NCI-H1581 Xenograft Model Service for NSCLC

Fig 1: NCI-H1581 xenograft model for NSCLC preclinical research.

The NCI-H1581 xenograft model serves as a preclinical benchmark for FGFR1-driven non-small cell lung cancer, harboring focal amplification of the FGFR1 locus alongside wild-type KRAS and BRAF status. Alfa Cytology delivers comprehensive NCI-H1581 xenograft services—from model establishment and longitudinal tumor tracking to pharmacodynamic biomarker analysis—empowering researchers to evaluate pan-FGFR inhibitors, irreversible tyrosine kinase blockers, and rational combination regimens in a genetically defined, oncogene-addicted tumor setting.

Overview of NCI-H1581 Xenograft Model for NSCLC

The NCI-H1581 cell line was established in January 1987 from the pleural effusion of a 44-year-old Caucasian male smoker with a 75 pack-year history and Stage IV non-small cell lung cancer. It exhibits a distinctive mixed growth pattern—forming floating aggregates of round epithelial-like cells alongside a minor adherent population—reflecting its large-cell carcinoma phenotype. Genomically, NCI-H1581 is defined by focal amplification of chromosome 8p11-12 encompassing FGFR1, which drives constitutive receptor tyrosine kinase signaling through autocrine and paracrine loops. This amplification is accompanied by elevated phosphorylation of the downstream adaptor FRS2, confirming active pathway engagement. Importantly, the line retains wild-type KRAS, BRAF, and LKB1 loci, offering an uncluttered genetic landscape in which FGFR1 dependency can be interrogated without confounding oncogenic signals.

Oral CPL304110 twice daily produced dose-dependent tumor growth inhibition in NCI-H1581 xenografts (TGI 38%, 66% and 85% at 10, 20 and 60 mg/kg), confirming robust FGFR1-pathway blockade of FGFR1-amplified lung cancer in vivo. (Popiel, Delfina, et al., 2024)

Functionally, NCI-H1581 demonstrates exquisite sensitivity to FGFR tyrosine kinase inhibition. Pharmacological blockade with agents such as PD173074, FIIN-1, CPL304110, and rogaratinib produces potent antiproliferative and anchorage-independent growth suppression, with shRNA-mediated FGFR1 knockdown resulting in marked viability loss that can be rescued by ectopic wild-type FGFR1 expression—formally establishing FGFR1 as the critical oncogenic driver. The line has also served as a prototype for studying acquired resistance, with p38 MAPK pathway upregulation identified as a compensatory escape mechanism following chronic FGFR inhibitor exposure. Collectively, these attributes render the NCI-H1581 xenograft an indispensable platform for validating FGFR-directed therapeutics, profiling on-target pharmacodynamics, and exploring resistance-prevention strategies in molecularly selected NSCLC.

Workflow of NCI-H1581 Xenograft Model Construction

Development of a reproducible NCI-H1581 xenograft model requires careful adaptation to its unique mixed growth phenotype and stringent quality controls to preserve FGFR1-driven oncogenic signaling throughout the study. At Alfa Cytology, all procedures are conducted under IACUC-approved protocols with documented SOPs. The workflow proceeds as follows:

  1. Cell Line Expansion and Pre-Implantation Characterization: NCI-H1581 cells are cultured in DMEM:F12 (1:1) with 10% FBS, with particular attention to maintaining the floating aggregate phenotype. Each batch is subjected to mycoplasma PCR, STR authentication, and FGFR1 expression verification by immunoblot prior to harvest. Only cultures with confirmed FGFR1 amplification and >95% viability are cleared for in vivo use.
  2. Recipient Animal Selection and Acclimation: Immunodeficient mice (BALB/c nude or NOD/SCID) undergo health screening and a minimum five-day acclimation period. Baseline body weights and clinical observations are documented to ensure cohort uniformity prior to tumor cell injection.
  3. Subcutaneous Tumor Cell Implantation: A single-cell suspension (1 × 106 to 5 × 106 cells in 100–200 µL of serum-free medium, optionally mixed 1:1 with Matrigel™) is injected subcutaneously into the flank under sterile conditions. Given the line's mixed growth properties, gentle dissociation protocols are employed to preserve cell viability. Tumors typically become palpable within 10–14 days.
  4. Tumor Monitoring and Cohort Randomization: Tumor dimensions are measured by digital caliper every 2–3 days, with volumes calculated using the modified ellipsoid formula (length × width2 / 2). Upon reaching the target volume (typically 100–150 mm3), animals are randomized into vehicle-control and treatment groups using stratified randomization based on tumor size and body weight.
  5. Therapeutic Administration and Clinical Monitoring: Test compounds are dosed according to sponsor-defined protocols (route, frequency, and duration). Mice are evaluated daily for body weight, clinical signs, and treatment-related adverse events, with strict enforcement of predefined humane endpoints.
  6. Terminal Analysis and Data Integration: At study termination, tumors are excised, weighed, photographed, and allocated for multi-modal analysis: histopathology (H&E, Ki-67, TUNEL), phospho-protein profiling (p-FRS2, p-FGFR1, p-ERK by IHC or Western blot), gene-expression analysis, and pharmacokinetic correlation. All data are compiled into a comprehensive report with statistical evaluation.

Case Study-NCI-H1581 Xenograft Model Development

In a preclinical program focused on FGFR1-amplified NSCLC, Alfa Cytology established subcutaneous NCI-H1581 xenografts to evaluate the antitumor efficacy of a novel irreversible pan-FGFR inhibitor, both as monotherapy and in combination with a p38 MAPK inhibitor designed to preempt compensatory resistance. Following tumor engraftment and randomization, animals received daily oral dosing over a four-week treatment window. Longitudinal caliper measurements demonstrated robust tumor growth inhibition in the monotherapy arm, with near-complete stasis observed in the combination cohort. Terminal pharmacodynamic analysis revealed profound suppression of intratumoral p-FRS2 and p-ERK in treated tumors, while p38 phosphorylation remained attenuated in the combination group relative to monotherapy-treated animals. Body-weight trajectories were stable across all cohorts, and no unexpected histopathological findings were observed in major organs. These findings highlight the translational utility of the NCI-H1581 platform for both validating FGFR-directed monotherapy and rationally designing combination strategies to forestall resistance in molecularly defined NSCLC.
