CE81T/VGH Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: CE81T/VGH Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The CE81T/VGH orthotopic mouse model recreates human esophageal squamous cell carcinoma within the native esophageal microenvironment, enabling preclinical evaluation of tumor growth, local invasion, and therapeutic response under conditions that closely resemble the patient disease. Backed by an experienced tumor modeling team and a well-equipped preclinical platform, Alfa Cytology delivers end-to-end CE81T/VGH orthotopic model construction, longitudinal bioluminescence monitoring, histopathological characterization, and efficacy assessment, all tailored to the specific goals of each research program.

Overview of CE81T/VGH Orthotopic Mouse Model for Esophageal Cancer

CE81T/VGH is a well-characterized human esophageal squamous cell carcinoma (ESCC) cell line originally established from a 57-year-old male patient of Chinese (Taiwanese) origin at Taipei Veterans General Hospital — the source of the "VGH" designation in its name. As an epithelial cell line derived directly from esophageal tumor tissue, it retains key malignant features of ESCC, including epithelial morphology, a relatively slow doubling time of approximately 72 hours, and documented tumorigenicity in immunodeficient mice. The line has been extensively used in oncology research, ranging from the selection of highly invasive subpopulations (CE81T-0 to CE81T-4) and studies on matrix metalloproteinase regulation, betel quid/arecoline exposure, and mTOR signaling, to apoptosis and cell cycle analyses involving p53, Bax, Bcl-2, and caspase pathways in vitro, with findings validated in xenograft studies in vivo.

While conventional subcutaneous xenografts remain widely used, they fail to reproduce the anatomical and stromal context of the primary tumor. The orthotopic approach addresses this limitation by introducing CE81T/VGH cells directly into the esophageal wall of immunodeficient mice, typically in the abdominal esophagus near the diaphragm. Tumors developing at this site invade the muscularis propria, progressively compress the esophageal lumen to produce stricture-like symptoms, and are associated with body weight loss — features that mirror the clinical course of esophageal cancer far better than flank tumors. When the cells are labeled with a luciferase reporter, orthotopic tumor burden can be tracked non-invasively and quantitatively by bioluminescence imaging, making the CE81T/VGH orthotopic model a powerful platform for preclinical drug screening, radiotherapy and chemotherapy response studies, and mechanistic investigations of ESCC progression.

Workflow of CE81T/VGH Orthotopic Mouse Model Construction

Construction of the CE81T/VGH orthotopic model combines reporter-labeled tumor cell preparation with refined microsurgical techniques and longitudinal imaging. The standard workflow is outlined below.

  1. Cell line authentication and reporter labeling: CE81T/VGH cells are verified by STR profiling and tested for mycoplasma before use, then stably transduced with a luciferase reporter to enable non-invasive tumor tracking.
  2. Pre-inoculation cell preparation: Cells are expanded under standardized culture conditions, harvested during logarithmic growth, counted and assessed for viability, and resuspended at a defined concentration in an ice-cold medium/Matrigel mixture (typically on the order of 2 × 105 cells in 10 μL for orthotopic inoculation).
  3. Anesthesia and surgical exposure: Immunodeficient mice are anesthetized and provided analgesia; a small upper-abdominal incision is made, the liver is gently elevated, and traction on the stomach exposes the abdominal esophagus near the diaphragm.
  4. Orthotopic inoculation: Using a fine 30-gauge needle, the cell suspension is injected into the esophageal wall (muscularis externa), with the local bulging at the injection site confirming intramural delivery; Matrigel solidifies rapidly at body temperature to minimize cell leakage.
  5. Closure and perioperative care: The abdominal wall and skin are sutured, and mice receive supportive care with close monitoring of body weight, hydration, and general status during recovery.
  6. Longitudinal tumor monitoring: Orthotopic tumor growth is followed by weekly bioluminescence imaging, together with body weight and survival records, allowing quantitative tracking of tumor burden in vivo.
  7. Endpoint analysis and reporting: At study end, esophageal tumors and major organs are collected for size and weight measurement, H&E and immunohistochemical staining, and metastasis assessment, with all data compiled into a comprehensive study report.
