H22 Syngeneic Model Service for Liver Cancer

Fig 1: H22 Syngeneic Model for Liver Cancer preclinical research.

The H22 syngeneic model is the most published immunocompetent platform for hepatocellular carcinoma research, engrafting a murine hepatoma line — maintained for decades as an ascites tumor before solid and orthotopic formats were standardized — into BALB/c hosts with rapid, reproducible growth and documented checkpoint responsiveness. Alfa Cytology delivers this workhorse model as a complete preclinical service — authenticated cell banking with the line's ascites-passage tradition, subcutaneous, intraperitoneal ascites, and orthotopic left-lobe liver implantation formats, scheduled monitoring, and full histopathology — giving sponsors a fast, flexible HCC system without managing strain-matched husbandry and hepatic microsurgery in-house.

Overview of H22 Syngeneic Model for Liver Cancer

H22 is arguably the most extensively used murine hepatoma line in the world, and its biography explains why: it was derived from the ascites of a mouse bearing hepatocellular carcinoma and propagated for generations as an ascites tumor — intraperitoneal passage from mouse to mouse, with tumor cells harvested from the peritoneal fluid — before laboratories standardized solid flank tumors and orthotopic liver implants. That ascites lineage gives the line an unusual personality: it grows in suspension with lymphoblast-like morphology, proliferates explosively (a 25-fold expansion in a week documented for a derived subline), secretes hepatocyte growth factor, and forms tumors through virtually every route — subcutaneous, intraperitoneal, intravenous, portal vein, and direct hepatic implantation. The standard syngeneic host is BALB/c, in which subcutaneous inoculation of 106 cells in 50 μL produces palpable tumors within 4–6 days at take rates above 95%, with in vivo doubling on the order of 3–4 days. Orthotopically, 5 × 105 cells implanted into the left liver lobe recreate the hepatic microenvironment — and the orthotopic setting measurably reshapes the immune landscape, expanding regulatory T cells and myeloid-derived suppressor cells in tumor, spleen, and bone marrow just as human HCC does.

Characterization of the half-life-extended anti-CTLA-4 VHH validated in the H22 syngeneic hepatoma model — H11 and H11-HLE construct design, binding to mouse CTLA-4 (EC50 0.048 nM and 0.064 nM), and plasma/tumor pharmacokinetics showing H11-HLE achieves 53 h plasma and 79 h tumor half-life versus 2.6 h and 4.5 h for H11, underpinning its anti-tumor efficacy in H22 tumor-bearing BALB/c mice. (Sato, Yosuke, et al., 2020)

The model's publication record spans virtually every drug class tested against liver cancer. Checkpoint blockade is validated — anti-PD-1 at 5 mg/kg weekly for three doses is a published reference regimen in the flank model — and a landmark Science Translational Medicine study used the H22 orthotopic platform to show that the HDAC8 inhibitor PCI-34051 potentiates anti-PD-L1 therapy, with combination-treated animals surviving beyond fifteen months and resisting rechallenge. H22 has carried oncolytic herpes virotherapy delivered intravenously against orthotopic disease, sorafenib-derivative evaluation, AFP-cDNA dendritic-cell vaccination, microRNA mechanism studies, hepatic stellate cell co-implantation experiments that expose how activated stellate cells drive angiogenesis and immune suppression, and portal-vein injection models of metastatic spread. Two niche roles deserve note: the line is the default vehicle for hepatoprotective and natural-product pharmacology in the Chinese literature, and the derived H22-F25/L subline — selected through a lymphatic metastasis model — forms tumors in 615 mice with 50% lymph node and 10% lung metastasis rates, extending the family's metastatic utility. Luciferase-tagged H22-Luc and H22-Luc-GFP derivatives enable quantitative bioluminescence tracking of flank and orthotopic disease, and one honest caution from the recent literature: H22 is BALB/c-lineage, so studies run in C57BL/6 hosts risk allogeneic confounding — a design question Alfa Cytology addresses by defaulting to BALB/c unless a program explicitly requires otherwise.

Cell Line Information: H22

H22 (Hepatoma-22; Cellosaurus CVCL_H613; Cytion 305163; National Infrastructure of Cell Line Resource, Beijing) is a murine hepatocellular carcinoma line with an ascites-tumor lineage, distributed by Chinese national repositories and international cell banks. Luciferase- and fluorescent-protein-tagged derivatives (H22-Luc; H22-Luc-GFP-Puro, lentiviral, puromycin-selected) support optical tracking studies. Core characteristics are compiled below.

Parameter Description

Our Services

Engaging Alfa Cytology for the H22 syngeneic model consolidates a complete, IACUC-compliant program under one accountable team: authenticated, low-passage H22 banks are maintained — honoring the line's ascites-passage tradition and suspension-culture discipline — BALB/c hosts are sourced from validated breeders and acclimated, tumors are established by subcutaneous, intraperitoneal ascites, or orthotopic left-lobe liver routes at documented take rates, and treatment arms from checkpoint inhibitors to natural-product formulations and oncolytic vectors run through scheduled caliper and bioluminescence monitoring to terminal hepatic and pulmonary histopathology, all returned as an audit-ready data package.

Workflow of H22 Syngeneic Model Construction

H22 studies offer three formats — flank, ascites, and orthotopic liver — united by the line's suspension-culture discipline and BALB/c strain fidelity. A standard program proceeds as follows.

  1. Cell preparation and quality control: H22 cells (or H22-Luc derivatives) are expanded in suspension culture, confirmed mycoplasma-free with bacterial, fungal, and Helicobacter screening where the ascites tradition demands, and luciferase activity validated in vitro; inocula are prepared fresh at defined concentrations.
  2. Host selection and acclimation: Female BALB/c mice, typically 6–8 weeks of age, are sourced from validated breeders, quarantined, and acclimated in a specific-pathogen-free barrier facility, with C57BL/6 substitution avoided given the documented allogeneic confound.
  3. Implantation: subcutaneous route. 106 cells in 50 μL sterile PBS are injected into the flank; the site is compressed briefly on needle withdrawal, and palpable tumors typically appear within 4–6 days.
  4. Implantation: ascites route. Cells in 0.2 mL saline (classically 1 × 107 cells/mL) are injected intraperitoneally; on the standard 7-day cycle, ascites is harvested aseptically for passage, counts, or direct study of malignant effusion biology.
  5. Implantation: orthotopic liver route. Under isoflurane anesthesia, a left subcostal laparotomy exposes the left lateral lobe, and 5 × 105 cells in 20 μL PBS are injected into the hepatic parenchyma with a fine needle; the lobe is returned and the abdomen closed in layers.
  6. Randomization and treatment: Mice are randomized at a defined tumor volume (for example day 7 for flank tumors) or post-implantation interval for orthotopic cohorts, and test articles — checkpoint inhibitors, oncolytic viruses, targeted agents, or natural-product formulations — are dosed per protocol with body weight and clinical signs recorded.
  7. Longitudinal monitoring: Flank tumors are measured by caliper every 2–3 days (volume = length × width2 / 2); luciferase-tagged cohorts undergo bioluminescence imaging after intraperitoneal D-luciferin injection, and orthotopic studies track survival against 100-day monitoring windows.
  8. Endpoint analysis and reporting: At termination, tumors and livers are resected and weighed; tissues undergo H&E with hepatic histology, immunohistochemistry (CD8, FoxP3, F4/80, CD31, PD-L1 panels), flow-cytometric tumor-infiltrating-leukocyte and bone-marrow/splenic immune profiling where specified, and metastatic organ screening, with all data consolidated into the final report.
