U14 Syngeneic Model Service for Cervical Cancer

Fig 1: U14 Syngeneic Model for Cervical Cancer preclinical research.

The U14 syngeneic model of cervical cancer propagates a chemically induced murine cervical squamous carcinoma in fully immunocompetent 615 or C57BL/6 hosts, pairing a metastasis-rich phenotype with a six-decade publication heritage. Alfa Cytology delivers the complete U14 workflow—authenticated cell banking, subcutaneous, footpad or intraperitoneal implantation, burden-balanced randomization, scheduled dosing and metastasis-inclusive endpoint analysis—giving sponsors decision-ready preclinical datasets shaped to their program, strictly within a preclinical scope.

Overview of U14 Syngeneic Model for Cervical Cancer

U14 is a genuine artifact of experimental oncology’s carcinogen era: in 1958, researchers at the Chinese Academy of Medical Sciences and Peking Union Medical College applied 20-methylcholanthrene to the uterine cervix of a mouse and, from the tumor that arose, established a transplantable line that has been maintained ever since. Pathology settled its identity in the 1980s as an undifferentiated squamous carcinoma of the cervix, and the classical transplant statistics remain arresting—lymph-node metastasis in 95% of recipients, lung metastasis in 80%, and a mean survival of 27 days—earning U14 its reputation as a bidirectional (lymphatic and hematogenous) metastasis strain. A modern re-establishment and characterization (Gu and colleagues, 2008) described epithelioid cells that grow adherent with partial suspension, stain positive for cytokeratin, divide every ~21.8 hours, and carry a hypotetraploid karyotype of 64–68 chromosomes, with 100% tumorigenicity in both 615 and C57BL/6 mice. The same laboratory produced a fully GFP-positive monoclonal derivative, while parallel work selected organ-preference substrains—the pulmonary line U14AP11 (cell line P11-90) and the lymphatic line U14AL10 (L10-90)—that remain useful for studying metastatic organotropism.

As a syngeneic platform, U14 has served immunotherapy research for decades, most notably the B7-1 (CD80)-transfected vaccine studies in 615 mice that demonstrated protective and therapeutic antitumor immunity, and it offers a feature rare among cervical models: HPV16 E6- and E7-transfected derivatives whose altered latency (tumor appearance extending from 5–7 to 8–14 days) and dampened metastasis make them purpose-built substrates for E6/E7-targeted strategies. Practical formats are flexible—subcutaneous flank tumors for efficacy screening, footpad implants that enrich for nodal spread, and intraperitoneal inoculation as low as 1 × 105 cells, which generates trackable ascites within days in fluorescent derivatives. Gefitinib (ZD1839) has been run through this model as an oral dose-response benchmark, confirming its utility for tyrosine-kinase-inhibitor screening. The caveats are specific and manageable: BALB/c is only partially compatible (roughly half of recipients take), the classic 615 strain is maintained by a limited circle of institutions, and the line’s brisk kinetics compress treatment windows—all outweighed, for many programs, by an immunocompetent host with a uniquely well-documented metastatic pedigree.

HPV16 E6/E7 transfectants E6+ and E7+ clones show delayed tumor appearance (8–14 days vs. 5–7 days for wild type) and reduced lymph-node (30–40% vs. 90%) and lung (10–20% vs. 60%) metastasis — substrates for E6/E7-targeted research
Vaccine heritage B7-1 (CD80)-transfected U14 vaccines elicited protective and therapeutic antitumor immunity in 615 mice
Ascites / dissemination format Intraperitoneal inoculation of as few as 1 × 105 cells (U14-GFP) produces fluorescent ascites within ~6 days
Drug-testing precedent Oral gefitinib (ZD1839) run as a dose-response benchmark in 615 mice, with tumor-growth inhibition and apoptosis measured by FCM and TUNEL
Typical subcutaneous inoculum 1 × 106 cells in 100–200 µL PBS per flank (published range 105–107 depending on format); footpad implants enrich nodal spread
Considerations BALB/c partial incompatibility; 615 strain sourcing limited to certain institutions; brisk kinetics compress dosing windows

Workflow of U14 Syngeneic Model Construction

Construction of the U14 syngeneic model is an implantation workflow whose quality rests on verified cells, strain-matched immunocompetent recipients, standardized technique and a disciplined measurement cadence. A standard engagement proceeds as follows:

  1. Source and qualify cells: U14 stocks are cleared by mycoplasma PCR and identity verification (cytokeratin-positive epithelial phenotype confirmed); U14-GFP or luciferase labeling is titrated in vitro before expansion where imaging readouts are required.
  2. Prepare recipients: 615 or C57BL/6 mice (typically 6–8 weeks old; BALB/c avoided given partial incompatibility) complete acclimation and health screening and are individually marked before implantation.
  3. Prepare the inoculum: log-phase cells are harvested, washed, counted and resuspended in PBS (commonly 1 × 107/mL for a 100 µL dose); an intraperitoneal ascites format uses as few as 1 × 105 cells.
  4. Implant: 1 × 106 cells are injected subcutaneously into the flank, or into the footpad where lymphatic spread is the readout of interest, with consistent anatomical positioning for reproducible caliper geometry.
  5. Randomize: when tumors reach palpability (typically day 5–7 for wild-type cells), animals are assigned to groups balanced by tumor volume and body weight.
  6. Dose and follow: test articles are administered on schedule; tumors are measured with calipers two to three times weekly (volume typically 0.52 × length × width2), with fluorescence or bioluminescence substituted for labeled cohorts; body weight and clinical status are logged in parallel, and survival is recorded where the design requires.
  7. Terminate and analyze: tumors are resected and weighed; draining lymph nodes, lungs and peritoneal cavity are inspected for metastasis; tissues then enter histopathology, immunohistochemistry, flow-cytometric immune profiling and molecular analysis.
