L1210 Syngeneic Model Service for Leukemia

Fig 1: L1210 Syngeneic Model for Leukemia preclinical research.

The L1210 syngeneic model service for leukemia is an immune-competent DBA/2 platform built on the most historically consequential mouse leukemia in existence — the methylcholanthrene-induced L1210 lymphocytic leukemia that anchored the NCI drug screen for decades — offering ultra-rapid disease kinetics and the classic %ILS survival endpoint. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated L1210 expansion and banking, intraperitoneal, subcutaneous, or intravenous inoculation into DBA/2 or CD2F1 hosts, daily survival monitoring with strict quality-control ranges, and full leukemic histopathology — all configured around each sponsor’s agent, schedule, and survival or efficacy endpoint.

Overview of L1210 Syngeneic Model for Leukemia

L1210’s history is inseparable from the history of chemotherapy itself. Established in 1949 from a methylcholanthrene-induced tumor arising in an eight-month-old female DBA mouse (subline 212), maintained by serial heterotransplantation in DBA/2 mice, and brought into suspension culture in the 1960s, the line became the backbone of the National Cancer Institute’s in vivo drug screen — so thoroughly that an entire era of anticancer pharmacology is expressed in its native metric, the percentage increase in lifespan (%ILS), comparing treated and control survival times. The biology is brutally efficient: lymphoblast-doubling times of 8–10 hours in vitro become median survivals of roughly eight days after intraperitoneal challenge of 1 × 105 cells in syngeneic DBA/2 or semi-syngeneic CD2F1 (BALB/c × DBA/2 F1) hosts, and so extreme is its aggressiveness that in fully histocompatible CD2F1 mice a challenge of as few as one cell can prove lethal. Untreated animals die with leukemic ascites, splenomegaly, and hepatomegaly — a reproducible, well-documented terminal picture that made L1210 the ideal first-pass filter for cytotoxic discovery.

In vivo antileukemic activity and immunogenicity of NRRP-treated L1210 cells in the syngeneic DBA/2 model — survival curves of mice challenged with L1210 cells or NRRP-pretreated L1210 cells, serum cytokine profiling (m-CSF, LIF, MIP-2, IL-4, IL-2, RANTES, CXCL11, MCP-1, G-CSF) 20 h after treatment, and protection against rechallenge showing long-lasting antitumor immunity. (Batenchuk, C., et al., 2013)

Two legacies make the model uniquely valuable today. The first is its family of drug-resistant derivatives — among the most studied resistance reagents in cancer biology: the cisplatin-selected L1210/DDP series (23- to 108-fold resistance), the DACH-platinum-selective L1210/DACH line, methotrexate-resistant variants driven by dihydrofolate reductase amplification, and the ribonucleotide-reductase-inhibitor-resistant ED1/ED2 lines — enabling matched sensitive-versus-resistant in vivo comparisons. The second is its role in early immunotherapy lessons: serotherapy studies with the L.1 monoclonal antibody in CD2F1 hosts, published in 1983, delivered sobering insights about complement inactivation and antigen heterogeneity that still inform antibody design. Contemporary providers have re-validated the model with modern agents — cisplatin at 3 mg/kg intraperitoneally, dosed weekly, significantly extended survival in the validated DBA/2 configuration — and position it for CAR-T and other immunotherapy evaluation. L1210 remains the fastest, cheapest route to a statistically powered survival readout for any antileukemic mechanism.

Cell Line Information: L1210

The table below consolidates the key biological, cultural, and modeling characteristics of the L1210 line and its classic resistant derivatives.

