PC-9 Xenograft Model Service for NSCLC

Fig 1: PC-9 xenograft model for NSCLC preclinical research.

The PC-9 xenograft model, harboring the canonical EGFR exon 19 deletion in a PTEN-intact, KRAS-wild-type background, serves as the preclinical gold standard for evaluating first- and third-generation EGFR tyrosine kinase inhibitors in lung adenocarcinoma. Alfa Cytology provides authenticated PC-9 cell line-derived xenograft services with verified EGFR mutational status, enabling robust assessment of EGFR-directed monotherapy and combination strategies in early-phase NSCLC development.

Overview of PC-9 Xenograft Model for NSCLC

PC-9 was established from a human lung adenocarcinoma that retains differentiated glandular architecture. The cell line carries the sensitizing EGFR exon 19 deletion (delE746-A750), which results in constitutive receptor activation and robust downstream signaling through the PI3K/AKT and RAS/RAF/MEK/ERK cascades. Its PTEN-positive, KRAS-wild-type, BRAF-wild-type, and ALK-wild-type genotype isolates EGFR as the dominant oncogenic driver, making PC-9 an ideal system for dissecting EGFR-dependent tumor biology without confounding compensatory pathway activation.

Pharmacodynamic IHC of PC-9 xenograft tumors demonstrated that AZD9291 durably suppressed pEGFR, pERK, pS6 and pPRAS40 across 30 hours, confirming the sustained target engagement underlying deep tumor regressions in the PC-9 model. (Cross, Darren AE, et al., 2014)

As a subcutaneous xenograft, PC-9 exhibits rapid and homogeneous tumor formation with palpable nodules appearing within 7–10 days and volumes reaching 600–900 mm3 by 4–5 weeks post-inoculation. Histologically, the tumors recapitulate moderately to well-differentiated adenocarcinoma with glandular and solid structures, strong membrane-localized EGFR expression, and robust Ki-67 staining. The model demonstrates predictable sensitivity to gefitinib, erlotinib, and afatinib, and has been extensively employed to benchmark osimertinib analogs and to elucidate mechanisms of acquired T790M-mediated resistance.

Cell Line Information: PC-9

The PC-9 cell line was originally deposited with the Riken BioResource Center and is distributed by ECACC (Catalogue No. 90071810). Our Services

Alfa Cytology leverages authenticated PC-9 cultures to construct xenografts with documented EGFR exon 19 deletion fidelity, offering flexible study designs that range from single-agent TKI dose-response characterization to combination regimens targeting compensatory signaling nodes, supported by comprehensive pharmacodynamic and histopathological endpoint analysis.

Workflow of PC-9 Xenograft Model Construction

PC-9 xenograft construction adheres to a protocol tailored to its adherent/suspension growth phenotype and rapid proliferation rate, ensuring consistent engraftment and interpretable pharmacological readouts. The procedure unfolds through six sequential stages:

  1. Cell Expansion and Molecular Verification: PC-9 cells are propagated in RPMI-1640 with 2 mM glutamine and 10% FBS. Each batch is authenticated by STR profiling, tested for mycoplasma, and confirmed for EGFR exon 19 deletion status by targeted sequencing prior to implantation.
  2. Harvest and Matrigel Preparation: Cultures are detached with 0.05% trypsin-EDTA, washed, and counted. Cells are resuspended at 1 × 107 cells/mL in a 1:1 blend of serum-free RPMI-1640 and high-concentration Matrigel to preserve epithelial architecture and promote rapid stromal interaction.
  3. Subcutaneous Implantation: A 100 µL inoculum containing 1 × 106 viable cells is injected subcutaneously into the flank of immunodeficient mice. Tumor nodules generally become palpable within 7–10 days post-injection.
  4. Tumor Monitoring and Group Allocation: Tumor dimensions are measured twice weekly with digital calipers, and volume is calculated as (length × width2) / 2. Once tumors reach 100–150 mm3, mice are randomized into treatment cohorts matched for tumor volume and body weight.
  5. Compound Administration and In-Life Monitoring: Test articles are dosed according to the predefined schedule. Tumor volumes and body weights are recorded at regular intervals. Optional blood sampling permits pharmacokinetic and pharmacodynamic correlation.
  6. Endpoint Analysis and Tissue Processing: At study termination, tumors are excised, weighed, and photographed. Tissues are fixed in 10% neutral buffered formalin for H&E and immunohistochemistry, with aliquots snap-frozen for genomic, transcriptomic, or proteomic analyses.

Case Study-PC-9 Xenograft Model Development

In a recent preclinical engagement, the PC-9 xenograft was utilized to evaluate the antitumor activity of a novel allosteric MEK inhibitor administered in combination with a standard-of-care EGFR-TKI. Following subcutaneous engraftment in athymic nude mice, animals bearing established tumors were randomized and treated for 21 days. The combination regimen produced marked tumor growth inhibition relative to vehicle and single-agent controls, with responder tumors showing reduced phospho-ERK and phospho-AKT staining upon post-study immunohistochemical analysis. Notably, E-cadherin expression remained robust in treated tumors, indicating preservation of epithelial differentiation. These preclinical findings provided mechanistic rationale for downstream combination development and biomarker validation efforts.
