Carbon Tetrachloride-Induced Liver Fibrosis Model Service

Fig 1: Carbon Tetrachloride-Induced Liver Fibrosis Model preclinical research.

The carbon tetrachloride-induced liver fibrosis model is the most widely validated toxin-based system for generating rapid, robust, and reproducible hepatic fibrosis in rodents, delivering bridging fibrosis within weeks rather than months. Alfa Cytology runs this classic platform with modern operational discipline, executing scheduled CCl4 dosing, oil-vehicle control management, staged histological readouts, and quantitative fibrosis scoring so that antifibrotic candidates can be benchmarked quickly and credibly.

Overview of Carbon Tetrachloride-Induced Liver Fibrosis Model

Carbon tetrachloride (CCl4) is a colorless, non-polar solvent that earned its place in experimental hepatology because the liver itself converts this otherwise inert compound into a potent toxin. Hepatic cytochrome P450, principally the CYP2E1 isoform concentrated in pericentral zone 3 hepatocytes, reductively dehalogenates CCl4 into the trichloromethyl radical (•CCl3), which in the presence of oxygen yields the even more aggressive trichloromethyl peroxy radical (•CCl3OO•). These species attack polyunsaturated fatty acids in cellular and mitochondrial membranes, unleashing lipid peroxidation that collapses membrane integrity, deranges calcium homeostasis, and kills hepatocytes across the centrilobular region. A single exposure produces acute, self-limiting necrosis that the liver repairs within days, but when dosing is repeated on a weekly rhythm, each wave of death, inflammation, and regeneration layers new extracellular matrix onto the last, and fibrosis accumulates predictably.

Rat CCl4 cirrhosis protocol comparison at 12 weeks: liver-to-body weight ratio (A) and quantitative fibrosis area by Sirius Red (B) and Masson's trichrome (C), plus representative histology panels — H&E (A–C), Sirius Red (D–F), and Masson's trichrome (G–I) for control (A,D,G), once-weekly CCl4 (B,E,H), and twice-weekly CCl4 (C,F,I) groups — showing that only the twice-weekly gavage protocol produces advanced cirrhosis with elevated ALT/AST/ALP/bilirubin, increased INR, decreased albumin, and thrombocytopenia. (Fortea, José I., et al., 2018)

What keeps the CCl4 model in continuous use after more than half a century is the degree of control it hands the investigator. Standard regimens, two or three intraperitoneal injections per week at 0.5–1 mL/kg diluted in vegetable oil, produce significant fibrosis within four weeks, bridging fibrosis around five to seven weeks, and cirrhosis by eight to nine weeks in rats, a pace unmatched by dietary or genetic alternatives. Fibrosis severity tracks dose and duration, strains differ predictably in sensitivity (BALB/c being highly responsive, C57BL/6 intermediate, FVB/N comparatively resistant), and, crucially, fibrosis induced for fewer than about sixty days largely regresses once exposure stops, making the system uniquely suited to regression and resolution studies. Because the pathology springs from hepatocyte injury, the model is also regarded as a faithful analogue of virus-driven chronic hepatitis fibrosis and remains the reference standard against which new antifibrotic mechanisms are tested in vivo.

Principles of Carbon Tetrachloride-Induced Liver Fibrosis Model Construction

Constructing the CCl4 model successfully requires attention to bioactivation geography, dosing cadence, vehicle choice, and strain sensitivity, because each variable measurably shifts the fibrotic output. The essential elements are tabulated below.

