Fig 1: TRAMP-C1 / TRAMP-C1-luc Orthotopic Mouse Model for Prostate Cancer preclinical research.

The TRAMP-C1 / TRAMP-C1-luc orthotopic mouse model is a fully immunocompetent, syngeneic platform that recapitulates prostate tumor growth in its native anatomical niche, enabling realistic evaluation of novel therapeutics against prostate cancer. Alfa Cytology delivers this model as a complete, ready-to-use preclinical service — from cell line authentication and surgical orthotopic implantation to longitudinal bioluminescence tracking, treatment administration, and full histopathological endpoints — so your team can focus on the science while we handle the animal work with rigor and reproducibility.

Overview of TRAMP-C1 / TRAMP-C1-luc Orthotopic Mouse Model for Prostate Cancer

TRAMP-C1 is a murine prostate adenocarcinoma cell line established in 1996 from a heterogeneous 32-week primary tumor arising in the TRAMP (transgenic adenocarcinoma of the mouse prostate) model. The parental TRAMP strain carries a construct in which the minimal rat probasin promoter drives prostate-specific epithelial expression of the SV40 large T antigen, so TRAMP males spontaneously progress from prostatic intraepithelial neoplasia to adenocarcinoma with distant metastases. The TRAMP-C1 line retains an epithelial phenotype, expresses cytokeratin, E-cadherin, and androgen receptor, and — critically — forms tumors reliably when grafted into syngeneic C57BL/6 hosts, making it one of the most widely used immunocompetent prostate cancer models for preclinical research. Because host and tumor are genetically matched, the model preserves an intact immune system, allowing investigators to study immunotherapies, checkpoint inhibitors, vaccines, and combination regimens in a setting that closely mirrors the human disease milieu.

When TRAMP-C1 cells are implanted orthotopically — directly into the prostate gland rather than a subcutaneous site — the resulting tumors develop within the authentic prostatic microenvironment, with proper stromal interactions, tissue-specific vasculature, and drainage to para-aortic lymph nodes. This configuration supports spontaneous local invasion and metastatic spread and yields drug exposure kinetics that more faithfully reflect clinical pharmacology than flank xenografts. The luciferase-engineered derivative, TRAMP-C1-luc, adds a firefly luciferase reporter that permits noninvasive, serial bioluminescence imaging of tumor onset, growth kinetics, response to treatment, and relapse in the same animal over time, substantially reducing cohort sizes and inter-animal variability. Together, the TRAMP-C1 and TRAMP-C1-luc orthotopic models provide a versatile, translationally relevant system for interrogating prostate cancer biology and benchmarking therapeutic candidates before further development.

Cell Line Information: TRAMP-C1 / TRAMP-C1-luc

TRAMP-C1 was derived in 1996 by Foster and colleagues from a 32-week primary prostate tumor of a PB-Tag C57BL/6 (TRAMP) transgenic mouse, and is distributed by the American Type Culture Collection (ATCC CRL-2730). The TRAMP-C1-luc derivative is generated by stable transduction with a firefly luciferase reporter construct, enabling bioluminescence-based tracking without altering tumorigenicity in syngeneic hosts. Key characteristics are summarized below.

Workflow of TRAMP-C1 / TRAMP-C1-luc Orthotopic Mouse Model Construction

Construction of the orthotopic model combines careful cell quality control, microsurgical implantation into the prostate gland, and longitudinal noninvasive monitoring. The workflow below summarizes how a typical study proceeds from cell preparation to endpoint analysis.

  1. Cell preparation and quality control: TRAMP-C1 or TRAMP-C1-luc cells are expanded under standardized conditions, confirmed mycoplasma-free, authenticated, and verified for luciferase activity in vitro before inoculation.
  2. Host selection and acclimation: Male C57BL/6 mice, typically 6–10 weeks of age, are housed in a specific-pathogen-free barrier facility and allowed to acclimate for at least one week prior to surgery.
  3. Surgical exposure of the prostate: Under general anesthesia, a lower midline abdominal incision is made, and the anterior prostate lobe is gently externalized by retraction of the seminal vesicle, exposing the injection site while minimizing tissue trauma.
  4. Intra-prostatic inoculation: A single-cell suspension (commonly around 1 × 106 cells in 25–50 μL PBS, optionally mixed 1:1 with Matrigel) is injected into the prostate lobe using a fine-gauge Hamilton syringe; uniform blanching or engorgement of the lobe confirms successful delivery.
  5. Closure and perioperative recovery: The prostate is returned to the abdominal cavity, the incision is closed in layers with absorbable suture, and mice recover on a heated pad with analgesia and close postoperative observation.
  6. Tumor monitoring and randomization: For TRAMP-C1-luc studies, bioluminescence imaging is performed after intraperitoneal D-luciferin injection on a weekly schedule to confirm engraftment and track tumor burden; ultrasound or caliper measurements complement imaging, and mice are randomized into treatment groups based on signal intensity.
  7. Treatment and longitudinal assessment: Test articles, antibodies, radiation, or combination regimens are administered according to the study design, with body weight, health status, and tumor signal recorded at defined intervals.
  8. Endpoint necropsy and analysis: At study termination, tumors and prostate-draining lymph nodes are excised and weighed, tumor dimensions are recorded, and tissues are processed for H&E and immunohistochemical staining, immune profiling by flow cytometry, and metastasis assessment, with all data compiled into a final report.