Tumor type Murine lymphocytic leukemia (lymphoblast morphology)
Growth properties Suspension-adapted lymphoblasts of extraordinary proliferative capacity; population doubling time approximately 8–10 hours — among the fastest of any murine leukemia line
Recommended culture conditions Classically maintained in Dulbecco’s modified Eagle medium with 10% horse serum, or RPMI-1640 with 10% fetal bovine serum, at 37 °C in 5% CO₂; routine MAP pathogen panel negative (Sendai, polyoma, LCM, MHV, LDV, Mycoplasma pulmonis, and others)
Standard inoculum and routes Intraperitoneal challenge of 1 × 105 cells (NCI acceptable control survival range 8–11 days); contemporary validation used 1 × 106 cells intraperitoneally with treatment from day 2; subcutaneous challenge supported by the NCI in vivo screen; intravenous and intracerebral routes historically used for disseminated and CNS-directed therapy studies
Disease kinetics and endpoints Fulminant ascites leukemia with median survival ≈8 days at the NCI challenge dose; results expressed as T/C ratios and %ILS (percentage increase in lifespan); in fully histocompatible CD2F1 hosts, even a single intraperitoneal cell can be lethal; terminal necropsy picture of ascites with splenomegaly and hepatomegaly
Documented in vivo responses Cisplatin 3 mg/kg intraperitoneally weekly for three weeks significantly extended survival versus vehicle (validated DBA/2 configuration); cyclophosphamide pretreatment and combination immunochemotherapy studies extensively benchmarked; responsive to the classical antimetabolite and alkylator classes on which the NCI screen was built
Resistance biology L1210/DDP lines display 23- to 108-fold cisplatin resistance with defined DACH-platinum selectivity patterns; methotrexate resistance maps to DHFR amplification and marker-chromosome changes; ED1/ED2 lines overexpress ribonucleotide reductase with altered effector-binding subunit regulation — enabling matched resistant-arm designs

Our Services

Few models compress an entire efficacy experiment into ten days the way L1210 does, and Alfa Cytology runs the line with the historical discipline it deserves: authenticated banks of the parental and classic resistant derivatives, validated DBA/2 and CD2F1 inoculation with strict control-survival quality ranges, daily monitoring with %ILS reporting, and terminal histopathology — all under one accountable preclinical operation.

Workflow of L1210 Syngeneic Model Construction

Construction follows the NCI-validated methodology: L1210 cells are expanded under authenticated conditions, formulated at the study-specified intraperitoneal dose, inoculated into DBA/2 or CD2F1 hosts, randomized, treated, and followed daily for survival with %ILS analysis and terminal pathology. The workflow comprises the following stages.

  1. Cell authentication and banking: L1210 or matched resistant derivatives are identity-verified, confirmed mycoplasma-negative with the full MAP pathogen panel documented, and expanded into low-passage working banks under standardized suspension culture conditions.
  2. Cell harvest and formulation: Log-phase cultures are counted and viability-checked, then resuspended at the study-specified dose of approximately 1 × 105 cells (NCI range) to 1 × 106 cells (contemporary validation dosing) in 100 – 200 μL of balanced salt solution per animal.
  3. Anesthesia and inoculation: DBA/2 or CD2F1 mice receive the intraperitoneal challenge (subcutaneous, intravenous, or intracerebral variants follow published alternatives), with dose verification recorded as a primary quality indicator.
  4. Randomization and quality-control check: Animals are randomized by body weight; control-arm survival is tracked against the validated 8–11 day median range, and off-range runs trigger predefined corrective review.
  5. Treatment administration: Experimental arms — classical agents, novel cytotoxics, platinum/antifolate classes against matched resistant derivatives, CAR-T or antibody constructs, and combinations — are dosed per study design with daily body-weight and clinical monitoring.
  6. Daily survival monitoring: Mortality is recorded daily; survival curves are plotted from inoculation, and interim censored animals are documented per protocol.
  7. Endpoint and histopathological analysis: Endpoints are expressed as median survival, %ILS, and statistical significance with terminal necropsy confirming ascites, splenomegaly, and hepatomegaly, plus histopathology (H&E confirming lymphoblast morphology with Ki67 and marker-specific immunohistochemistry) and peripheral-blood or ascites cytology where required.
