MC38-hPD-L1 Syngeneic Model Service for Colon Cancer

Fig 1: MC38-hPD-L1 Syngeneic Model for Colon Cancer preclinical research.

The MC38-hPD-L1 syngeneic model service for colon cancer is an immune-competent C57BL/6 platform in which the hypermutated MC38 colon adenocarcinoma is engineered to replace murine PD-L1 with its human counterpart, enabling direct evaluation of human-specific PD-L1 therapeutics — monoclonal antibodies, bispecifics, and antibody-drug conjugates — in a fully functional immune system. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated MC38-hPD-L1 expansion and banking, standardized implantation into wild-type or humanized C57BL/6 hosts, scheduled tumor-growth monitoring with optional luciferase imaging, and full histopathology with checkpoint-engagement pharmacodynamics — all configured around each sponsor’s therapeutic format, combination strategy, and endpoint plan.

Overview of MC38-hPD-L1 Syngeneic Model for Colon Cancer

MC38-hPD-L1 solves a problem every developer of human checkpoint biologics eventually faces: clinical-stage anti-PD-L1 antibodies such as durvalumab, atezolizumab, and avelumab do not recognize murine PD-L1, so they cannot be tested in conventional syngeneic systems. The model’s parent, MC38, is a chemically induced colon adenocarcinoma of C57BL/6 origin from the classical Corbett tumor panel (the carcinogen exposure is attributed to N-nitroso-N-methylurethane in the original pharmacology literature and to 1,2-dimethylhydrazine in more recent characterizations), and it is one of the most thoroughly decoded murine tumors in existence: whole-exome sequencing classifies it as a microsatellite-instability, hypermutated, neoantigen-rich tumor resembling human consensus molecular subtype 1 colorectal cancer, with mutations across Tp53, Pten, Smad2/4, Acvr2a, Braf, and truncation lesions in Axin2 — a genotype that explains its brisk, reproducible responsiveness to PD-1/PD-L1 blockade. In the hPD-L1 derivative, the endogenous murine Cd274 gene is knocked out and human CD274 is knocked into its place (or driven from an inserted construct that simultaneously disrupts the mouse locus), so tumor cells present exclusively human PD-L1 at the cell surface while the surrounding immune compartment remains entirely murine and intact.

In vivo efficacy of Caryophylli Cortex extract in the MC38-hPD-L1 humanized syngeneic model — experimental timeline of hPD-L1/MC-38 subcutaneous implantation in hPD-1/C57BL/6J mice, body weight stability, tumor growth curves, tumor suppression rates (73.03% at 100 mg/kg; 68.48% at 300 mg/kg) and final tumor weights after 15 days of daily oral WCC administration. The engineering has been validated at every level: by flow cytometry the lines express human PD-L1 without detectable murine PD-L1; by binding studies human PD-L1 engages murine PD-1, preserving the physiological checkpoint interaction in wild-type C57BL/6 hosts; and tumorigenicity matches parental MC38, with 1 × 106 cells reliably establishing flank tumors. Therapeutically, the model is where human-specific formats prove themselves — a clinically validated anti-hPD-L1 antibody reduced tumor growth at just 1 mg/kg in the published characterization, pembrolizumab, nivolumab, and atezolizumab produced up to 95% growth inhibition with complete regressions and long-term tumor-free survivors in humanized PD-1/PD-L1 knock-in hosts, and a PD-L1×VEGF bispecific achieved 81% growth inhibition versus 56% for a reference bispecific in head-to-head testing. The platform has since been extended into a family — multi-humanized derivatives carrying human CD47, HER2, DLL3, or FAP alongside hPD-L1, and luciferase-ZsGreen dual-reporter versions that render tumor burden visible in vivo — making MC38-hPD-L1 the touchstone system for human-specific checkpoint pharmacology in an immune-competent colon cancer setting.

Cell Line Information: MC38-hPD-L1

The table below consolidates the key biological, engineering, and modeling characteristics of the MC38-hPD-L1 line, its parent, and the derivative formats used in contemporary studies.

