LLC1 / LLC1-luc Orthotopic Mouse Model Service for Lung Cancer

Fig 1: LLC1 / LLC1-luc Orthotopic Mouse Model for Lung Cancer preclinical research.

The LLC1 / LLC1-luc orthotopic mouse model for lung cancer grows a KRAS-mutant Lewis lung carcinoma within the pulmonary environment of immunocompetent C57BL/6 mice, pairing an immune-intact tumor microenvironment with non-invasive bioluminescence readouts. Operating strictly in preclinical oncology, Alfa Cytology delivers this syngeneic platform as a complete service: authenticated LLC1 / LLC1-luc cell supply, route-matched orthotopic inoculation (tail vein, intrathoracic, or intranasal), scheduled in vivo imaging, and full histopathological endpoint analysis.

Overview of LLC1 / LLC1-luc Orthotopic Mouse Model for Lung Cancer

LLC1 (LL/2) descends from the Lewis lung carcinoma, one of the oldest transplantable mouse tumors in existence, which arose spontaneously in a C57BL/6 mouse in 1951 and was passed through generations of syngeneic hosts ever since. The widely used ATCC version, catalogued as CRL-1642, was established by Bertram and Janik in 1980 and is now understood—through whole-exome sequencing—to originate from a male C57BL/6L donor and to carry an unusually high mutation burden accumulated during decades of laboratory propagation. Genetically, LLC1 mirrors major features of human non-small cell lung cancer: an activating KRAS G12C mutation, Trp53 and Cdkn2a alterations, and prominent Myc expression. Its long passage history in immune-competent animals has also selected a highly immune-evasive phenotype with a strongly immunosuppressive tumor microenvironment, making LLC1 the standard syngeneic lung cancer model—and a key counterpoint to immunotherapy-sensitive lines such as CT26—for preclinical immunology research.

Blood-derived CTCs from orthotopic LLC-luc tumors formed distal metastases detectable by in vivo and ex vivo bioluminescence in liver, adrenal gland, bone and brain after intracardiac injection, and metastatic distribution of the LLC-based syngeneic model mirrored human lung cancer patterns better than the A549 xenograft. (Picabea, Beñat, et al., 2025)

For orthotopic lung cancer work, luciferase-tagged LLC1 (LLC1-luc, including the commercially available CRL-1642-LUC2 reporter) can be introduced through several route-matched strategies, each answering a different question. Tail-vein injection of 5 × 105 to 1 × 106 cells is the classic approach, though it can seed multiple sites with incomplete penetrance; direct intrathoracic injection of ~5 × 105 cells in a 25 µl volume into the left lung produces confirmed thoracic signals within a week; intranasal instillation (two 50 µl doses of 0.5 × 106 cells) delivers cells non-invasively to the lower respiratory tract with documented reproducibility; and subcutaneous dosing of 5 × 105 cells yields palpable tumors by day five for pharmacology screens. Whole-animal bioluminescence after D-luciferin administration quantifies tumor burden longitudinally, works even in black-furred C57BL/6 animals, and supports two- to four-week therapeutic windows with survival, lung nodule scoring, and H&E confirmation at endpoint. Figure 1 outlines this orthotopic system.

Cell Line Information: LLC1 / LLC1-luc

LLC1 is among the most deeply studied syngeneic lung cancer lines, and its luciferase-tagged derivatives have made it the default choice for imaging-based orthotopic lung research. The defining characteristics are summarized below.

