The LM8 Syngeneic Model Service for Sarcoma provides a fully immunocompetent, spontaneously metastasizing osteosarcoma platform designed for rigorous preclinical evaluation of candidate therapeutics. Alfa Cytology delivers complete LM8 model construction and study execution — from cell preparation and C3H mouse inoculation to tumor monitoring, lung metastasis quantification, and tailored efficacy endpoint analysis — giving your sarcoma program a dependable in vivo foundation.

Overview of LM8 Syngeneic Model for Sarcoma

LM8 is a murine osteosarcoma cell line established by Asai and colleagues in 1998 through eight consecutive rounds of in vivo selection: parental Dunn osteosarcoma cells were repeatedly injected into the tail vein of syngeneic C3H mice, and cells harvested from resulting lung metastases were re-injected until pulmonary colonization occurred with 100% efficiency. Compared with the low-metastatic Dunn line, LM8 cells display faster growth, a fibroblast-like morphology with prominent filopodia, and elevated alkaline phosphatase (ALP) activity, vascular endothelial growth factor (VEGF) expression, and matrix metalloproteinase (MMP-2 and MMP-9) activity. These features, together with activated Cdc42 signaling and focal adhesion kinase (FAK) autophosphorylation, drive a mesenchymal mode of migration that underlies the line’s striking metastatic behavior.

(Kondo, Hiroya, et al., 2025)

When implanted into immunocompetent C3H hosts, LM8 cells form aggressive primary tumors following subcutaneous, intraosseous, or intravenous administration, and they reproducibly disseminate to the lung — the dominant metastatic site in human osteosarcoma — with metastases detectable within approximately four weeks. Because the model preserves an intact immune system, it captures tumor–immune interactions that xenografts in immunodeficient mice cannot, making it a benchmark platform for studying metastasis mechanisms, tumor dormancy, angiogenesis, and immunotherapeutic strategies in vitro and in vivo. Reporter-tagged derivatives such as LM8/luc and lacZ-labeled LM8 further allow non-invasive and single-cell-resolution tracking of metastatic dissemination.

Our Services

Alfa Cytology provides end-to-end LM8 syngeneic model services within a specialized preclinical in vivo pharmacology facility, combining authenticated cell resources, standardized C3H mouse colony management, and flexible study designs that accommodate subcutaneous, orthotopic, and experimental metastasis formats — so your sarcoma efficacy, mechanism-of-action, and combination-therapy questions are addressed with reproducible, publication-quality data.

Workflow of LM8 Syngeneic Model Construction

Construction of the LM8 syngeneic model follows a standardized, quality-controlled process that ensures robust tumor take and reproducible metastatic progression. The typical workflow comprises the following stages:

  1. Cell Line Verification and Expansion: LM8 cells are authenticated (e.g., STR profiling), tested for mycoplasma contamination, and expanded at low passage to preserve tumorigenicity and metastatic potential.
  2. Animal Acclimatization and Grouping: C3H mice (typically 5–8 weeks old) are acclimatized under pathogen-free conditions and randomly assigned to study groups according to the experimental design.
  3. Tumor Cell Inoculation: LM8 cells are harvested, viability-checked, and administered subcutaneously into the flank (commonly 1–10 × 106 cells), into the proximal tibia for orthotopic bone tumors, or via the tail vein for experimental lung metastasis.
  4. Tumor Development Monitoring: Primary tumor growth is measured with calipers, and body weight and general health status are recorded at regular intervals to track model establishment.
  5. Treatment Initiation: Once tumors reach the predefined size (e.g., approximately 100 mm3 for efficacy studies), animals are randomized into vehicle and treatment arms, and dosing of test articles begins.
  6. In-Life Assessments: Tumor volume, body weight, and clinical signs are monitored throughout the dosing period; bioluminescence imaging can be applied for LM8/luc models to follow metastatic burden non-invasively in vivo.
  7. Endpoint Necropsy and Sample Collection: At study termination, primary tumors are excised and weighed, and lungs and other organs are harvested for metastasis analysis by surface nodule counting, India ink staining, or histology.
  8. Data Analysis and Reporting: Tumor growth curves, metastatic foci counts, and statistical comparisons are compiled into a comprehensive study report with interpretive commentary.

Case Study-LM8 Syngeneic Model Development

In a representative preclinical collaboration, Alfa Cytology established the LM8 syngeneic sarcoma model for a sponsor evaluating a novel therapeutic strategy directed against osteosarcoma metastasis. The program encompassed model setup in C3H mice, treatment-group randomization, scheduled in-life monitoring, and terminal analysis of primary tumor burden together with quantification of pulmonary metastatic foci, delivering a complete dataset that supported the client’s go/no-go decision-making for subsequent preclinical development. Additional case formats — including orthotopic and reporter-imaging study designs — are available, and study parameters can be tailored to specific compound classes and mechanism-of-action questions.
