EC109 Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: EC109 Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The EC109 orthotopic mouse model for esophageal cancer recreates human esophageal squamous cell carcinoma within its native anatomical site, delivering a biologically faithful setting in which tumor behavior and therapy response can be evaluated preclinically. Alfa Cytology offers comprehensive development of this model, covering authenticated cell expansion, minimally invasive surgical implantation, longitudinal tumor monitoring, and full histopathological endpoint analysis, giving your oncology program a translationally relevant platform built on rigorous quality standards.

Overview of EC109 Orthotopic Mouse Model for Esophageal Cancer

The EC109 cell line is a well-characterized human esophageal squamous cell carcinoma (ESCC) line originally derived from a middle-esophageal tumor of a female patient of Chinese ethnicity. When established as an orthotopic xenograft, EC109 cells are implanted directly into the esophageal wall of immunodeficient mice rather than into the subcutaneous flank, allowing the tumor to develop in the anatomical and physiological context of the native esophagus. This setting preserves critical tumor–stroma interactions, local vascular supply, and organ-specific mechanical cues, so the resulting lesions more closely mirror the growth pattern, histology, and microenvironmental dependence of human esophageal tumors.

Compared with conventional subcutaneous xenografts, the orthotopic EC109 model more faithfully reproduces features that matter in preclinical decision-making: nutrient-limited growth kinetics, invasion into adjacent esophageal layers, and clinically relevant routes of regional progression. These characteristics make the model particularly valuable for testing chemotherapeutics, targeted agents, combination regimens, and experimental delivery strategies under conditions that better approximate the human disease. Orthotopic modeling also supports longitudinal assessment through non-invasive imaging, enabling tumor burden to be tracked over time without sacrificing cohorts at each time point.

Cell Line Information: EC109

EC109 (also cataloged as Eca-109, EC-109, or Eca109) is a continuously propagated human ESCC line that has been widely adopted in esophageal cancer research for decades. Because certain distributed stocks have been reported to carry HeLa contamination, the line is flagged by the International Cell Line Authentication Committee (ICLAC); accordingly, only lot-specific, STR-authenticated and mycoplasma-tested material should be used for model construction. Key documented attributes are summarized below.

Attribute Description

Key Markers Cytokeratins, EpCAM (CD326); CD44-positive phenotype documented; c-Met expression reported
Molecular Features Enrichment of alterations in cell cycle regulation, TP53, Hippo, and Wnt signaling pathways; up-regulated immune-related gene expression relative to other ESCC lines
Tumorigenicity Tumorigenic in BALB/c nude mice and other immunodeficient strains, forming well-defined xenografts with squamous differentiation features
Drug Sensitivity Parental line responsive to cisplatin, 5-fluorouracil, and paclitaxel; multidrug-resistant sublines available, including EC109/CDDP (cisplatin-selected) and EC109/Taxol (paclitaxel-selected)
Common Applications Tumor proliferation, migration and invasion studies; apoptosis and autophagy research; radiosensitivity and drug resistance studies; xenograft and orthotopic model development

Our Services

Alfa Cytology brings together surgical oncology expertise, authenticated cell line resources, and disciplined in vivo study management to deliver the EC109 orthotopic esophageal cancer model as a dependable preclinical tool. From feasibility assessment and pilot tumorigenicity runs to fully powered efficacy studies with imaging and histology readouts, every engagement is executed under a clearly defined protocol and quality framework—so you receive decision-ready data rather than raw readouts.

Workflow of EC109 Orthotopic Mouse Model Construction

Construction of the EC109 orthotopic model follows a standardized, quality-controlled sequence in which each phase feeds a defined checkpoint before the next begins. The typical workflow comprises the following steps:

  1. Cell line authentication and expansion: EC109 cells from a verified, low-passage stock are STR-profiled and tested for mycoplasma, then expanded under standardized conditions to generate a qualified inoculum.
  2. Pre-implantation cell preparation: Cultures are harvested at optimal confluence, counted and viability-checked, then formulated—often mixed with a basement membrane matrix—at a defined concentration (commonly on the order of 2 × 106 cells per inoculum).
  3. Animal preparation and anesthesia: Immunodeficient mice are acclimatized, randomized, and anesthetized; analgesia and aseptic technique are applied in line with institutional animal welfare requirements.
  4. Surgical exposure of the esophagus: A small cervical or abdominal incision provides controlled access to the esophageal segment selected for implantation, minimizing tissue trauma and operative time.
  5. Orthotopic implantation: The EC109 cell suspension is carefully introduced into the esophageal wall (or applied onto the exposed serosal surface), and the site is closed with fine sutures so the inoculum remains confined to the organ.
  6. Postoperative recovery: Animals are monitored until fully recovered, with body weight, feeding behavior, and surgical wound condition recorded daily during the early postoperative period.
  7. Longitudinal tumor monitoring: Tumor establishment and growth are tracked non-invasively where reporter-labeled cells are used, supplemented by clinical observation, so that randomization into treatment arms occurs at a consistent tumor burden.
  8. Endpoint analysis: At study termination, tumors are excised, measured, and processed for histopathology, immunohistochemistry, and molecular analyses, with full documentation delivered to the client.
