MCA205 Syngeneic Model Service for Sarcoma

Fig 1: MCA205 Syngeneic Model for Sarcoma preclinical research.

The MCA205 Syngeneic Model Service for Sarcoma offers a chemically induced, immunocompetent fibrosarcoma system that recapitulates the tumor–immune dynamics central to soft-tissue sarcoma research. Alfa Cytology supports the full study lifecycle of this model — C57BL/6 mouse inoculation, longitudinal tumor monitoring, immune-checkpoint and combination-therapy dosing, and detailed endpoint characterization — delivering dependable preclinical data for sarcoma programs.

Overview of MCA205 Syngeneic Model for Sarcoma

MCA205 is a murine fibrosarcoma cell line originally induced in C57BL/6 mice by the carcinogen 3-methylcholanthrene, then maintained in vivo through serial subcutaneous transplantation before being adapted to in vitro culture. Classified as a weakly immunogenic tumor, MCA205 occupies a valuable middle ground in immuno-oncology: it grows progressively in syngeneic hosts, yet expresses sufficient tumor-associated antigens and moderate MHC class I levels to engage the adaptive immune system. The line is therefore one of the most widely used models for inducing tumor-reactive T lymphocytes, dissecting antigen presentation, and benchmarking adoptive cell therapies and cytokine-based interventions.

Verapamil inhibits MCA205 fibrosarcoma in vitro and enhances anti-PD-1 efficacy in the MCA-205 syngeneic model: VEGF reduction, tumor growth curves, excised tumors, survival, and CD8+ T cell infiltration. (Sun, Jianwen, et al., 2024)

Following subcutaneous implantation into C57BL/6 mice, MCA205 tumors establish reliably with predictable kinetics and display notable sensitivity to immune-checkpoint blockade — responding robustly to anti-PD-1 and anti-PD-L1 antibodies and partially to standard-of-care agents such as doxorubicin — while still offering dynamic range for combination optimization. Because the host immune system remains fully functional, the model faithfully reproduces lymphoid and myeloid infiltration, CD8⁺ T-cell-mediated tumor control, and immunosuppressive microenvironment features (including myeloid-derived suppressor cells and M2 macrophages) that are difficult to study in xenograft settings. Luciferase-tagged MCA205 derivatives further enable non-invasive tracking in vivo, supporting both efficacy studies and mechanism-of-action investigations.

Our Services

Alfa Cytology runs the MCA205 syngeneic model within a dedicated preclinical in vivo facility, pairing standardized C57BL/6 colony operations with flexible study configurations — from straightforward single-agent efficacy arms to checkpoint-combination, rechallenge, and immune-profiling designs — so each sarcoma program receives data matched to its specific mechanism-of-action and decision-making needs.

Workflow of MCA205 Syngeneic Model Construction

Building a reliable MCA205 syngeneic study proceeds through a series of controlled stages designed to guarantee consistent tumor take and meaningful readouts. The workflow is outlined below:

  1. Cell Line Qualification: MCA205 cells are STR-authenticated, confirmed mycoplasma-negative, and expanded at low passage to retain growth and immunogenic characteristics.
  2. Mouse Acclimatization and Randomization: C57BL/6 mice (typically 6–8 weeks old) are acclimatized under specific-pathogen-free conditions and randomized to treatment groups.
  3. Tumor Cell Inoculation: Single-cell suspensions (commonly 0.5–2 × 106 cells in 100–200 µL) are injected subcutaneously into the flank; in vivo-luciferase variants are used where imaging is required.
  4. Model Establishment Monitoring: Implant sites are palpated and, once tumors become measurable, caliper readings begin alongside body-weight and health-status recording.
  5. Group Randomization and Dosing: When tumors reach the target volume (e.g., approximately 100 mm3), animals are re-randomized and dosing with vehicle, reference agents (e.g., anti-PD-1, anti-PD-L1), or test articles commences.
  6. In-Life Assessment: Tumor growth and body weight are tracked on a scheduled basis; bioluminescence imaging is performed for luciferase-tagged studies to follow disease burden non-invasively in vivo.
  7. Endpoint Analysis: At termination, tumors are excised and weighed, and tissues are processed for histology, flow-cytometric immune profiling, or molecular analysis as specified.
  8. Reporting: Tumor-growth curves, survival analysis, and immune-endpoint data are integrated into a final study report with statistical interpretation.

Case Study-MCA205 Syngeneic Model Development

In a recent preclinical engagement, Alfa Cytology established the MCA205 syngeneic sarcoma model for a sponsor assessing an immunomodulatory therapeutic approach. The engagement covered model setup in C57BL/6 mice, treatment-arm allocation with reference checkpoint inhibitors, scheduled in-life monitoring of tumor burden and body weight, and terminal immune-profiling of tumor tissue — yielding an integrated dataset that informed the client’s preclinical development strategy. Case formats can be adapted to single-agent, combination, rechallenge, or biomarker-oriented designs according to program requirements.