Cell Line Name LLC1 (synonyms: LL/2, LL2, 3LL, Lewis lung carcinoma line 1); luciferase-labeled derivatives: LLC1-luc, LLC-GLF, CRL-1642-LUC2 (ATCC)
Catalog / Accession ATCC CRL-1642 (parent) and CRL-1642-LUC2 (luciferase reporter)
Species / Tissue / Disease Mouse (Mus musculus) / lung / Lewis lung carcinoma, an undifferentiated carcinoma of lung origin
Derivation Descended from the Lewis lung carcinoma arising spontaneously in a C57BL/6 mouse (1951) and serially passaged in syngeneic hosts; the LL/2 line was established by Bertram and Janik in 1980
Genetic Profile KRAS G12C activating mutation (confirmed by WES and single-cell sequencing); Trp53 and Cdkn2a mutations; NRAS alteration also reported; high Myc expression; EGFR p.P992P variant; exceptionally high mutation burden from long-term passaging
Morphology / Growth Rounded cells, loosely attached or floating; mixed adherent/suspension population with epithelial appearance; adherent in culture with easy trypsin passaging
Culture Conditions DMEM-based medium with 10% heat-inactivated fetal bovine serum (37 °C, 5% CO₂); passage twice weekly with 0.25% trypsin-EDTA; strict mycoplasma control essential for immunology work
Syngeneic Host C57BL/6 (white- or black-furred; both compatible with bioluminescence imaging); ~100% tumor take with 5 × 105 subcutaneous cells, palpable by day 5
Immunological Character Highly immune-evasive with an immunosuppressive microenvironment; refractory to anti-PD-1 monotherapy—an informative baseline for combination-immunotherapy and microenvironment-reprogramming studies
Metastatic Behavior Classic model of spontaneous lung metastasis from subcutaneous primary tumors; intravenous and intranasal routes seed lung nodules with reproducible kinetics
Orthotopic Inocula (Published) Tail vein: 5 × 105–1 × 106 cells in 200 µl; intrathoracic: ~5 × 105 cells in 25 µl into the left lung; intranasal: 2 × (0.5 × 106) cells in 50 µl; CT-guided parenchymal injection down to 1 × 104 cells; BLI after D-luciferin (100–150 mg/kg i.p., or 3 mg s.c.)
LLC1-luc Derivative Stable firefly luciferase expression enabling quantitative whole-animal imaging; signals tracked for 3–4 weeks, with detectable photon output from week 3 in most animals
Research Applications Orthotopic lung cancer growth and metastasis studies, immunotherapy and microenvironment-reprogramming combinations, KRAS-pathway targeting, antiangiogenic and vascular-disrupting agent evaluation, radiotherapy and radiosensitizer testing

An LLC1 / LLC1-luc engagement with Alfa Cytology is executed as one continuous program: route selection matched to the scientific question, rigorous mycoplasma control for immunology-grade work, pre-agreed IVIS imaging cadence, and a terminal package combining lung nodule counts, ex vivo organ imaging, and flow-cytometric profiling of the tumor microenvironment.

Workflow of LLC1 / LLC1-luc Orthotopic Mouse Model Construction

Establishing an LLC1 or LLC1-luc orthotopic lung tumor begins with a route decision—each inoculation strategy carries distinct biological implications—followed by disciplined cell handling, imaging-driven monitoring, and structured endpoint analysis. The workflow below describes the standard execution of this platform.

  1. Cell Bank Verification and Expansion: LLC1 or LLC1-luc stocks are authenticated (ATCC-supplied reporter lines verified for growth, mycoplasma, and species identity) and expanded in DMEM-based medium, with strict contamination control given the line’s immunology applications.
  2. Luciferase Calibration (LLC1-luc Only): In vitro signal per cell is verified so that in vivo photon flux can be converted into relative tumor burden across cohorts.
  3. Host Preparation and Route Selection: C57BL/6 mice are acclimatized and randomized by body weight; the study design selects the inoculation route—tail vein for systemic seeding, intrathoracic injection for a single defined lung nodule, or intranasal instillation for a non-invasive, microenvironment-focused format.
  4. Orthotopic Inoculation: Cells are delivered per the chosen route: tail vein (5 × 105–1 × 106 cells in 200 µl), left-lung intrathoracic injection (5 × 105 cells in 25 µl under anesthesia with surgical staple closure), or two intranasal doses of 0.5 × 106 cells in 50 µl each with dropwise inhalation.
  5. Engraftment Confirmation: A first IVIS session a few days after inoculation confirms cell take and establishes baseline radiance for each animal.
  6. Longitudinal Monitoring and Treatment: Whole-animal bioluminescence is captured on a scheduled basis after D-luciferin (100–150 mg/kg i.p. or 3 mg s.c.), with body weight recorded in parallel; test agents or immunotherapy combinations are dosed per the study design.
  7. Scheduled Endpoint Harvesting: Animals are typically euthanized three to four weeks after inoculation (or at humane endpoints); lungs are resected, the five lobes are inspected for macroscopic nodules, and major organs are collected for ex vivo imaging.
  8. Histopathology and Immune Profiling: Lung sections are H&E stained for tumor burden quantification (e.g., QuPath image analysis), and tumor-digest flow cytometry profiles the microenvironment (CD8+ T cells, Tregs, macrophages, PD-L1) to complete the dataset.