Element Agent / Factor Function in Model Construction Representative Conditions
Hepatotoxin Carbon tetrachloride (CCl4), diluted in vegetable oil A non-polar solvent bioactivated by CYP2E1 in pericentral hepatocytes to •CCl3 and •CCl3OO• radicals; radical-driven lipid peroxidation destroys membrane integrity, provoking centrilobular necrosis and the release of damage signals that recruit inflammatory cells. 0.5–1 mL/kg body weight, intraperitoneal, two to three times weekly
Vehicle Corn oil, olive oil, or mineral oil The oil vehicle governs absorption kinetics and injury intensity; vehicle-matched controls are indispensable, and oil selection itself modulates the severity and pace of fibrosis. Typically a 1:3 to 1:4 dilution of CCl4 in oil; vehicle-only control group included
Dosing cadence Repeated injections at fixed intervals Because single-dose injury resolves within days, regular re-dosing sustains the injury–inflammation–regeneration cycles that convert acute necrosis into progressive matrix deposition and bridging fibrosis. Twice-weekly dosing for 4–6 weeks (fibrosis); extended schedules for cirrhosis
Fibrogenic machinery Kupffer cell activation, hepatic stellate cell transdifferentiation, TGF-β1/Smad3 signaling Damage-associated molecular patterns from dying hepatocytes activate Kupffer cells, whose cytokine output transdifferentiates stellate cells into collagen-secreting myofibroblasts; TGF-β1/Smad3 is the dominant profibrogenic axis, with α-SMA and collagen I as canonical readouts. Hydroxyproline content, Sirius red/Masson trichrome area, α-SMA immunostaining
Reversibility & staging Withdrawal-defined regression window Fibrosis induced for under ~60 days regresses substantially after exposure ceases, while longer exposure matures into relatively fixed cirrhosis; interim sacrifice points document the stage progression from pericentral fibrosis to bridging and nodular cirrhosis. Interim necropsies at weeks 2–4 and weeks 5–7; terminal analysis at week 6–9

Our Services

Every CCl4 engagement at Alfa Cytology is executed against written dosing calendars, vehicle-matched control arms, and prespecified interim staging points, so sponsors receive fibrosis data that is as auditable as it is reproducible.

Workflow of Carbon Tetrachloride-Induced Liver Fibrosis Model Construction

The CCl4 model rewards precision: its entire fibrotic output emerges in a compact six- to nine-week window, and small deviations in dose timing translate into visible differences in histology. A standard engagement proceeds as follows.

  1. Strain selection and study configuration: The scientific question sets the species and strain, mouse or rat, with sensitivity-guided choices such as BALB/c for maximal responsiveness or C57BL/6 where genetic tools are required, alongside group sizes and the therapeutic arms to be tested.
  2. Animal acclimatization and baseline sampling: Animals are habituated for at least one week; only animals meeting minimum body-weight criteria (≥20 g for mice) enter the study, and baseline body weights plus optional serum chemistry are recorded.
  3. Randomization and vehicle control setup: Cohorts are randomized by weight, and a vehicle-only control group receiving oil injections alone is established to isolate the specific effect of CCl4.
  4. Dosing solution preparation: CCl4 is freshly diluted in the chosen oil at the protocol ratio immediately before each session, and prepared volumes are used promptly to prevent evaporation-driven concentration drift.
  5. Scheduled CCl: 4 administration and monitoring. Animals receive intraperitoneal injections on the fixed weekly calendar, typically twice weekly, with body weight, activity, and injection-site condition checked after every dose and dosing adjusted against predefined tolerance criteria.
  6. Test article administration (therapeutic studies): Candidate antifibrotics or vehicle are dosed on the sponsor-defined schedule, commonly on alternating days relative to CCl4, so that treatment and toxin exposures never coincide.
  7. Interim staging necropsies: At designated milestones, subset animals are euthanized, and livers are harvested for early histology, hydroxyproline measurement, and oxidative stress indices such as MDA and GSH, capturing the fibrosis timeline as it unfolds.
  8. Terminal endpoint analysis: At the final time point, livers are weighed, sectioned, and processed for H&E necrosis scoring, Sirius red or Masson trichrome fibrosis quantification, α-SMA and collagen I immunohistochemistry, TGF-β1/Smad3 pathway analysis, and full serum ALT/AST panels.
