Karpas299 / Karpas299-luc Orthotopic Mouse Model Service for Lymphoma

Fig 1: Karpas299 / Karpas299-luc Orthotopic Mouse Model for Lymphoma preclinical research.

The Karpas299 / Karpas299-luc orthotopic mouse model disseminates the founding human anaplastic large cell lymphoma line — the very cell line in which the NPM-ALK fusion was first cloned — through the hematolymphoid system of immunodeficient mice, carrying both the ALK addiction that defines this disease and the CD30 surface antigen that makes it druggable by antibody–drug conjugates. Alfa Cytology delivers this oncogene-driven platform with subcutaneous, systemic, and luciferase-imaging configurations, benchmark-validated ALK inhibitor arms from crizotinib to next-generation compounds, paired crizotinib-resistant Karpas-299-CR06 cohorts, and CD30-directed ADC capability, so your targeted agents, combinations, and resistance strategies are tested against the lymphoma model the ALK field was built on.

Overview of Karpas299 / Karpas299-luc Orthotopic Mouse Model for Lymphoma

Karpas 299 was established by Abraham Karpas at the University of Cambridge from the peripheral blood of a male patient with Ki-1 (CD30)-positive anaplastic large cell lymphoma, and published in 1991; its place in history was secured three years later when Morris and colleagues cloned the t(2;5)(p23;q35) translocation from these very cells, identifying the NPM-ALK fusion oncogene that now defines ALK-positive lymphoma. The line is a suspension lymphoblast of T-cell lineage expressing the full ALCL signature — CD30, anaplastic lymphoma kinase, EMA, and CD25 — and it carries a homozygous TP53 p.Arg273Cys mutation. Its oncogene addiction is absolute and measurable: crizotinib inhibits NPM-ALK phosphorylation in vitro at 24 nM and proliferation at roughly 30 nM, and the signaling cascade downstream — PLCγ, STAT3, and AKT — collapses in step with the fusion kinase, giving every efficacy experiment a built-in pharmacodynamic readout.

Graphical abstract summarizing the deuterated ALK inhibitor AMX6001 with Karpas-299 in vitro IC₅₀ of 7.6 nM and oral in vivo efficacy in ALCL Karpas-299 xenografts achieving 95.8% tumor growth inhibition, outperforming the reference standard ceritinib. (Das, Debasis, et al., 2024)

In vivo, Karpas299 xenografts have anchored the entire ALK inhibitor development ladder. In SCID-beige mice, oral crizotinib at 100 mg/kg daily eradicated all tumors within fifteen days, with response tightly correlated to NPM-ALK phospho-inhibition and apoptosis in tumor tissue; subsequent benchmarks include dose-dependent tumor-growth inhibition by ceritinib, the deuterated next-generation inhibitor AMX6001 achieving 95.8% TGI at 25 mg/kg daily oral dosing and outperforming ceritinib, synergistic crizotinib–everolimus combinations (combination index below one in vitro and superior antitumor effect in vivo), and marked tumor shrinkage plus prolonged control when the anti-CD30 antibody–drug conjugate brentuximab vedotin is combined with ALK inhibitors. The model has even served ADC chemistry development, benchmarking stable maleimide linker designs in Nature Biotechnology. A crizotinib-selected resistant derivative, Karpas-299-CR06, extends the platform for resistance-overcoming studies, and the Karpas299-luc configuration enables quantitative whole-body bioluminescence in vivo for systemic disease designs. Few lymphoma models offer this combination of a single defining oncogene, two orthogonal druggable antigens (ALK and CD30), and three decades of head-to-head inhibitor benchmarks.

Cell Line Information: Karpas299 / Karpas299-luc

Karpas299 (ECACC 06072604; RRID CVCL_1324) grows in suspension in vitro in RPMI-1640 supplemented with 10–20% fetal bovine serum, doubling every one to two days, and remains the most widely cited ALK-positive ALCL substrate in oncology. Its identity is verifiable on multiple levels — STR profile, NPM-ALK fusion transcript by RT-PCR, CD30 and ALK immunostaining — and each of these becomes a study-level quality control attribute at in vivo program entry. The Karpas299-luc derivative adds firefly luciferase for quantitative imaging in vivo, with monoclonal selection of high-signal clones in vitro recommended for uniform bioluminescence.

Our Services

ALK programs live and die by pharmacodynamic proof: an agent that shrinks a tumor without suppressing NPM-ALK phosphorylation has not yet earned its conclusion. Alfa Cytology structures every Karpas299 study around that principle — fusion-transcript and CD30/ALK immunophenotype verification at banking, monoclonal luciferase quality control for imaging cohorts, benchmark arms spanning the published inhibitor ladder from crizotinib to AMX6001, and matched Karpas-299-CR06 cohorts where resistance is the question — with endpoint panels that pair tumor kinetics in vivo against phospho-ALK, STAT3, and AKT immunohistochemistry.

Workflow of Karpas299 / Karpas299-luc Orthotopic Mouse Model Construction

Karpas299 model construction follows compact, well-benchmarked protocols shared across the ALK inhibitor literature, with the study design organized around on-target pharmacodynamics. The workflow below details construction, imaging-guided staging, and mechanism-anchored endpoints.

  1. Cell Expansion and Multilevel Quality Control: Karpas299 or Karpas299-luc cells are expanded in vitro in RPMI-1640 + 10–20% FBS to the logarithmic phase; batches are confirmed mycoplasma-negative, STR-authenticated, verified for NPM-ALK fusion transcript and CD30/ALK expression, and — for luciferase configurations — monoclonal high-signal clones are confirmed by in vitro luminescence assay.
  2. Host Selection and Acclimatization: BALB/c nude, SCID-beige, or NSG mice, typically 5–7 weeks of age, are acclimatized for approximately one week under SPF conditions and health-screened before entering the study cohort.
  3. Route Selection and Inoculation: The disease configuration is chosen — subcutaneous flank implantation (classically 5–10 × 106 cells in ~100 µL PBS, sometimes with Matrigel) for localized benchmarking, or intravenous/intraperitoneal delivery for systemic disseminated designs — with implantation day recorded for the dosing calendar.
  4. Engraftment Verification and Randomization: Systemic disease is confirmed by whole-body bioluminescence imaging in vivo after D-luciferin administration once signal exceeds background; flank tumors are staged by caliper to a predefined volume, and mice are randomized into vehicle, benchmark inhibitor (e.g., crizotinib), and investigational or combination arms with matched starting burdens.
  5. Treatment Administration: Test agents are dosed on study-defined schedules — oral ALK inhibitors at benchmark or escalating doses, everolimus-class mTOR partners, or anti-CD30 antibody–drug conjugates alone and in combination — with body weight and clinical condition recorded throughout.
  6. Longitudinal Monitoring: Tumor burden is tracked twice weekly by caliper (flank) or bioluminescence quantification (systemic luciferase designs) in vivo, and plasma is collected serially where fusion-transcript kinetics are included as a circulating biomarker readout.
  7. Survival Follow-Up: Animals are followed to humane endpoints with Kaplan–Meier analysis, reflecting the survival designs used in published ceritinib and combination studies in this model family.
  8. Endpoint Analysis and Reporting: At termination, tumors and spleens are harvested and weighed, disseminated organs are inspected and imaged ex vivo for luciferase studies, and tissues are processed for phospho-ALK, STAT3, and AKT immunohistochemistry confirming on-target engagement, alongside CD30 staining, TUNEL apoptosis assessment, and H&E; tumor-growth inhibition, bioluminescence kinetics, pharmacodynamic markers, and survival data are compiled into the final report.
