Pan02 (Panc02) Syngeneic Model Service for Pancreas Cancer

Fig 1: Pan02 (Panc02) Syngeneic Model for Pancreas Cancer preclinical research.

The Panc02 syngeneic model service for pancreas cancer is an immune-competent C57BL/6 platform built on the 1984 Corbett carcinogen-induced ductal adenocarcinoma — a poorly differentiated, broadly chemo-resistant tumor that remains the most widely used syngeneic pancreatic cancer system across subcutaneous, orthotopic, and metastatic study formats. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated Panc02 / Panc02-H7 / luciferase-engineered expansion and banking, Matrigel-cushioned orthotopic implantation, scheduled caliper and in vivo bioluminescence monitoring, and full ductal histopathology with immune-microenvironment and senescence pharmacodynamics — all configured around each sponsor’s chemotherapy, vaccine, checkpoint, stromal, or combination hypothesis.

Overview of Panc02 (Pan02) Syngeneic Model for Pancreas Cancer

Panc02 is the founding syngeneic line of pancreatic ductal adenocarcinoma research, created in 1984 by Corbett and colleagues at the Southern Research Institute through a piece of deliberately primitive technology: cotton threads saturated with the carcinogen 3-methylcholanthrene were implanted directly into the pancreas of female C57BL/6 mice, and the resulting transplantable ductal adenocarcinomas — Panc02 and its sibling Panc03 — became the field’s first durable PDAC tools. The line is poorly differentiated, polygonal and epithelial, and notoriously insensitive to a broad range of cytotoxic agents — a chemo-resistant phenotype that usefully mimics one of human PDAC’s defining clinical traits. One documented caveat frames modern use: Panc02 lacks the canonical human PDAC driver mutations, with no KRAS, TP53, or CDKN2A/p16 lesions and a documented SMAD4 deletion (with KRAS single-nucleotide and SMAD4 mutation reports appearing in the deeper characterization literature) — so genotype-dependent questions are today routed to KPC-derived lines, while Panc02 retains its role for immune, stromal, delivery, and chemoresistance studies.

In vivo, the model’s breadth is its signature. Subcutaneous grafts give rapid, measurable tumors for screening; orthotopic implantation into the pancreatic tail — where the thin pancreatic parenchyma makes cell-suspension extravasation a documented failure mode, prompting the Matrigel-cushion technique developed by Partecke and colleagues — supports bioluminescence-tracked growth and local invasion; and the in vivo-selected Panc02-H7 derivative, established by serial passage through metastatic sites, provides an aggressively metastatic counterpart with distinct TGF-β/SMAD defects. The immunotherapy record is deep and mechanistically rich: intratumoral dendritic-cell vaccination with tumor-RNA-pulsed, α-galactosylceramide-loaded, or glycoepitope-armed DCs proved superior to subcutaneous or intravenous routes in the orthotopic setting; and a systematic CTLA-4 dose-titration study quantified myeloid-derived suppressor cells and tumor-cell PD-L1 while mapping antitumor effect against immune-related adverse events for local versus systemic administration — an early template for checkpoint-delivery design. Gemcitabine at 50 mg/kg weekly is the standard benchmark positive control, and contemporary studies extend the platform into senescence-targeting combinations (etoposide-induced senescence modulated by the Bcl-2 inhibitor navitoclax). Luciferase- and GFP-engineered derivatives support quantitative burden tracking across all formats. For sponsors, Panc02 remains the fastest route into an immune-competent PDAC microenvironment — with its genotype caveat stated plainly and its matched H7 metastatic arm available when dissemination is the question.

Cell Line Information: Panc02 (Pan02)

The table below consolidates the key biological, genetic, and modeling characteristics of the Panc02 line, its Panc02-H7 metastatic derivative, and its engineered variants.

