MNNG/HOS / MNNG/HOS-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: MNNG/HOS / MNNG/HOS-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The MNNG/HOS / MNNG/HOS-luc orthotopic mouse model engrafts the chemically transformed malignant member of the classic TE85 progression series into the tibial marrow cavity of immunodeficient mice, delivering rapid primary bone tumor growth with a defined, moderate metastatic signature. Alfa Cytology provides this model as a complete preclinical service — authenticated human cell banking, precision intratibial microsurgery, scheduled X-ray and bioluminescence monitoring, therapeutic intervention, and comprehensive terminal histopathology — giving sponsors a fast-growing, well-characterized bone-sarcoma platform without assembling specialized surgical and imaging teams internally.

Overview of MNNG/HOS / MNNG/HOS-luc Orthotopic Mouse Model for Sarcoma

MNNG/HOS occupies a special place in cancer biology: it is one of the first human cell lines ever created by deliberate chemical carcinogenesis. Isolated by J. S. Rhim and reported in Nature in 1975, the line was generated by treating HOS — a line originally cultured from the tibial osteosarcoma of a 13-year-old Caucasian female — with 0.01 μg/mL MNNG (N-methyl-N′-nitro-N-nitrosoguanidine), a potent carcinogenic nitrosamine. The treatment converted a relatively indolent cell population into a genuinely malignant one: MNNG/HOS grows to high saturation density, forms colonies efficiently in soft agar, carries a hypertriploid karyotype with a modal chromosome number of 70–71, is of G6PD type B, and is strongly tumorigenic in nude mice, producing tumors at 100% frequency within about three weeks after subcutaneous inoculation. Mechanistically, the transformation is attributed to activation of the c-Met oncogene — encoding the receptor tyrosine kinase for hepatocyte growth factor — with downstream engagement of PI3K and MEK/MAPK signaling. A cell-cycle-dependent pattern of telomerase activity in this line was among the early observations linking telomere dynamics to tumor cell proliferation.

(Ren, Ling, et al., 2015)

The line's deepest value, however, lies in its pedigree. Together with the parental TE85/HOS cells and the KRAS-transformed derivative 143B, MNNG/HOS forms a three-tier in vitro–in vivo progression system that models the stages of osteosarcoma evolution. When GFP-tagged cells were injected into the proximal tibia of athymic mice, TE85 cells were neither tumorigenic nor metastatic; MNNG/HOS cells formed primary tumors efficiently but metastasized to the lung at a markedly lower rate than 143B — roughly 50-fold fewer pulmonary lesions; and 143B combined rapid growth with heavy metastatic burden. This graduated behavior makes the MNNG/HOS orthotopic model the middle rung of a uniquely controlled comparative ladder. Tumors are fast — subcutaneous xenografts in NSG mice can approach 1000 mm3 within 10–20 days — and orthotopic primary lesions are readily imaged by whole-body fluorescence or luciferase bioluminescence, while pulmonary metastases are enumerated at necropsy. The luciferase-tagged MNNG/HOS-luc variant extends this utility with quantitative IVIS-class tracking of both sites, making the platform a workhorse for osteosarcoma pharmacology, matrix-remodeling research, and comparative progression studies.

Cell Line Information: MNNG/HOS / MNNG/HOS-luc

MNNG/HOS (ATCC CRL-1547; ECACC 87070201), designated MNNG/HOS Cl #5 [R-1059-D] or MNNG/HOS (TE85, Clone F-5), is deposited with the American Type Culture Collection and the European Collection of Authenticated Cell Cultures. The luciferase-tagged MNNG/HOS-luc derivative, generated by stable reporter transduction, is used for quantitative bioluminescence tracking while retaining parental growth behavior. Our Services

Commissioning Alfa Cytology for the MNNG/HOS / MNNG/HOS-luc orthotopic model consolidates the entire study under one accountable team: authenticated human cell banks are expanded at low passage and luciferase-verified, immunodeficient hosts are prepared and acclimated, the Matrigel-supported inoculum is delivered into the tibial marrow cavity by microsurgeons practiced in the technique, primary tumors are followed by scheduled X-ray and bioluminescence imaging, and your treatment arms run through preset endpoint criteria to terminal bone and lung histopathology under IACUC-compliant conditions — all returned as an audit-ready data package.

Workflow of MNNG/HOS / MNNG/HOS-luc Orthotopic Mouse Model Construction

Orthotopic MNNG/HOS work moves quickly, so discipline is everything: a compact Matrigel-supported inoculum, tight surgical technique, and scheduled dual-modality monitoring. A standard program proceeds as follows.

  1. Cell preparation and quality control: MNNG/HOS or MNNG/HOS-luc cells are expanded at low passage, confirmed mycoplasma-free and STR-authenticated, and luciferase activity validated in vitro; cells are kept in culture only briefly before inoculation to preserve aggressive growth behavior.
  2. Host selection and acclimation. Male or female immunodeficient mice: athymic nude or NSG strains, typically 4–6 weeks of age — are quarantined and acclimated in a specific-pathogen-free barrier facility.
  3. Anesthesia and tibial access: After pre-emptive analgesia such as meloxicam and induction of isoflurane anesthesia, a small incision exposes the proximal tibia, and the knee is flexed to present the tibial plateau.
  4. Intratibial inoculation. A fine needle is guided through the growth plate into the marrow cavity: a subtle change in resistance confirms correct placement — and roughly 5 × 105–106 cells in 10–20 μL PBS, mixed with Matrigel to a total volume near 30 μL, are injected slowly.
  5. Closure and postoperative care: The needle is withdrawn carefully, the incision is closed, and mice recover on warming with daily monitoring of weight and gait over the following three days.
  6. Primary tumor monitoring: Tumors typically become palpable within three to five weeks; caliper measurements every two to three days are paired with scheduled X-ray and, for luciferase-tagged cohorts, bioluminescence imaging after intraperitoneal D-luciferin injection.
  7. Randomization and treatment: Mice are randomized into arms at a defined tumor volume, test articles are dosed according to protocol, and body weight, clinical signs, and imaging signals are captured at preset intervals.
  8. Endpoint analysis and reporting: Mice are euthanized when tumors reach the protocol threshold (for example ~1.5 cm); tumor-bearing limbs and lungs are resected, metastatic nodules are enumerated and processed for H&E and immunohistochemistry, imaging signals are reconciled with histology, and all findings are consolidated into the final report.
