L5178-R (LY-R) Syngeneic Model Service for Lymphoma

Fig 1: L5178-R (LY-R) Syngeneic Model for Lymphoma preclinical research.

The L5178-R (LY-R) syngeneic model service for lymphoma is an immune-competent DBA/2 platform built on the X-ray-resistant thymic lymphoma line LY-R and its radiosensitive counterpart LY-S, providing a uniquely paired system for radiation biology, genotoxicity-informed drug evaluation, and checkpoint immunotherapy studies. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated LY-R / LY-S expansion with the platform’s documented culture-stability controls, subcutaneous, intraperitoneal, intravenous, and intracranial inoculation formats, scheduled burden monitoring, and full lymphoma histopathology with DNA-damage-response pharmacodynamics — all configured around each sponsor’s radiation, radiosensitizer, genotoxicity, or immunotherapy hypothesis.

Overview of L5178-R (LY-R) Syngeneic Model for Lymphoma

L5178-R (LY-R) is one of the most scientifically consequential mouse lymphoma lines ever derived — a 3-methylcholanthrene-induced thymic lymphoma arising in an eight-month-old female DBA/2 mouse in 1951, carried for decades as a transplantable ascites tumor by weekly intraperitoneal passage, and adapted to culture by Fischer in 1958. Its fame rests on a singular property: resistance to ionizing radiation. LY-R cells exhibit an X-ray mean lethal dose (D₀) of 0.91 grays yet remain exquisitely sensitive to ultraviolet light (D₀ = 0.7 J/m²), brief heat treatment at 43 °C, and several antineoplastic drugs including cisplatin — a sensitivity pattern rooted in differential DNA repair efficiency. The line’s paired derivative, LY-S, is X-ray sensitive (D₀ = 0.56 Gy) but UV resistant, and the two together became the canonical experimental system for dissecting the relationship between DNA repair capacity and tumor behavior. Molecularly, every L5178Y subline examined — LY-R, LY-S, and the mutagenicity-test clone — carries the same heterozygous codon 170 (TGC→CGC) mutation in p53, placing the platform’s radiation phenotypes in a defined tumor-suppressor context.

In vivo, LY-R is highly tumorigenic in syngeneic DBA/2 mice while LY-S is practically nontumorigenic — a tumorigenicity difference that tracks with repair capacity and that makes the pair uniquely informative when radiation or DNA-damaging agents are part of the study question. A phenomenon that any user of this platform must respect was documented definitively in 1983: after prolonged culture (five months to four years), LY-R cells can convert into the LY-S phenotype, losing both X-ray resistance and tumorigenicity — evidence that the line’s behavior is a maintained, metastable state rather than a fixed genotype, and the reason why low-passage banking and scheduled phenotypic verification are essential quality steps for this model. The platform’s reach extends well beyond radiobiology: its tk+/− clone 3.7.2C is the OECD 476 mouse lymphoma assay standard for genotoxicity testing; intracranial inoculation into B6D2F1 hybrids supports central-nervous-system leukemia studies with temozolomide-plus-PARP-inhibitor combinations; and L5178Y cells have served since the 1970s as classic substrates for macrophage phagocytosis and tumor-associated-macrophage interaction studies, being one of the first lines shown to be ingested by activated peritoneal macrophages and to prime macrophages through CD40:CD40L contact. Checkpoint-inhibitor response data are also documented for the syngeneic flank format, rounding out a model whose 70-year history touches mutagenesis, radiation oncology, macrophage biology, and immuno-oncology alike.

Cell Line Information: L5178-R (LY-R)

The table below consolidates the key biological, cultural, and modeling characteristics of the LY-R line, its LY-S counterpart, and the tk+/− mutagenicity-test clone.

