Saos-2 / Saos-2-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: Saos-2 / Saos-2-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The Saos-2 / Saos-2-luc orthotopic mouse model of sarcoma delivers a strongly osteoblastic human tumor directly into the tibial bone bed of immunodeficient mice, where it builds mineralizing, bone-forming primary tumors with measurable spread to lung and beyond. From authenticated cell expansion and intratibial implantation surgery to serial luciferase imaging, CT-based bone assessment and metastasis confirmation, Alfa Cytology delivers complete Saos-2 / Saos-2-luc orthotopic study programs configured around each sponsor’s compound, schedule and endpoint needs—exclusively within preclinical research.

Overview of Saos-2 / Saos-2-luc Orthotopic Mouse Model for Sarcoma

Saos-2 was established in 1973 by Jorgen Fogh from the osteogenic sarcoma of an 11-year-old Caucasian girl and has since become the benchmark human line for osteoblast biology. The cells carry a truncating TP53 mutation and produce no detectable p53 protein, which made them a favorite substrate for reconstitution studies of the p53 pathway; they are epithelial in appearance, adhere as a monolayer, and carry an aneuploid genome of roughly 56 chromosomes riddled with complex rearrangements. Their defining trait, however, is how faithfully they behave like osteoblasts: basal alkaline phosphatase activity is exceptionally high, receptors for 1,25-dihydroxyvitamin D3 and parathyroid hormone are functional—the latter driving a robust cAMP response—and under osteogenic induction the cells deposit calcified nodules that stain with Alizarin Red while upregulating late markers such as SOST, DMP1, PHEX and MEPE. Intriguingly, these highly differentiated cells refuse to grow as subcutaneous tumors in immunocompromised mice, yet when placed directly into bone they engraft and progress, revealing how decisively the organ microenvironment governs their malignant expression.

In the orthotopic setting, a bone-adapted derivative of the line (Saos-2-B) carrying a lentiviral Luc2 luciferase reporter—often alongside the fluorescent mKate2 tag—is loaded into the proximal tibial cavity of immunodeficient mice, suspended in a small Matrigel volume that curbs immediate vascular washout. Engraftment is efficient (five of six NSG mice), bioluminescence is detectable within five days of implantation, and the primary tumor outgrows most comparator osteosarcoma xenografts, exceeding 1010 photon-scale signal by roughly six weeks in NSG animals. On CT the lesion is unmistakably osteoblastic: aberrant new bone condenses within and beyond the cortex, accompanied by periosteal reaction, cortical breach and only focal lysis—the mirror image of the predominantly osteolytic phenotype seen with other osteosarcoma lines. Pulmonary metastases appear between days 26 and 42 in NSG recipients, and unlike the usual pattern the model also delivers sporadic lesions to spleen and to the opposite, uninjected bone. Growth and spread are markedly slower in nude than in NSG hosts, and the immunodeficient requirement excludes immuno-oncology questions; within those boundaries the model excels for bone-matrix-targeting agents, mineralization and biomaterial evaluation, p53-pathway research, chemotherapy-resistance studies with methotrexate-adapted derivatives, and any program that needs a fast, faithfully osteoblastic primary tumor.

Cell Line Information: Saos-2 / Saos-2-luc

Saos-2 and its luciferase-labeled derivative Saos-2-luc descend from the same human osteogenic sarcoma; orthotopic studies typically employ the bone-adapted variant, and the reporter line additionally stably expresses firefly luciferase for quantitative optical tracking. Key characteristics are summarized below.

