KMS-11 / KMS-11-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: KMS-11 / KMS-11-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The KMS-11 / KMS-11-luc orthotopic mouse model disseminates one of the most genetically extreme human myeloma lines — carrying three separate immunoglobulin heavy chain translocations including t(4;14) with an activating FGFR3 Y373C mutation — into the bone marrow of immunodeficient mice, where its bortezomib-sensitive parent and JCRB-curated bortezomib-resistant derivative define both sides of the proteasome-inhibitor response axis. Alfa Cytology delivers this high-risk cytogenetic platform with translocation-verified banking, subcutaneous, intramedullary, or systemic luciferase-imaging configurations, validated FGFR3-targeting reference arms, and epigenetic-pathway endpoint panels around the MMSET/WHSC1 axis, so your kinase, proteasome, and chromatin-targeting strategies are tested against the cytogenetics that actually drive aggressive MM.

Overview of KMS-11 / KMS-11-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

KMS-11 was established by T. Otsuki from the pleural effusion of a 67-year-old Japanese woman with IgG-kappa myeloma and is curated by JCRB (JCRB1179) as a t(4;14)(p16.3;q32.3)-positive line with small round-cell morphology, partly multinucleated, that grows uniquely among myeloma lines as a largely adherent population with a minor suspension fraction. Its translocation architecture is exceptional even by MM standards: long-read sequencing characterization has resolved three distinct IgH rearrangements — t(4;14) with the classical switch-mu breakpoint juxtaposing the IGH enhancer to NSD2/MMSET, t(14;16), and a complex t(8;14) placing MYC under IGH control — more IgH rearrangements than a normal cell possesses loci. The t(4;14) lesion drives deregulated FGFR3 expression carrying the activating Y373C substitution (heterozygous, IIIc isoform), the same class of mutation found in autosomal dominant skeletal disorders, making KMS-11 a canonical substrate for FGFR3-pathway pharmacology and MMSET-driven epigenetic biology alike.

NSG female mice implanted with KMS-11 human myeloma cells (day -14), followed by T-cell infusion (day 0) and biweekly BCMA×CD3 bispecific antibody dosing (0.15 mg/kg i.v., 7 doses), showing weekly bioluminescence where the no-antibody and control groups exhibited progressive tumor signal (total flux rising to ~10¹¹ photons/sec by day 38) while BsAb treatment reduced the KMS-11-Luc tumor signal to background by day 38, with GFP/T-cell immunofluorescence (GrB, p-Lck, tubulin) confirming immunological synapse formation. In vivo, KMS-11 xenografts have anchored two distinct therapeutic literatures. On the kinase side, the FGFR3-targeting antibody R3Mab — which binds human and murine FGFR3 with similar affinity — almost completely abolished KMS-11 tumor growth, and the FGFR-selective small-molecule inhibitor PD173074 prolonged survival of KMS-11 xenograft-bearing mice, establishing FGFR3 dependence as a druggable vulnerability in vivo; KMS-11 cells are correspondingly bortezomib-sensitive in vitro, and the JCRB-curated derivative KMS-11/BTZ provides a matched bortezomib-resistant arm. On the chromatin side, the MMSET/WHSC1–NF-κB–MYC transcriptional axis that the t(4;14) rearrangement installs has been directly targeted in KMS-11 xenografts, where the SETD2-pathway inhibitor EZM0414 at 30 mg/kg twice daily produced 91% tumor-growth inhibition alone and greater than 95% with regression when combined with bortezomib at 0.5 mg/kg twice weekly — one of the cleanest published regression benchmarks in a t(4;14) model. Orthotopically, KMS-11 can be delivered systemically for marrow-homing disease or directly into bone for intramedullary designs, and the KMS-11-luc configuration adds quantitative whole-body bioluminescence in vivo with monoclonal quality control. Few MM models pack this much targetable biology — kinase, proteasome, and epigenetic writer — into one well-curated line.

