K562 / K562-luc Orthotopic Mouse Model Service for Leukemia

Fig 1: K562 / K562-luc Orthotopic Mouse Model for Leukemia preclinical research.

The K562 / K562-luc orthotopic mouse model for leukemia engrafts a Philadelphia chromosome-positive human chronic myeloid leukemia line into the blood and bone marrow compartments of immunodeficient mice, where disseminated disease can be quantified non-invasively over time. Exclusively a preclinical CRO, Alfa Cytology delivers this model as a complete service, from authenticated K562 / K562-luc cell supply through intravenous or intraosseous inoculation, scheduled in vivo bioluminescence imaging, and terminal organ-level analysis.

Overview of K562 / K562-luc Orthotopic Mouse Model for Leukemia

K562 occupies a unique place in cancer research: it was the first human leukemia cell line ever established, derived in 1970 by Lozzio and Lozzio from the pleural effusion of a 53-year-old female patient with chronic myeloid leukemia (CML) in terminal blast crisis. The line carries the defining lesion of CML, the Philadelphia chromosome translocation t(9;22) that generates the BCR-ABL1 e14a2 (p210) fusion tyrosine kinase, and exhibits an undifferentiated, erythroleukemia-like phenotype in suspension culture. K562 cells are characteristically deficient in surface MHC class I molecules, which makes them exquisitely sensitive to natural killer (NK) cell lysis and has turned them into the standard target cell for NK activity assays worldwide. The line is remarkably prolific in the literature, with well over ten thousand published studies, and responds to BCR-ABL1 tyrosine kinase inhibitors such as imatinib, dasatinib, and nilotinib, anchoring its role in CML drug research.

(Anna, François, et al., 2021)

For orthotopic modeling, K562 is introduced into the natural disease compartments of the host. Luciferase-labeled derivatives (K562-luc) are delivered intravenously into sublethally irradiated NSG (NOD-scid IL2rγnull) mice, typically at inocula of 2–5 × 106 cells, or injected directly into the femoral cavity as a bone marrow-residing model at around 105 cells. Engrafted cells seed spleen, lymph nodes, bone marrow, and liver, with imaging studies showing that K562 produces its strongest bioluminescent signals in lymph nodes, a distribution pattern distinct from other leukemia lines. Whole-animal bioluminescence after D-luciferin administration then reports systemic disease burden longitudinally, enabling therapy response to be quantified days before clinical decline and supporting survival studies that typically run several weeks. The model is the platform of choice for BCR-ABL1-targeted agent evaluation, TKI resistance studies, and adoptive cell therapy programs involving NK or CAR-engineered effectors. Figure 1 depicts the overall architecture of this orthotopic system.

Cell Line Information: K562 / K562-luc

As the prototypical BCR-ABL1-positive CML line, K562 has been characterized exhaustively. The defining attributes of the parent line and its luciferase-tagged derivative are summarized below.

Cell Line Name K562; luciferase-labeled derivative: K562-luc (also used as K562-LucGFP dual-tagged reporter)
Catalog / Accession ATCC CCL-243
Species / Tissue / Disease Human (Homo sapiens) / pleural effusion / chronic myeloid leukemia in terminal blast crisis (erythroleukemia phenotype)
Origin Established in 1970 by Lozzio and Lozzio from pleural effusion of a 53-year-old female patient; first reported in 1975; the first CML-derived leukemia cell line
Morphology / Growth Highly undifferentiated, round cells growing in suspension; no attachment required
Culture Conditions RPMI-1640 with 10% fetal bovine serum and 2 mM L-glutamine (37 °C, 5% CO₂); routinely maintained at ~0.5 × 106 cells/mL with regular media replenishment
Karyotype Genomically unstable; reported as hypodiploid in short-term culture and near-triploid in long-term culture; Philadelphia chromosome positive
Defining Molecular Lesion BCR-ABL1 fusion, e14a2 (b3a2) transcript encoding p210 BCR-ABL1 constitutively active tyrosine kinase
Drug Responsiveness Sensitive to BCR-ABL1 tyrosine kinase inhibitors (imatinib, dasatinib, nilotinib); TKI-resistant sublines and colony-forming variants are widely used
Immunological Notes Lacks surface MHC class I expression, rendering the cells highly susceptible to NK-mediated cytotoxicity; the standard target cell for NK killing assays and antibody-dependent cytotoxicity studies
K562-luc Derivative Stably expresses firefly luciferase (often with GFP), permitting quantitative in vivo bioluminescence imaging; organ distribution is line-specific, with the strongest K562 signals localizing to lymph nodes
Engraftment Parameters (Published) Intravenous inoculation of 2.5–5 × 106 cells into sublethally irradiated NSG mice; intra-femoral bone marrow-residing models use ~105 cells; BLI performed after intraperitoneal D-luciferin (150 mg/kg)
Research Applications BCR-ABL1-targeted drug and TKI resistance evaluation, CML blast crisis biology, adoptive NK / CAR / γδ T-cell therapy testing, graft-versus-leukemia research, hematopoietic differentiation and gene regulation studies

Our Services

Every K562 / K562-luc program run by Alfa Cytology is anchored by verified cell banks, conditioning regimens suited to the chosen host, calibrated imaging schedules, and a terminal package combining organ-level bioluminescence, flow cytometry, and survival statistics.

Workflow of K562 / K562-luc Orthotopic Mouse Model Construction

Standing up a K562 / K562-luc orthotopic leukemia study follows a clear sequence in which cell authentication, host conditioning, route-specific inoculation, and serial imaging each play a defined role. The sequence below reflects the standard execution of this platform.

  1. Cell Bank Verification and Expansion: K562 or K562-luc stocks are authenticated by STR profiling and mycoplasma testing, then expanded in suspension culture with regular feeding to preserve logarithmic growth.
  2. Luciferase Calibration (K562-luc Only): In vitro signal output per cell is verified, confirming that in vivo photon flux can be interpreted as relative disease burden.
  3. Host Conditioning and Randomization: NSG or related immunodeficient mice are randomized by body weight and, where the protocol requires, conditioned with sublethal irradiation to facilitate human cell engraftment under approved animal protocols.
  4. Orthotopic Route-Specific Inoculation: A single-cell suspension is delivered by tail-vein injection (commonly 2–5 × 106 cells) to model systemic leukemia, or by intra-femoral injection (~105 cells) to establish a bone marrow-residing tumor that demands effector homing.
  5. Engraftment Confirmation: Whole-animal bioluminescence is captured a few days after inoculation, typically after intraperitoneal D-luciferin (150 mg/kg), to verify cell take and establish baseline disease loads.
  6. Scheduled Longitudinal Imaging: Mice are imaged on a defined schedule to chart disease progression in vivo, with body weight and clinical condition recorded alongside.
  7. Treatment Administration: Test agents or adoptive cell products are dosed per the client’s regimen, with response read out through the next imaging cycles.
  8. Endpoint Analysis and Survival Statistics: At humane endpoint, bone marrow, spleen, lymph nodes, and blood are collected for ex vivo imaging, human CD45+ engraftment quantification by flow cytometry, and histopathology; survival data are compiled for Kaplan–Meier analysis.
