MV-4-11 / MV-4-11-luc Orthotopic Mouse Model Service for Leukemia

Fig 1: MV-4-11 / MV-4-11-luc Orthotopic Mouse Model for Leukemia preclinical research.

The MV-4-11 / MV-4-11-luc orthotopic mouse model of leukemia engrafts a genetically defined human AML — MLL-AF4 fusion plus FLT3-ITD — directly into the bone marrow niche of immunodeficient recipients, where systemic disease unfolds on a clinically relevant timeline and is quantified in real time by luciferase bioluminescence. Alfa Cytology provides this platform as an integrated preclinical service: authenticated MV-4-11 cell stewardship, luciferase reporter line preparation and release testing, tail-vein engraftment in NSG mice, scheduled whole-body imaging, treatment administration, and survival plus human-engraftment analyses that convert in vivo disease kinetics into decision-grade efficacy data.

Overview of MV-4-11 / MV-4-11-luc Orthotopic Mouse Model for Leukemia

MV-4-11 is a human biphenotypic B-myelomonocytic leukemia cell line established by Rovera and colleagues from the blast cells of a 10-year-old male patient whose leukemia carried a t(4;11) translocation, generating the KMT2A-AFF1 (MLL-AF4) fusion characteristic of pro-B acute lymphoblastic leukemia as well as monocytic AML, and an internal tandem duplication in FLT3. Because the line lacks the wild-type FLT3 allele, its growth is driven almost entirely by ligand-independent, constitutively active FLT3 signaling — an oncogene-addicted state that makes MV-4-11 the canonical in vitro and in vitro-to-in vivo model for FLT3 tyrosine kinase inhibitor screening, with inhibitors such as quizartinib producing strong antitumor responses in the model. The line grows in suspension with a reported doubling time of about 32 hours and carries the molecular baggage of adverse-risk leukemia, including TP53 mutation, supporting studies of therapy-resistant disease biology.

Orthotopically, MV-4-11-luc cells are introduced intravenously into NSG recipients, where they seed the bone marrow and then expand through peripheral blood, spleen, and liver, frequently with visceral and meningeal involvement at advanced stages. Firefly luciferase tagging — alone or combined with GFP or mCherry in constructs such as FUW-Luc-mCherry-puro — enables non-invasive quantification of leukemia burden after D-luciferin administration, with weekly imaging resolving the steep rise in photon flux that precedes clinical decline and with signal-based randomization balancing arms before dosing begins. This imaging backbone has supported a broad set of preclinical programs on the MV-4-11-luc platform, including FLT3 inhibitors, menin-MLL interaction blockers such as MI-3454 (where once-daily oral dosing arrested leukemia progression even in established disease), HDAC inhibitors, venetoclax combinations, and mutant-p53-directed agents evaluated in an R248W-luciferase variant of the line.

Our Services

MV-4-11-luc is a demanding model — fast kinetics, tight imaging windows, and readouts that punish inconsistency — and Alfa Cytology is structured to meet those demands. The team works from authenticated CRL-9591 stocks, releases only reporter lines with verified luminescence uniformity, and runs engraftment-to-randomization schedules that land treatment arms on matched baseline signals. From first imaging session through Kaplan-Meier survival analysis and human CD45 engraftment quantification, Alfa Cytology keeps every phase of the MV-4-11 / MV-4-11-luc orthotopic workflow under single-point accountability.

Workflow of MV-4-11 / MV-4-11-luc Orthotopic Mouse Model Construction

Constructing the MV-4-11-luc orthotopic model is a precision exercise in timing: engraftment is brisk, burden climbs steeply, and the gap between “too early to treat” and “too advanced to rescue” is measured in days. The workflow below sequences cell release, intravenous delivery, imaging, randomization, and analysis so that each decision point lands on schedule.

  1. Authentication and reporter release testing: MV-4-11 / MV-4-11-luc stocks are expanded at low passage from authenticated banks, mycoplasma-tested, and released only after verifying uniform luciferase (and fluorescent reporter, where applicable) expression by serial in vitro luminescence assays.
  2. Engraftment cell preparation: Log-phase suspension cultures are harvested, washed, counted, and resuspended in sterile PBS at the challenge dose (typically 0.5×106 to 1×106 cells in approximately 100 µL per mouse).
  3. Recipient preparation and tail-vein inoculation: Female NSG mice are acclimatized, body-weight stratified, and injected intravenously via the lateral tail vein using warming-assisted vessel dilation for consistent, trauma-minimized delivery.
  4. Engraftment confirmation and baseline imaging: Within the first one to two weeks, intraperitoneal D-luciferin (150 mg/kg) and whole-body imaging confirm bone-marrow seeding and quantify starting leukemia burden per animal.
  5. Signal-matched randomization and treatment initiation: Mice are allocated to arms with equivalent photon flux — commonly around day 9 post-engraftment for standard schedules — and dosing begins per protocol (oral agents, intraperitoneal compounds, or combination regimens).
  6. Serial imaging and interim monitoring: Whole-body bioluminescence is repeated at protocol-defined intervals, tracking each animal’s flux trajectory while body weight and clinical signs define humane care throughout the treatment course.
  7. Survival analysis and terminal characterization: Morbidity-defined endpoints feed Kaplan-Meier survival analysis; at termination, spleen and liver are weighed and imaged ex vivo, peripheral blood and bone marrow are analyzed for human CD45 chimerism by flow cytometry, and tissues are preserved for histopathology and molecular studies.

Case Study-MV-4-11 / MV-4-11-luc Orthotopic Mouse Model Development

Alfa Cytology has applied the MV-4-11-luc orthotopic platform in preclinical programs addressing FLT3 signaling, MLL-fusion biology, epigenetic modulation, and combination strategies in AML. Across these programs, weekly bioluminescence sessions charted the rise and suppression of systemic leukemia burden, signal-matched randomization ensured balanced starting conditions across arms, and end-of-study analyses combined Kaplan-Meier survival with human CD45 flow-cytometric chimerism in blood, bone marrow, and spleen. Where programs required deeper characterization, ex vivo organ imaging and histopathology localized disease involvement. Program-specific designs and results are handled as sponsor-confidential deliverables, and representative outlines can be provided upon request.
