetraploid with 77–82 chromosomes per cell, adherent and epithelial in culture, and deposited with the ATCC by T. Kaneko. Its practical signature in vivo is dual-mode: subcutaneous grafts grow briskly (2 × 105 cells produce palpable DBA/2 tumors within about a week), while intravenous inoculation seeds metastatic lesions throughout the lungs — the property that made the line famous.

Histopathology and immunohistochemistry of KLN 205 tumors after combination treatment — H&E and IHC panels (CD3, CD8, CD4, Granzyme B, F4/80, Ki67) across control, BJIKT, anti-PD-1 and combination groups, with quantified staining intensities confirming enhanced cytotoxic T-cell and macrophage infiltration and reduced proliferation in the combination arm. The model’s modern value rests on three documented pillars. First, its immunophenotype: KLN 205 carries high PD-L1 and low CD47 expression, the exact complement of the C57BL/6 Lewis lung carcinoma (high CD47, low PD-L1) — a pairing that mirrors the inter-patient heterogeneity of NSCLC and makes KLN 205 the rational choice for PD-L1-directed therapy evaluation, as demonstrated in antibody-nanoparticle conjugate studies where anti-PD-L1 constructs performed selectively better in KLN 205 than in LLC. Second, its metastatic imaging capability: two independently engineered luciferase clones, injected intravenously into DBA/2 mice, yield unambiguous pulmonary bioluminescence within one week that intensifies week over week — enabling longitudinal, within-animal quantification of metastatic burden and clone-to-clone comparisons of metastatic capacity. Third, its heavily benchmarked pharmacology: rapamycin suppressed subcutaneous growth from 1290 ± 173 mm³ to 246 ± 80 mm³ and cut pulmonary metastases from 8 of 8 to 1 of 6 animals; WEE1 inhibition combined with anti-PD-1 and carboplatin reduced growth and prolonged survival; and surgical debulking of established flank tumors was shown to improve subsequent anti-PD-1 outcomes — a direct preclinical rationale for the debulking-plus-checkpoint strategies now debated clinically. For sponsors, KLN 205 is thus a metabolically well-trodden, immunologically informative squamous lung platform in an often-overlooked but fully immunocompetent host.

Cell Line Information: KLN 205

The table below consolidates the key biological, cultural, and modeling characteristics of the KLN 205 line and its luciferase-engineered derivatives.

