EMT6 Syngeneic Model Service for Breast Cancer

Fig 1: EMT6 Syngeneic Model for Breast Cancer preclinical research.

The EMT6 syngeneic model is the radiobiology-born workhorse of immunocompetent breast cancer research, engrafting a triple-negative murine mammary adenocarcinoma into BALB/c hosts to deliver rapid, reproducible tumors validated across checkpoint blockade, radiation combinations, and hypoxia-targeted therapy. Alfa Cytology delivers this benchmark model as a complete preclinical service — authenticated cell banking from the 1972-characterized reference line, subcutaneous or orthotopic mammary-fat-pad implantation, scheduled bioluminescence and caliper monitoring, and full histopathology — giving sponsors the triple-negative syngeneic system whose immunology helped define modern combination design.

Overview of EMT6 Syngeneic Model for Breast Cancer

EMT6 — Experimental Mammary Tumour-6 — entered the scientific record through one of the most careful characterizations of its era: Sara Rockwell, Robert Kallman, and Luis Fajardo described the line in the Journal of the National Cancer Institute in 1972, having selected it in tissue culture from the 25th transplant generation of the KHJJ tumor, itself born from a hyperplastic alveolar nodule in a BALB/c mouse. Its very name encodes its provenance, and its subsequent career split into two intertwined identities. The first is radiobiological: because the founder's laboratory specialized in tumor hypoxia, EMT6 became a founding model for hypoxic-cell radiosensitizer development, fractionation studies, and photodynamic-therapy agent testing, its solid tumors developing the hypoxic regions that make radioresistance questions experimentally tractable. The second is immunological: the line is a hormone receptor-negative, triple-negative mammary adenocarcinoma that is highly immunogenic with brisk kinetics — 106 subcutaneous cells producing consistent tumors randomizable within a week — making it one of the most responsive BALB/c platforms for checkpoint and combination immunotherapy.

(Khan, Kabir A., et al., 2020)

The model's defining scientific moment arrived in 2018, when Mariathasan and colleagues used EMT6 to show that intratumoral TGF-β signaling excludes T cells from tumor parenchyma and thereby attenuates response to PD-L1 blockade — and that combining TGF-β inhibition with anti-PD-L1 converted immune-excluded tumors into inflamed, regressing lesions. That single finding, replicated across the field, reshaped combination immunotherapy strategy and cemented EMT6's place in the canon. Around that landmark the platform offers exceptional tooling: the EMT6.8 cloned line for antigen studies; EMT6-LM2, a lung-metastasis-selected derivative for experimental metastasis work; OVA- and PSA-secreting transfectants that elicit adjuvant-level antibody responses in syngeneic hosts; HER-2/neu-transfected variants for receptor-targeting questions; and eGFP/Fluc reporter lines for quantitative bioluminescence. Orthotopic implantation into the mammary fat pad delivers the anatomically faithful setting, with late pulmonary metastasis for dissemination studies. For sponsors, EMT6 offers a rare pedigree: radiobiology heritage, triple-negative biology, and the immunology that taught the field how to break immune exclusion.

Cell Line Information: EMT6 Syngeneic Model

EMT6 (ATCC CRL-2755; Cellosaurus CVCL_1923) is the tissue-culture-adapted derivative of the KHJJ transplantable mammary tumor, deposited by S. Rockwell in 1971 and characterized in the landmark 1972 JNCI paper. Engineered derivatives extend the platform from lung-metastasis selection to antigen-defined immunology. Core characteristics are compiled below.

Orthotopic tumorigenicity 106 cells into the lower-right mammary fat pad: reproducible orthotopic tumors (~100 mm3 by day 7) with caliper monitoring and late pulmonary metastasis; less aggressive metastatically than 4T1 but adopts pulmonary spread readily
Metastatic derivatives EMT6-LM2: lung-metastasis-selected derivative for experimental metastasis studies; i.v. lung-colony formation quantifiable after scatter-factor (HGF/SF) stimulation, which enhances motility, invasiveness, and urokinase expression ~1.7-fold in lung colonization

Our Services

Engaging Alfa Cytology for the EMT6 syngeneic model places a half-century of radiobiological and immunological precedent under one accountable team: authenticated, low-passage EMT6 banks are maintained with mycoplasma-free certification, syngeneic BALB/c hosts are sourced and acclimated, tumors are established subcutaneously or in the mammary fat pad at the published 106-cell geometry, and treatment arms — from checkpoint and TGF-β-pathway combinations to radiation-immunotherapy regimens and hypoxia-targeted agents — run through scheduled caliper and bioluminescence monitoring to terminal mammary and pulmonary histopathology, all under IACUC-compliant conditions and returned as an audit-ready data package.

Workflow of EMT6 Syngeneic Model Construction

EMT6 studies divide into rapid flank screening and anatomically faithful mammary-fat-pad modeling, both resting on the line's brisk kinetics and published checkpoint benchmark geometry. A standard program proceeds as follows.

  1. Cell preparation and quality control: EMT6 cells (or engineered derivatives) are expanded at low passage, confirmed mycoplasma-free and STR-authenticated, and luciferase activity validated in vitro where reporter formats are used; single-cell suspensions are prepared fresh at 106 cells per inoculum.
  2. Host selection and acclimation: Female BALB/c mice, typically 6–8 weeks of age, are quarantined and acclimated in a specific-pathogen-free barrier facility.
  3. Implantation: subcutaneous route. 106 cells in 0.1 mL sterile PBS are injected into the rear flank; the site is compressed briefly on needle withdrawal, and consistent tumors reach randomization volume (~150 mm3) within about a week.
  4. Implantation: orthotopic mammary-fat-pad route. The lower-right mammary fat pad is exposed through a small incision and 106 cells in a defined volume are delivered into the gland; the incision is closed and tumors reach ~100 mm3 within a comparable window with caliper monitoring.
  5. Randomization and treatment. Mice are randomized by tumor volume into arms, and test articles: anti-PD-1 or TGF-β-pathway combinations, paclitaxel benchmarks, radiation-immunotherapy regimens, or hypoxia-targeted agents — are dosed per protocol with body weight recorded.
  6. Longitudinal monitoring: Tumors are measured by caliper twice weekly; luciferase-tagged cohorts undergo bioluminescence imaging after intraperitoneal D-luciferin injection, and late-stage cohorts are screened for pulmonary dissemination.
  7. Metastatic-arm execution where specified: For dissemination studies, EMT6-LM2 cells are administered by the designated route with lung-colony enumeration at termination, including scatter-factor-stimulated arms where motility enhancement is under study.
  8. Endpoint analysis and reporting: At termination, primary tumors and lungs are resected and weighed; tissues undergo H&E with mammary histology, immunohistochemistry (CD8, FoxP3, F4/80, CD31, phospho-SMAD2/3, PD-L1 panels), hypoxia-marker staining (pimonidazole or HIF-1α) where radiation arms are included, and metastatic nodule enumeration, with all data consolidated into the final report.
