KYSE-30-luc Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: KYSE-30-luc Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The KYSE-30-luc orthotopic mouse model pairs a luciferase-labeled human esophageal squamous cell carcinoma line with implantation into the native esophageal wall, allowing tumor growth, local invasion, and treatment response to be tracked in real time through non-invasive bioluminescence imaging. Drawing on proven surgical techniques and quantitative imaging expertise, Alfa Cytology supplies end-to-end support for KYSE-30-luc orthotopic model construction, serial monitoring, histopathological validation, and preclinical efficacy testing.

Overview of KYSE-30-luc Orthotopic Mouse Model for Esophageal Cancer

KYSE-30-luc is the bioluminescent derivative of KYSE-30, one of the best-known members of the KYSE panel of human esophageal squamous cell carcinoma (ESCC) lines established at Kyoto University from a well-differentiated tumor of a 64-year-old untreated Japanese male patient. The parental line grows rapidly (doubling time around 20.8 hours) and carries an extensively characterized genome that includes a TP53 intron-6 splice mutation, a HRAS p.Gln61Leu mutation, a truncating CDKN2A (p16) alteration, amplification of MYC, CCND1, and HER2, and exceptionally high EGFR expression. To convert this line into a trackable imaging tool, KYSE-30 cells are engineered to stably express firefly luciferase — introduced with a pLVX-Luciferase vector, enriched under stepwise G418 selection, and confirmed for strong luciferase activity by dual-luciferase reporter assay in vitro — without compromising their tumorigenic behavior.

The real value of the luciferase tag emerges when the cells are returned to their organ of origin. Injected into the muscularis externa of the abdominal esophagus near the diaphragm, KYSE-30-luc cells form tumors with a 100% take rate, and because the emitted photons scale with viable tumor mass, whole-body imaging captures tumor initiation and progression in vivo week by week without sacrificing animals. Histologically, these orthotopic tumors faithfully mirror human ESCC: nodular growth causing esophageal luminal stricture, invasion through the adventitia, lymphovascular invasion, and perineural infiltration. The platform's utility has been demonstrated functionally as well — AKT shRNA knockdown cells showed significantly suppressed orthotopic tumor growth compared with controls, underscoring its value for gene-function studies, metastasis research, and therapeutic regimen testing in a physiologically relevant microenvironment.

Our Services

Alfa Cytology equips researchers with a turnkey KYSE-30-luc orthotopic solution: authenticated luciferase-positive cells, precision intramural implantation surgery, scheduled IVIS imaging with quantitative signal analysis, and terminal histopathology — all executed under one quality-managed preclinical workflow.

Workflow of KYSE-30-luc Orthotopic Mouse Model Construction

The KYSE-30-luc orthotopic model is established by direct microinjection of luciferase-labeled cells into the esophageal wall, followed by serial whole-body bioluminescence imaging. The workflow typically proceeds as follows.

  1. Luciferase labeling and clone selection: KYSE-30 cells are transfected with pLVX-Luciferase plasmids, placed under stepwise G418 selection to enrich luciferase-positive cells, and isolated as stable clones.
  2. Reporter verification and quality control: Luciferase activity is quantified by dual-luciferase reporter assay, and cell identity is confirmed by STR profiling with mycoplasma testing before expansion.
  3. Inoculum preparation: KYSE-30-luc cells are expanded in RPMI 1640 with 10% FBS, harvested during exponential growth, counted, and resuspended in plain medium at a working concentration delivering 2 × 105 cells in 10 μL per mouse.
  4. Anesthesia and surgical exposure: Athymic nude mice are anesthetized and positioned supine; a small upper-abdominal incision is made, retractors hold the wound open, the left liver lobe is gently raised, and caudal traction on the stomach straightens the abdominal esophagus for clear visualization under a dissecting microscope.
  5. Intramural microinjection: Using a 30-gauge insulin syringe, the cell suspension is injected into the muscularis externa of the esophagus; formation of a local edema at the injection site confirms correct intramural delivery, and a cotton swab is pressed on the site as the needle is withdrawn to prevent leakage into the peritoneal cavity.
  6. Closure and postoperative care: The peritoneum is closed with Vicryl absorbable sutures and the skin with wound clips; mice are warmed until fully mobile and monitored daily for body weight, hydration, appetite, and behavior.
  7. Serial imaging and endpoint analysis: Tumor growth is monitored weekly by IVIS bioluminescence imaging after intraperitoneal D-luciferin (150 mg/kg) in vivo; at study end, orthotopic tumors are excised, weighed, and processed for H&E histology to confirm adventitia invasion, lymphovascular invasion, and perineural infiltration.

Case Study-KYSE-30-luc Orthotopic Mouse Model Development

Alfa Cytology has implemented the KYSE-30-luc orthotopic platform in several preclinical programs covering imaging-based model development, longitudinal efficacy tracking of investigational agents, and signaling-pathway interrogation. In these projects, luciferase signal kinetics in vivo provided early, quantitative readouts of tumor growth, while terminal histopathology confirmed characteristic ESCC invasion patterns. Project specifics and tailored study designs can be discussed upon request.