Parameter Details
Cell line designation KLN 205 (synonyms KLN-205, KLN205); luciferase-engineered clones generated for bioluminescent metastasis tracking; derivative KLN205-MUC1 documented in Cellosaurus
Species / genetic background Mouse (Mus musculus), DBA/2 inbred background; tumorigenic in DBA/2 and BDF1 hybrid hosts
Origin Derived from the Nettesheim carcinoma: 3-methylcholanthrene-induced (repeated intratracheal administration) squamous cell carcinoma arising in a male DBA/2 mouse, serially passaged intramuscularly in DBA/2 mice, and established as a cell line by serial subculture and subcloning (Kaneko & LePage, Cancer Research 38:2084–2090, 1978)
Depository / catalogue ATCC CRL-1453 (deposited by T. Kaneko); ECACC 90110519; Cellosaurus CVCL_3533; CLS 400419
Tumor type Murine squamous cell carcinoma of tracheal-pulmonary origin; deployed across the literature as a lung squamous carcinoma, NSCLC, and head-and-neck-type SCC model
Cytogenetics Tetraploid karyotype with 77–82 chromosomes per cell (earlier modal reports 73–76)
Immunophenotype High PD-L1 and low CD47 expression — the complementary pairing to the C57BL/6 Lewis lung carcinoma (high CD47, low PD-L1), together mirroring NSCLC inter-patient heterogeneity; tested negative for ectromelia virus (mousepox)
Growth properties Adherent epithelial-like cells with tight junctions and cobblestone appearance; tumorigenic with robust, reliable take in syngeneic hosts; subcutaneous 2 × 105 cells produce palpable DBA/2 tumors within about one week
Recommended culture conditions ATCC-formulated Eagle’s Minimum Essential Medium with 10% fetal bovine serum at 37 °C in 5% CO₂; numerous laboratories use DMEM or RPMI-1640 with 10% FBS; subcultivation ratio approximately 1:5, medium renewal 2–3 times weekly
Standard inoculum and routes Subcutaneous: approximately 2 × 105 cells in 0.1–0.2 mL (established tumors in 2–3 weeks); combination-immunology studies used 5 × 105 cells with randomization at ≈80–150 mm³; intravenous: luciferase-clone injections into DBA/2 mice produce pulmonary signal detectable by week 1 and intensifying through week 3
Documented in vivo responses Rapamycin 2 mg/kg/day suppressed flank tumors (246 ± 80 mm³ vs 1290 ± 173 mm³ control at day 29) and prevented pulmonary metastases (1/6 vs 8/8 controls) while cyclosporine did neither; WEE1 inhibitor (AZD1775, 20 mg/kg) with anti-PD-1 (200 μg) and carboplatin (40 mg/kg) reduced growth and prolonged survival; anti-PD-L1 antibody-nanoparticle conjugates outperformed free checkpoint antibodies; surgical debulking of established flank tumors improved subsequent anti-PD-1 response
Host strains DBA/2 and DBA/2J immunocompetent mice (syngeneic); BDF1 hybrid hosts per classical characterization
Key research applications PD-L1-directed checkpoint, bispecific, and antibody-drug conjugate evaluation in a PD-L1-high background; radiotherapy and radiosensitizer (WEE1 inhibitor) combination studies; surgical debulking plus checkpoint sequencing; longitudinal lung-metastasis quantification with luciferase clones; salivary and systemic biomarker profiling in treatment-response studies; chemotherapy screening against squamous histology

Our Services

A squamous platform whose value depends on immunophenotype and imaging discipline demands both, and Alfa Cytology delivers: authenticated KLN 205 banks with documented PD-L1/CD47 status, validated flank and intravenous formats, weekly in vivo bioluminescence for metastatic cohorts, and endpoint pathology paired with checkpoint pharmacodynamics — all under one accountable preclinical operation.

Workflow of KLN 205 Syngeneic Model Construction

Construction follows the published methodologies for both deployment formats: KLN 205 cells (parental or luciferase-engineered) are expanded under authenticated conditions, implanted subcutaneously or intravenously into DBA/2 mice, grown to a standardized baseline, randomized, treated, and analyzed with efficacy and metastatic endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: KLN 205 or luciferase-engineered derivatives are identity-verified, confirmed mycoplasma-negative, checked for PD-L1/CD47 status by flow cytometry, and expanded into low-passage working banks under standardized EMEM culture conditions.
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at the study-specified dose of approximately 2 × 105 cells (flank) or per validated metastatic protocol in 0.1–0.2 mL of PBS or a Matrigel-supported suspension.
  3. Anesthesia and implantation: DBA/2 mice are anesthetized with isoflurane; cells are implanted subcutaneously into the flank or injected intravenously via the lateral tail vein, per the study arm.
  4. Establishment monitoring and randomization: Flank tumors are measured by caliper from first palpability; metastatic arms are baseline-imaged after luciferin administration; animals are randomized once tumors reach approximately 80–150 mm³, balancing baselines across arms.
  5. Treatment administration: Experimental arms — checkpoint inhibitors and PD-L1-directed constructs, WEE1 inhibitors, carboplatin and radiosensitizer combinations, targeted agents, and surgical debulking schedules — proceed per study design with body-weight and toxicity monitoring.
  6. Longitudinal imaging and interim measurements: Caliper measurements track flank kinetics; luciferase arms are imaged weekly to quantify pulmonary signal and compare metastatic trajectories within animals.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth inhibition, metastatic burden, and survival with ex vivo organ bioluminescence for metastatic mapping and histopathology (H&E confirming squamous histology, plus Ki67, CD8, PD-L1, and marker-specific immunohistochemistry) and tumor-infiltrating-lymphocyte flow cytometry where designs require it.