Item Details
Cell line name Saos-2; ATCC HTB-85; established 1973 (J. Fogh)
Species / host requirement Homo sapiens; classically non-tumorigenic in subcutaneous assays even in immunocompromised mice, but forms osteoblastic tumors when implanted directly into bone — orthotopic engraftment 5/6 in NSG mice
Donor profile 11-year-old Caucasian female; osteogenic sarcoma
Karyotype Aneuploid; ~56 chromosomes with complex rearrangements
Morphology / growth Epithelial-like, adherent monolayer; reported doubling time of roughly 48 hours
TP53 status Truncating TP53 mutation with no detectable p53 protein; classic substrate for p53-pathway reconstitution studies
Osteoblastic phenotype Very high basal alkaline phosphatase; functional receptors for 1,25-dihydroxyvitamin D3 and parathyroid hormone (cAMP response); produces osteocalcin and osteopontin
Mineralization Gold-standard osteogenic capacity: robust calcified nodules (Alizarin Red positive) under osteogenic induction with β-glycerophosphate, ascorbic acid and dexamethasone; late markers SOST, DMP1, PHEX and MEPE upregulated under mineralizing conditions
Orthotopic tumor behavior Intratibial tumors grow faster than comparator osteosarcoma xenografts (signal >1010 by ~40–50 days in NSG); CT shows osteocondensation — aberrant new bone with periosteal reaction and focal lysis — first noted around day 41; histology predominantly osteoblastic with occasional fibroblastic components
Metastatic behavior Lung metastases detected 26–42 days after intratibial implantation in NSG mice; sporadic spleen and contralateral-bone lesions also reported; spread markedly slower in nude hosts
Reporter variant (Saos-2-luc) Lentiviral Luc2 firefly luciferase, frequently co-expressed with mKate2 (>90% positive by flow cytometry); validated by in vitro cell-titer titration; in vivo signal detectable from day 5 post-implantation
Culture conditions DMEM + 10% FBS (+ 1% penicillin/streptomycin); 37 °C, 5% CO2, humidified incubator
Typical orthotopic inoculum 1.5 × 106 cells in 10 µL Matrigel (4 mg/mL) delivered into the proximal tibial medullary cavity of NSG mice; a paratibial variant with periosteum activation is established
Representative applications Bone-matrix and mineralization targeting, biomaterial and osseointegration evaluation, p53-pathway research, osteosarcoma metastasis-kinetics studies, chemotherapy-resistance programs using methotrexate-adapted derivatives

Workflow of Saos-2 / Saos-2-luc Orthotopic Mouse Model Construction

Orthotopic construction of the Saos-2 / Saos-2-luc model is a survival-surgery workflow performed under inhalation anesthesia with pre-emptive analgesia, aseptic technique and disciplined post-operative observation, reflecting the model’s brisk growth in highly immunodeficient hosts. The procedure typically advances through the following stages:

  1. Cell line qualification: Saos-2 or Saos-2-luc vials are mycoplasma-tested and STR-verified; reporter performance is confirmed in vitro by serial cell-titer bioluminescence titration with D-luciferin before expansion.
  2. Animal preparation: immunodeficient mice (NSG preferred given superior engraftment; nude acceptable, typically 6–8 weeks old) are acclimatized under appropriate containment, health-screened and randomized by body weight ahead of surgery.
  3. Anesthesia, analgesia and asepsis: mice receive isoflurane anesthesia with pre-emptive buprenorphine, the surgical leg is clipped and disinfected, and a sterile field with sterile instruments is maintained throughout.
  4. Surgical exposure: a small incision over the knee exposes the proximal tibia, providing access to the medullary cavity under magnification.
  5. Intratibial inoculation: Saos-2 / Saos-2-luc cells (typically 1.5 × 106 in 10 µL Matrigel at 4 mg/mL) are injected into the proximal tibial cavity with a fine needle; the Matrigel suspension limits immediate spread into the circulation.
  6. Closure and recovery: soft tissue and skin are closed with absorbable sutures and wound clips, post-operative analgesia is maintained, and gait, body weight and leg deformation are monitored on a defined schedule.
  7. Longitudinal monitoring: primary tumor growth is tracked in vivo by weekly bioluminescence imaging after intraperitoneal D-luciferin for Saos-2-luc, with periodic CT scans documenting osteocondensation and cortical change; NSG cohorts frequently reach endpoint within 10–12 weeks.
  8. Endpoint and analysis: at termination the tibial tumor is excised and weighed, lungs, spleen and contralateral bone are removed for ex vivo bioluminescence and metastasis counting, and tissues are processed for histology (including HES and luciferase staining), immunohistochemistry and molecular analysis.