Parameter Details
Cell line designation L5178-R (LY-R; synonyms L5178Y-R, L5178YR, LYR); paired radiosensitive derivative L5178-S (LY-S); mutagenicity-test clone L5178Y tk+/− 3.7.2C (OECD 476 standard)
Species / genetic background Mouse (Mus musculus), DBA/2 inbred background; syngeneic in DBA/2 immunocompetent hosts; H-2d haplotype shared with BALB/c enables H-2-matched alternative hosts
Origin 3-methylcholanthrene-induced thymic lymphoma arising in an 8-month-old female DBA/2 mouse (July 1951, Yale); maintained as a transplantable ascites tumor by weekly intraperitoneal passage (classically 1 × 106 cells); in vitro suspension culture first described by Fischer (1958)
Depository / catalogue ATCC CRL-1722 (LY-R); ATCC CRL-1723 (LY-S); Cellosaurus CVCL_4234 (LY-R), CVCL_4235 (LY-S), CVCL_2097 (parent L-5178-Y)
Tumor type Murine thymic (T-cell) lymphoblastic lymphoma/leukemia
Radiation phenotype LY-R: X-ray resistant (D₀ = 0.91 Gy), UV sensitive (D₀ = 0.7 J/m²), heat sensitive (43 °C, up to 60 min), sensitive to certain antineoplastics (e.g., cisplatin); LY-S: X-ray sensitive (D₀ = 0.56 Gy), UV resistant (D₀ = 5.5 J/m²), heat and drug resistant — differences paralleled by DNA-repair efficiency
Culture-stability phenomenon LY-R cells cultured for 5 months to 4 years convert to the LY-S phenotype, simultaneously losing X-ray resistance and tumorigenicity — scheduled phenotypic verification and low-passage banking are therefore mandatory quality steps (Beer et al., Cancer Research 43:4736–4742, 1983)
Molecular features All L5178Y sublines examined (LY-R, LY-S, tk+/− 3.7.2C) are heterozygous for a codon 170 (TGC→CGC) p53 mutation; multicolor FISH shows only two normal chromosomes (3 and 10) with eight documented translocations and extensive aneuploidy
Growth properties Suspension lymphoblasts with population doubling times of 12–15 hours (LY-R) and ≈10 hours (LY-S); maintain logarithmic cultures between 3 × 104 and 1 × 106 cells/mL
Recommended culture conditions RPMI-1640 with 10% fetal bovine serum (LY-R standard); Fischer’s medium with 2 mM glutamine and 10% horse serum for the tk+/− 3.7.2C clone; 37 °C, 5% CO₂
Implantation formats Classical ascites maintenance by intraperitoneal passage of 1 × 106 cells (or 5 × 106 in alternative protocols); subcutaneous hindlimb inoculation at 4 × 106 cells (0.2 mL of 2 × 107 cells/mL) for solid tumors; systemic intravenous models in DBA/2 (CrownBio documented both subcutaneous and systemic formats); intracranial 1 × 104 cells in 0.03 mL into B6D2F1 (C57BL/6 × DBA/2) hybrids for CNS leukemia studies
Documented in vivo responses Temozolomide (100–200 mg/kg intraperitoneally) combined with PARP inhibition enhanced survival in the intracranial CNS leukemia model (Blood 99:2241, 2002); LY-R highly tumorigenic in DBA/2 while LY-S is practically nontumorigenic; live-cell vaccination with IL-4-secreting L5178 variants induced long-lasting CD4⁺/CD8⁺-dependent systemic protection with dormant bone-marrow persistence correlating to immunity duration (PNAS, 1999)
Host strains DBA/2 immunocompetent mice (syngeneic); B6D2F1 (C57BL/6 × DBA/2) hybrids for intracranial studies; H-2d-matched BALB/c hosts used historically
Key research applications Radiosensitizer and radioprotector evaluation with an isogenic resistant-versus-sensitive pair; genotoxicity bridging via the OECD 476 tk+/− assay; CNS-directed therapy studies in the intracranial format; macrophage and tumor-associated-macrophage interaction biology; checkpoint inhibitor benchmarking in the flank format; DNA-repair-capacity correlation studies with paired survival and tumorigenicity endpoints

Our Services

A platform whose defining phenotype is metastable demands the strictest culture governance in the portfolio, and Alfa Cytology runs LY-R accordingly: authenticated banks with scheduled X-ray-resistance and tumorigenicity re-verification, all four validated inoculation formats, and endpoint analysis that pairs survival and tumorigenicity with DNA-damage-response pharmacodynamics — all under one accountable preclinical operation.

Workflow of L5178-R (LY-R) Syngeneic Model Construction

Construction follows the classical and contemporary methodologies for this platform: LY-R or LY-S cells are expanded under authenticated, passage-controlled conditions, verified for their defining radiation phenotype, implanted into DBA/2 or hybrid hosts in the study-selected format, treated, and analyzed with radiation- and immune-appropriate endpoints. The workflow comprises the following stages.

  1. Cell authentication and phenotype verification: LY-R / LY-S cells are identity-verified, confirmed mycoplasma-negative, and re-verified for their defining phenotype (X-ray D₀ by clonogenic assay where the design requires) at defined passage intervals to guard against the documented LY-R → LY-S conversion.
  2. Cell harvest and formulation: Log-phase cultures are counted and viability-checked, then resuspended at the study-specified dose: 1 × 106 cells intraperitoneally for ascites maintenance, 4 × 106 cells subcutaneously for solid tumors, or 1 × 104 cells in 0.03 mL for intracranial CNS studies, in PBS or complete medium per validated protocol.
  3. Anesthesia and inoculation: DBA/2 or B6D2F1 mice are anesthetized with isoflurane (or ketamine/xylazine per legacy protocols); cells are implanted by the study-selected route — subcutaneous hindlimb, intraperitoneal, intravenous, or intracranial through a stereotactically guided frontal-bone entry.
  4. Establishment monitoring: Ascites and subcutaneous formats are tracked by scheduled palpation, caliper measurement, and body-weight; intracranial cohorts are monitored for neurological signs as humane endpoints.
  5. Randomization and treatment administration: Experimental arms — radiation fractions and radiosensitizers, temozolomide-plus-PARP-inhibitor combinations, genotoxic agents, checkpoint inhibitors, and macrophage-targeting constructs — proceed per study design with paired LY-S comparison arms where the mechanism requires.
  6. Interim burden and DNA-damage monitoring: Tumor kinetics are tracked on schedule; in vitro companion assays (clonogenic survival, γ-H2AX foci, comet assay) are run on recovered cells to connect in vivo response to repair-capacity changes.
  7. Endpoint and histopathological analysis: Endpoints integrate survival, ascites or tumor burden, and — for paired designs — tumorigenicity re-challenge, with histopathology (H&E confirming T-lymphoblastic morphology, plus γ-H2AX, Ki67, CD8, and marker-specific immunohistochemistry) and organ-level metastatic or CNS mapping.
