A498 / A498-luc Orthotopic Mouse Model Service for Renal Cancer

Fig 1: A498 / A498-luc Orthotopic Mouse Model for Renal Cancer preclinical research.

The A498 / A498-luc orthotopic mouse model implants a VHL-deficient human clear cell renal carcinoma line directly beneath the kidney capsule, reproducing primary renal tumor growth and its propensity for distant spread in a physiologically authentic setting. Alfa Cytology delivers this model as a turnkey preclinical service — authenticated cell sourcing, precision subcapsular implantation surgery, serial bioluminescence tracking, dosed intervention, and comprehensive terminal pathology — giving kidney cancer programs a translationally faithful platform without the burden of specialized in-house surgical infrastructure.

Overview of A498 / A498-luc Orthotopic Mouse Model for Renal Cancer

A498 is one of the founding human renal cell carcinoma cell lines, established in the 1970s from the kidney of a 52-year-old Caucasian male with renal carcinoma, and later distributed worldwide as ATCC HTB-44. Among the first RCC lines assembled for the US National Cancer Institute's 60-cell-line anticancer drug screen, it remains a workhorse of kidney cancer research. The line exhibits epithelial morphology, an adherent monolayer growth pattern, and a bimodal, tetraploid karyotype. Genomically, A498 carries a frameshift mutation that abolishes von Hippel-Lindau (VHL) protein expression — the hallmark lesion of clear cell renal cell carcinoma — together with SETD2 and MLL3 mutations, while PBRM1 and NF2 remain intact; transcriptomically it clusters with the more aggressive ccB subtype. A-498-derived xenografts recapitulate classic ccRCC histology, forming nests of malignant cells with clear cytoplasm, and the line's well-documented upregulation of mTOR signaling and its responsiveness to rapamycin made it an early model for targeted therapy studies in RCC.

Orthotopic implantation of A498 beneath the renal capsule of immunodeficient mice converts this familiar cell line into a substantially more instructive research tool. Compared with flank xenografts, subcapsular tumors grow within the true renal microenvironment with its organ-specific vasculature, permitting studies of tumor–parenchyma interactions, local invasion, and spontaneous metastasis to lung, liver, and other organs. Luciferase-labeled A498 (A498-luc) enables quantitative, noninvasive bioluminescence tracking of engraftment, tumor expansion, and metastatic dissemination on IVIS-class imagers — a capability that has made the orthotopic A498 platform a standard choice for validating oncogenic drivers, testing targeted agents and kinase inhibitors, and dissecting resistance mechanisms in clear cell renal cancer research.

Cell Line Information: A498 / A498-luc

A498 (ATCC HTB-44) is a human renal cell carcinoma line of clear cell subtype, established from a primary renal tumor and maintained by the American Type Culture Collection. The luciferase-tagged derivative, A498-luc, is generated by stable transduction with a firefly luciferase reporter and retains the parental tumorigenic and metastatic behavior. Principal characteristics are summarized below.

Workflow of A498 / A498-luc Orthotopic Mouse Model Construction

Orthotopic renal modeling combines exacting cell quality control, left-kidney subcapsular microsurgery, and longitudinal optical readouts. A standard construction proceeds through the following stages.

  1. Cell preparation and quality control: A498 or A498-luc cells are expanded under authenticated culture, verified mycoplasma-free and STR-confirmed, and luciferase activity is validated in vitro before implantation.
  2. Host selection and acclimation. Immunodeficient mice: athymic nude or NOD/SCID mice around 5–6 weeks of age, and humanized NSG strains where immune-oncology questions require them — are quarantined and acclimated in a barrier facility.
  3. Anesthesia and exposure of the left kidney: Under isoflurane anesthesia, a left flank or dorsal incision is made, the peritoneum is opened, and the left kidney is gently exteriorized onto a sterile drape for subcapsular access.
  4. Subcapsular inoculation. A single-cell suspension: on the order of 105–106 cells in a small volume, frequently mixed 1:1 with Matrigel to support take rates — is injected beneath the renal capsule through a fine needle, producing a visible subcapsular bleb while sparing the renal parenchyma.
  5. Repositioning and closure: The kidney is returned to the abdominal cavity, the peritoneum and musculature are sutured, the skin is closed, and analgesia is administered with close postoperative monitoring.
  6. Engraftment verification and serial imaging: Bioluminescence imaging is performed after intraperitoneal D-luciferin injection to confirm delivery and take, then on a weekly schedule to track primary tumor growth and detect distant metastases in individual animals.
  7. Randomization and treatment: Mice are assigned to arms on the basis of tumor signal, and test articles, targeted agents, or combination regimens are dosed according to protocol with body weight and clinical status recorded at defined intervals.
  8. Endpoint analysis and reporting: At termination, primary kidney tumors and metastatic organs are resected and weighed; tissues undergo H&E and immunohistochemical staining (including VHL/HIF-pathway markers), metastatic burden is quantified, and all data are consolidated into the final report.