Parameter Details
Cell line designation Panc02 (synonyms Pan02, Panc-02, Panc 02, Panc02-H0); highly metastatic derivative Panc02-H7; luciferase/GFP derivatives (Pan-02-Luc, Panc02-luc, Panc02-Luc-GFP-Puro) and engineered knockout lines documented
Species / genetic background Mouse (Mus musculus), C57BL/6 inbred background; syngeneic in C57BL/6 immunocompetent hosts
Origin Established by Corbett et al. (Cancer Research 44:717–726, 1984) from a transplantable ductal adenocarcinoma induced by implantation of 3-methylcholanthrene-saturated cotton threads into the pancreas of female C57BL/6 mice (sibling line Panc03)
Depository / catalogue Cellosaurus CVCL_D627 (Panc02), CVCL_D628 (Panc02-H7), CVCL_D7FV (Pan-02-Luc), CVCL_A8QL (Panc02-luc); distributed historically through NCI-Frederick and the MD Anderson channel
Tumor type Murine pancreatic ductal adenocarcinoma, poorly differentiated; polygonal epithelial morphology
Genetic profile (documented caveat) Lacks the canonical human PDAC driver mutations — no KRAS, TP53, or CDKN2A/p16 mutations; SMAD4 deletion documented; the deeper characterization literature reports a KRAS single-nucleotide polymorphism and SMAD4 mutations distinguishing Panc02 from Panc02-H7 (Wang et al., 2012) — genotype-dependent studies should consider KPC-derived alternatives while immune/stromal/delivery questions remain Panc02’s strength
Panc02-H7 derivative Highly metastatic variant established by in vivo selection of Panc02 through metastatic sites (Wang et al., International Journal of Pancreatology 29:37–46, 2001); more aggressive phenotype with distinct TGF-β/SMAD-pathway defects and shorter host survival than parental Panc02
Growth properties and chemoresistance Adherent epithelial cells with rapid proliferation and reliable syngeneic take; documented insensitivity to a wide range of cytotoxic agents — a chemo-resistant phenotype mirroring human PDAC
Recommended culture conditions RPMI-1640 with 10% fetal bovine serum and penicillin/streptomycin (most common formulation); DMEM with 10% FBS also widely used — medium fixed per validated laboratory protocol; mycoplasma and viral contamination testing documented
Implantation formats and doses Subcutaneous: approximately 5 × 105 cells in 50 μL for flank efficacy studies; orthotopic: approximately 5 × 105 cells injected into the pancreatic tail with a Matrigel cushion to prevent extravasation and intra-abdominal seeding (Partecke technique); luciferase formats tracked by in vivo bioluminescence from early timepoints
Documented in vivo responses Gemcitabine 50 mg/kg weekly from day 8 is the standard positive control in orthotopic models; intratumoral DC vaccination (tumor-RNA-pulsed, α-galactosylceramide-loaded, or C-ter-J28 glycoepitope-armed) superior to subcutaneous or intravenous routes; CTLA-4 local-versus-systemic dose titration mapped antitumor effect against MDSC burden, tumor PD-L1, and immune-related adverse events; senescence-induction (etoposide, irradiation) with senolytic modulation (navitoclax) benchmarked in vitro and in vivo
Host strains C57BL/6 immunocompetent mice (syngeneic); immunodeficient hosts for designated xenograft-control arms
Key research applications Chemotherapy and combination-regimen screening against a resistant phenotype; orthotopic vaccine and checkpoint-delivery studies with intratumoral-route comparisons; stromal and delivery-platform (nanoparticle, oncolytic virus) evaluation; metastasis biology with the Panc02-H7 matched arm; senescence and senolytic-therapy studies; comparative benchmarking against KPC-derived syngeneic lines

Our Services

A platform with a stated genotype caveat and three validated formats rewards exactly the operational candor Alfa Cytology brings: authenticated Panc02 and H7 banks with documented genetic screening, Matrigel-cushioned orthotopic technique, bioluminescence-tracked monitoring, and endpoint pathology pairing ductal morphology with MDSC/PD-L1 and senescence pharmacodynamics — all under one accountable preclinical operation.

Workflow of Panc02 (Pan02) Syngeneic Model Construction

Construction follows the Corbett/Partecke methodology: Panc02 or derivative cells are expanded under authenticated conditions, implanted subcutaneously or orthotopically into C57BL/6 mice, tracked by caliper and bioluminescence, treated after standardization, and analyzed with PDAC-appropriate efficacy and immune endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: Panc02 or derivative lines are identity-verified (STR), confirmed mycoplasma-negative, and documented for the genetic profile relevant to the study (including the KRAS/TP53/CDKN2A wild-type status and SMAD4 deletion caveat), then expanded into low-passage working banks under the validated RPMI-1640 or DMEM formulation.
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at approximately 5 × 105 cells in 50 μL of complete medium; for orthotopic arms, cells are mixed 1:1 with cold Matrigel to form the implantation cushion.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane; cells are implanted subcutaneously into the flank, or injected into the exposed pancreatic tail through a small abdominal approach with the Matrigel cushion limiting extravasation.
  4. Establishment monitoring and randomization: Flank tumors are measured by caliper from first palpability; orthotopic arms are tracked by in vivo bioluminescence after D-luciferin administration; animals are randomized once tumors reach protocol-specified baselines.
  5. Treatment administration: Experimental arms — gemcitabine and cytotoxic combinations, intratumoral or systemic DC vaccines, checkpoint inhibitors with local-versus-systemic dosing, stromal and senolytic agents — proceed per study design with body-weight and toxicity monitoring.
  6. Immune and pharmacodynamic monitoring: Tumor and draining-node infiltrates are profiled for MDSCs, Tregs, CD8⁺ T cells, and PD-L1 status by flow cytometry; senescence-arm studies quantify SA-β-gal activity and Cdkn1a/Mdm2 expression in explanted tumors.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth inhibition, bioluminescence kinetics, and survival with histopathology (H&E confirming poorly differentiated ductal adenocarcinoma, plus Ki67, α-SMA for stroma, CD8, PD-L1, and marker-specific immunohistochemistry) and ex vivo metastatic mapping where designs require.