Parameter Details
Cell line designation MC38-hPD-L1 (murine MC38 colon adenocarcinoma engineered for human PD-L1); multi-humanized derivatives (hPD-L1 plus hCD47, hHER2, hDLL3, or hFAP) and luciferase-ZsGreen dual-reporter versions are commercially established
Species / genetic background Mouse (Mus musculus), C57BL/6 inbred background; syngeneic in C57BL/6 immunocompetent hosts, with optional pairing to C57BL/6 humanized PD-1/PD-L1 knock-in mice
Parental line origin MC38 (MC-38; historically “colon adenocarcinoma 38”): chemically induced grade III colon adenocarcinoma arising in a female C57BL/6 mouse within the Corbett et al. transplantable tumor panel (Cancer Research 35:2434–2439, 1975); carcinogen attribution varies between NMU (classical pharmacology literature) and 1,2-dimethylhydrazine (recent characterizations)
Parental molecular profile Microsatellite-instability (MSI), hypermutated tumor resembling human CMS1 colorectal cancer; mutations across Tp53, Pten, Smad2, Smad4, Acvr2a, Braf, with Axin2 truncation; high neoantigen burden underpinning strong anti-PD-1/PD-L1 responsiveness
Engineering strategy Endogenous murine Pd-l1 (Cd274) inactivated by CRISPR/Cas9 knockout or promoter-driven insertion, with human CD274 coding sequence knocked into the mouse locus or expressed from an exogenous promoter; clonal selection followed by sequencing and flow-cytometric verification
Expression validation Flow cytometry confirms homogeneous high human PD-L1 surface expression with no detectable murine PD-L1; ELISA and flow cytometry confirm that human PD-L1 binds murine PD-1, preserving the physiological checkpoint interaction
Tumorigenicity validation Subcutaneous growth in C57BL/6 mice comparable to parental MC38 at 1 × 106 cells per site; tumors reliably established for treatment initiation at approximately 100–150 mm³
Documented in vivo efficacy Clinically validated anti-hPD-L1 antibody reduced tumor growth at 1 mg/kg in wild-type C57BL/6; in humanized PD-1/PD-L1 knock-in mice, nivolumab, pembrolizumab, and atezolizumab achieved 70–95% growth inhibition with complete regressions and tumor-free survivors at day 77; pembrolizumab produced 64.9% TGI at day 18 with increased intratumoral IFN-γ in CD4⁺ and CD8⁺ T cells; a PD-L1×VEGF bispecific achieved 81.3% TGI versus 55.6% for a reference bispecific
Recommended culture conditions Standard complete medium (RPMI-1640 or DMEM) with 10% fetal bovine serum at 37 °C in a humidified 5% CO₂ atmosphere, with selection antibiotic maintained as appropriate to the engineering construct
Standard implantation practice Approximately 1 × 106 cells (5 × 105 in humanized-host configurations) implanted subcutaneously into the flank; treatment typically initiated one week post-implantation at approximately 100–150 mm³; reporter versions support pre-palpable detection by luminescence imaging
Host configurations Wild-type C57BL/6 for anti-human-PD-L1 therapeutics (tumor-side humanization suffices because hPD-L1 engages murine PD-1); C57BL/6 humanized PD-1/PD-L1 knock-in mice when both sides of the checkpoint interaction must be human (e.g., anti-human-PD-1 antibodies such as nivolumab and pembrolizumab)
Key research applications Efficacy screening of human-specific anti-PD-L1 monoclonal antibodies; bispecific and multispecific checkpoint constructs (PD-L1×VEGF and beyond); antibody-drug conjugates and fusion proteins; combination regimens with chemotherapy, targeted agents, or radiation; pharmacokinetic/pharmacodynamic bridging between surrogate murine reagents and clinical candidates; imaging-tracked burden studies using luciferase reporter derivatives

Our Services

Translating a human-specific checkpoint construct into an immune-competent setting is exactly where engineering rigor pays off, and Alfa Cytology runs the MC38-hPD-L1 program to that standard: authenticated banks of the knock-in line, both validated host configurations, scheduled dosing with luminescence tracking where reporters are used, and endpoint pathology paired with checkpoint-engagement pharmacodynamics — all under one accountable preclinical operation.

Workflow of MC38-hPD-L1 Syngeneic Model Construction

Construction follows the published methodology for the platform’s two host configurations: MC38-hPD-L1 cells are expanded under authenticated conditions with selection maintained, implanted subcutaneously into wild-type or humanized C57BL/6 mice, grown to a standardized baseline, randomized, treated, and analyzed with checkpoint-focused endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: MC38-hPD-L1 cells are STR/identity-verified, confirmed mycoplasma-negative, re-verified for exclusive human PD-L1 surface expression by flow cytometry, and expanded into low-passage working banks under selection.
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at approximately 1 × 106 cells (5 × 105 for humanized-host arms) in 100 μL of PBS or a Matrigel-supported suspension.
  3. Anesthesia and implantation: Wild-type or humanized PD-1/PD-L1 C57BL/6 mice are anesthetized with isoflurane, and cells are implanted subcutaneously into the flank; reporter-arm animals are baseline-imaged where applicable.
  4. Establishment monitoring and randomization: Tumors are measured by caliper (and by luminescence for reporter arms) from first signal; animals are randomized once tumors reach approximately 100–150 mm³, balancing baselines across arms.
  5. Treatment administration: Experimental arms — human-specific anti-PD-L1 monoclonals, bispecifics, or ADCs alone and in combination with chemotherapy, targeted agents, or radiation — are dosed per study design (clinical benchmark schedules include intraperitoneal dosing three times weekly) with body-weight and toxicity monitoring.
  6. Imaging and interim measurements: Caliper measurements track growth kinetics; luciferase-ZsGreen arms are imaged on a scheduled cadence to quantify tumor burden and detect response before palpable change.
  7. Endpoint and pharmacodynamic analysis: Endpoints integrate tumor-growth inhibition, regression and survival with tumor-infiltrating-lymphocyte profiling by flow cytometry (CD4⁺/CD8⁺ T cells, IFN-γ, checkpoint expression), pharmacodynamic confirmation of target engagement, and histopathology (H&E with Ki67, CD8, and human PD-L1 immunohistochemistry).