Cell Line Information: KMS-11 / KMS-11-luc

KMS-11 (JCRB1179) grows in vitro in RPMI-1640 with 10% fetal bovine serum, seeded at about 5 × 104 cells/mL and subcultured twice weekly by dilution; its partial adherence is operationally distinctive — floating cells are removed, trypsin inhibitors washed away, and the adherent fraction trypsinized for passage. The line is identity-verified by JCRB with cytogenetic documentation of t(4;14). The KMS-11-luc derivative adds firefly luciferase for quantitative imaging in vivo, with monoclonal selection of high-signal clones in vitro recommended for uniform bioluminescence.

Our Services

A line with three IgH translocations and a kinase mutation demands more from its operator than a routine xenograft: translocation architecture defines which benchmarks are meaningful, partial adherence defines how cultures must be handled, and the parent–resistant pairing defines what resistance claims can credibly be made. Alfa Cytology runs KMS-11 programs with long-read-informed translocation verification at banking, adherent-fraction-aware expansion SOPs, monoclonal luciferase quality control, benchmark arms from the published FGFR3 and EZM0414–bortezomib records, and matched KMS-11/BTZ cohorts where proteasome resistance is the question — with endpoint panels spanning phospho-FGFR3, MMSET/WHSC1, NF-κB, and MYC readouts that connect tumor response to the biology driving it.

Workflow of KMS-11 / KMS-11-luc Orthotopic Mouse Model Construction

KMS-11 model construction must honor the line's two operational signatures — partial adherence and translocation-defined benchmark selection. The workflow below covers expansion, the three validated implantation configurations, and pathway-anchored endpoints.

  1. Adherent-Aware Cell Expansion and Quality Control: KMS-11 or KMS-11-luc cells are expanded in vitro in RPMI-1640 + 10% FBS, seeded at ~5 × 104 cells/mL and subcultured twice weekly by dilution; batches are confirmed mycoplasma-negative, STR-authenticated, verified for t(4;14) and FGFR3 Y373C status, and — for luciferase configurations — monoclonal high-signal clones are confirmed by in vitro assay.
  2. Host Selection and Acclimatization: NSG, NOD-SCID, or nude mice, typically 6–8 weeks of age, are acclimatized under SPF conditions and health-screened before assignment to study cohorts; where marrow engraftment is prioritized, sub-lethal irradiation may precondition recipients.
  3. Configuration Selection and Inoculation: The study design selects among three validated configurations — subcutaneous flank implantation (2–10 × 106 cells, with Matrigel where required), systemic intravenous delivery for marrow-homing disease, or direct intratibial/intrafemoral injection for bone-disease designs — with implantation day recorded for the dosing calendar.
  4. Engraftment Verification and Randomization: KMS-11-luc cohorts are imaged by bioluminescence in vivo after D-luciferin administration; systemic configurations are confirmed by marrow signal in long bones and vertebrae, subcutaneous arms by caliper, and intramedullary arms by defined radiographic or imaging thresholds, before flux- or volume-matched randomization.
  5. Treatment Administration: Test agents are dosed on study-defined schedules — benchmark arms include FGFR3-targeting antibodies or inhibitors, bortezomib at 0.5 mg/kg twice weekly, and MMSET-axis epigenetic agents — with body weight and clinical condition recorded throughout.
  6. Longitudinal Monitoring: Disease burden is tracked weekly by bioluminescence quantification in vivo and by caliper for flank cohorts; intramedullary studies add periodic micro-CT or radiographic assessment of osteolytic burden where bone endpoints are included.
  7. Survival Follow-Up: Animals are followed to humane endpoints with Kaplan–Meier analysis, and KMS-11/BTZ arms monitor on-treatment progression to evaluate resistance-overcoming strategies.
  8. Endpoint Analysis and Reporting: At termination, tumors and femoral/tibial marrow are harvested; human myeloma cells are enumerated by CD38/CD138 or idiotype flow cytometry, bone architecture is assessed by micro-CT and TRAP histomorphometry for intramedullary studies, and tissues are processed for phospho-FGFR3, MMSET/WHSC1, phospho-p65/NF-κB, and MYC immunohistochemistry alongside cleaved caspase-3 and H&E; ex vivo bioluminescence maps signal distribution, and all imaging, flow, pathway, and survival data are compiled into the final report.
