HOS / HOS-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: HOS / HOS-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The HOS / HOS-luc orthotopic mouse model of sarcoma engrafts a classic human osteosarcoma line into the marrow cavity of the tibia in immunodeficient mice, where it grows as a slow, osteolytic primary tumor and ultimately seeds the lung. From authenticated cell expansion and intratibial implantation surgery to weeks-long luciferase monitoring, CT-based bone assessment and metastasis confirmation, Alfa Cytology delivers complete HOS / HOS-luc orthotopic study programs shaped around each sponsor’s timeline, compound and endpoint needs—exclusively within preclinical research.

Overview of HOS / HOS-luc Orthotopic Mouse Model for Sarcoma

HOS is among the oldest continuously cultured human osteosarcoma lines, maintained by ATCC as CRL-1543, and was isolated decades ago from a primary osteosarcoma of the distal right femur in a 13-year-old Caucasian girl. In culture the cells are elongated and polygonal, adherent as a monolayer with a mesenchymal bearing, and alkaline-phosphatase positive—yet their full osteogenic program only switches on inside organized three-dimensional matrices, since routine monolayer culture does not deposit calcium apatite. The line occupies a special place in osteosarcoma research as the parental stock from which two workhorse derivatives were raised: 143B, transformed with mutant K-ras into a fiercely aggressive, highly metastatic line, and MNNG/HOS, transformed with a chemical carcinogen and prized in chemotherapy-resistance studies. Compared with these offspring, HOS itself is comparatively restrained—a property that turns out to be advantageous in vivo.

In the orthotopic format, luciferase-labeled HOS-luc cells (commonly a lentiviral Luc2 reporter, often co-expressed with a fluorescent tag) are loaded into the proximal tibial cavity of immunodeficient mice, suspended in a small volume of Matrigel that limits immediate washout into the circulation. Primary tumors engraft with 100% consistency in NSG mice but expand slowly and diffusely, showing a predominantly osteolytic profile on CT scans—bone destruction rather than the dense osteoblastic reaction seen with some other lines—while pulmonary metastases typically declare themselves around day 26 after implantation and appear as numerous but small nodules in most animals, confirmed at necropsy by ex vivo imaging and histology. Engraftment is markedly better in NSG than in nude recipients, and the protracted disease course (studies may extend beyond 100 days) suits long treatment windows and resistance-emergence designs, including the methotrexate- and doxorubicin-resistant HOS derivatives already characterized in this exact setting. The trade-offs are equally clear: the immunocompromised host rules out immunotherapy questions, and the slow kinetics demand patience and disciplined imaging schedules—both of which the luciferase readout, detectable within days of implantation, makes far easier to manage.

Cell Line Information: HOS / HOS-luc

HOS and its luciferase-labeled derivative HOS-luc descend from the same human primary tumor; the reporter line additionally stably expresses firefly luciferase (typically the Luc2 variant) for quantitative optical tracking. Key characteristics are summarized below.

Item Details
Cell line name HOS (human osteosarcoma); ATCC CRL-1543; RRID: CVCL_0312
Species / host requirement Homo sapiens; requires immunodeficient hosts — engraftment is superior in NSG mice than in athymic nude mice
Donor profile 13-year-old female; primary osteosarcoma of the distal right femur
Derived variants Parental line of 143B (K-ras-transformed, highly metastatic) and MNNG/HOS (carcinogen-transformed) — together a standard comparative panel for osteosarcoma research
Morphology / growth Elongated, polygonal cells growing as an adherent monolayer; restrained growth compared with aggressive derivatives
Osteogenic biology Alkaline phosphatase positive; mineralization (apatite deposition) occurs only within organized three-dimensional matrices, not in routine 2D culture; mesenchyme-like organization
Orthotopic tumor behavior Intratibial tumors engraft with 100% take (5/5) in NSG mice but grow slowly; predominantly osteolytic morphology on CT
Metastatic behavior Lung metastases detected from ~day 26 after implantation in most NSG mice; nodules numerous but small (<29 per lung); occasional spleen and bone lesions reported
Reporter variant (HOS-luc) Lentiviral Luc2 firefly luciferase, frequently co-expressed with mKate2; bioluminescence detectable within days of implantation and validated in vitro by cell-titer titration with D-luciferin
Culture conditions DMEM + 10% FBS (+ 1% penicillin/streptomycin); 37 °C, 5% CO2, humidified incubator
Typical orthotopic inoculum 1.5 × 106 cells in 10 µL Matrigel (4 mg/mL) delivered into the proximal tibial medullary cavity of NSG mice
Drug-resistant derivatives Methotrexate-resistant (HOS-Luc/mKate2/MTX) and doxorubicin-resistant (HOS-Luc/mKate2/DOXO) variants characterized in the same orthotopic system
Representative applications Long-window targeted-therapy evaluation, multidrug-resistance functional imaging (e.g., 99mTc-sestamibi washout with P-glycoprotein inhibitors), natural-compound screening, mineralization and 3D-tumor biology, metastasis-kinetics research

Our Services

Combining meticulous bone surgery, quantitative bioluminescence and CT imaging and in-house histopathology, Alfa Cytology adapts every HOS / HOS-luc orthotopic engagement to the sponsor’s goals—setting implantation conditions, imaging cadence and endpoint panels so that even slow-burn tumor kinetics resolve into clear, decision-ready preclinical findings.

Workflow of HOS / HOS-luc Orthotopic Mouse Model Construction

Orthotopic construction of the HOS / HOS-luc model is a survival-surgery workflow performed under inhalation anesthesia with pre-emptive analgesia, aseptic technique and sustained post-operative observation, reflecting the long study horizons this slow-growing model demands. The procedure typically advances through the following stages:

  1. Cell line qualification: HOS or HOS-luc vials are mycoplasma-tested and STR-verified; reporter performance is confirmed in vitro by serial cell-titer bioluminescence titration with D-luciferin before expansion.
  2. Animal preparation: immunodeficient mice (NSG preferred; athymic nude acceptable, typically 6–8 weeks old) are acclimatized under appropriate containment, health-screened and randomized by body weight ahead of surgery.
  3. Anesthesia, analgesia and asepsis: mice receive isoflurane anesthesia with pre-emptive buprenorphine, the surgical leg is clipped and disinfected, and a sterile field with sterile instruments is maintained throughout.
  4. Surgical exposure: a small incision over the knee exposes the proximal tibia, providing access to the medullary cavity under magnification.
  5. Intratibial inoculation: HOS / HOS-luc cells (typically 1.5 × 106 in 10 µL Matrigel at 4 mg/mL) are injected into the proximal tibial cavity with a fine needle; the Matrigel suspension limits immediate spread into the circulation.
  6. Closure and recovery: soft tissue and skin are closed with absorbable sutures and wound clips, post-operative analgesia is maintained, and gait, body weight and leg deformation are monitored on a defined schedule.
  7. Longitudinal monitoring: primary tumor growth is tracked in vivo by weekly bioluminescence imaging after intraperitoneal D-luciferin for HOS-luc, with periodic CT scans documenting osteolytic bone change; given the slow kinetics, study timelines commonly extend beyond 100 days.
  8. Endpoint and analysis: at termination the tibial tumor is excised and weighed, lungs and spleen are removed for ex vivo bioluminescence and metastasis counting, and tissues are processed for histology (including HES and luciferase staining), immunohistochemistry and molecular analysis.
