SJSA-1 / SJSA-1-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: SJSA-1 / SJSA-1-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The SJSA-1 / SJSA-1-luc orthotopic mouse model for sarcoma is a human osteosarcoma xenograft platform built on a genetically distinctive foundation — MDM2 amplification with wild-type TP53 — in which tumor cells are grafted into the bone of immunodeficient mice to enable p53-reactivation therapy, resistance-emergence, and bone-lesion studies. Alfa Cytology delivers this model as a complete preclinical capability — spanning authenticated SJSA-1 / SJSA-1-luc expansion and luciferase-stable banking, precision intraosseous implantation, scheduled in vivo bioluminescence tracking of primary bone tumors, and complete skeletal histopathology with pharmacodynamic p53-pathway readouts — all configured around each sponsor’s mechanism of action and endpoint plan.

Overview of SJSA-1 / SJSA-1-luc Orthotopic Mouse Model for Sarcoma

SJSA-1 occupies a special position among osteosarcoma models because of a single defining abnormality: the MDM2 gene is amplified roughly 45-fold, flooding the cell with the E3 ubiquitin ligase that tags p53 for degradation, while TP53 itself remains wild-type. Established in 1982 from the primary tumor of an 18-year-old male with osteosarcoma, the line therefore represents the classic clinical scenario in which p53 is functionally silenced not by mutation but by overexpression of its negative regulator — a phenotype estimated to occur in a substantial fraction of sarcomas. This genotype makes SJSA-1 the reference system for p53-reactivation pharmacology: among a panel of ten cancer cell lines it showed the strongest apoptotic response to MDM2 antagonists, precisely because MDM2 amplification appears to be the principal defect in its p53 pathway. The line additionally carries an NRAS Q61K mutation, making it a useful substrate for MAPK-pathway studies, and it grows well in both culture and immunodeficient hosts.

In animals, SJSA-1 has produced some of the most dramatic proof-of-concept results in the MDM2-inhibitor field. Early work showed that nutlin-3 at 200 mg/kg twice daily for 20 days inhibited tumor growth in nude mice without apparent systemic toxicity, while optimized antagonists went further: RG7112 produced dose-dependent tumor regression, RG7388 at 25 mg/kg oral daily achieved outright tumor regression, and SAR405838 induced rapid, complete, and durable tumor regression — then, tellingly, treated tumors that eventually regrew yielded sublines that have become the standard reagents for studying how resistance emerges under p53-pathway pressure, both in vitro and in vivo. Locating this biology in the bone amplifies its translational relevance: orthotopic bioluminescent osteosarcoma xenograft methodology is established in Swiss nude and NSG mice, and the luciferase-labeled derivative SJSA-1-luc allows tumor take and growth in the operated limb to be tracked longitudinally by in vivo imaging, with micro-CT and histopathology confirming bone involvement. The result is a platform where p53-reactivation agents, MAPK-pathway inhibitors, and combination regimens can be evaluated with both imaging readouts and mechanism-resolving pharmacodynamic endpoints.

Cell Line Information: SJSA-1 / SJSA-1-luc

The table below consolidates the key biological, cultural, and modeling characteristics of the SJSA-1 parental line and its luciferase-labeled derivative SJSA-1-luc. Both are human-derived and require immunodeficient hosts for in vivo work.

Parameter Details
Cell line designation SJSA-1 (parental line; also known as OSA); SJSA-1-luc (luciferase-labeled derivative)
Species / donor Human (Homo sapiens), male; established in 1982 from the primary tumor of an 18-year-old donor with osteosarcoma
Depository / catalogue ATCC CRL-2098; RRID CVCL_1697
Tumor type Human osteosarcoma
Defining genotype MDM2 amplification at approximately 45-fold copy number with correspondingly high MDM2 protein, coupled to wild-type TP53 — the signature MDM2-amplified, p53-intact phenotype
Additional mutations NRAS Q61K mutation, making the line a tractable model for MAPK-pathway inhibitor studies (e.g., trametinib); SJSA-1 cells respond to such inhibitors with growth arrest but minimal apoptosis due to persistent p53 suppression
Pharmacological responsiveness Strongest apoptotic response to MDM2 antagonists among a ten-line cancer panel; growth-inhibited by nutlin-3, RG7112, RG7388, SAR405838, milademetan (IC50 < 100 nmol/L), and related compounds; treatment-selected resistant sublines (p53 mutations such as R273C, L130R/C277F, C176F) are documented reagents
Growth properties Adherent cells with robust, aggressive proliferation; amenable to transfection and selection workflows used to generate labeled derivatives
Recommended culture conditions RPMI-1640 supplemented with 10% fetal bovine serum at 37 °C in a humidified 5% CO₂ atmosphere
Reporter labeling (SJSA-1-luc) Stable firefly luciferase expression introduced by lentiviral transduction; enables in vitro reporter assays and longitudinal in vivo bioluminescence imaging of primary bone tumors
Standard orthotopic inoculum Established orthotopic bioluminescent osteosarcoma methodology in Swiss nude and NSG mice implants approximately 1 × 106 cells in 10–20 μL of PBS or a Matrigel-supported suspension into the proximal tibial medullary cavity (subcutaneous reference dosing is 1–5 × 106 cells per site)
Host strains Athymic nude (Swiss nude), SCID, and NSG mice
Tumor behavior Subcutaneous xenografts respond to MDM2 antagonists with growth inhibition through complete regression; post-treatment regrowth provides material for resistance mechanism studies; bone-orthotopic grafting places this pharmacology in a skeletal microenvironment
Key research applications MDM2-p53 interaction inhibitor efficacy and pharmacodynamic studies; acquired-resistance mechanism research under p53-pathway pressure; MAPK-pathway inhibitor evaluation in an NRAS-mutant background; combination and sequencing studies (MDM2 inhibitors with chemotherapy or targeted agents); bone-lesion biology and osteosarcoma drug discovery

Our Services

Few models reward mechanistic rigor the way MDM2-amplified osteosarcoma does, and Alfa Cytology runs the SJSA-1 / SJSA-1-luc orthotopic program accordingly: authenticated cell banks, standardized intraosseous implantation, weekly in vivo bioluminescence with micro-CT correlation, and endpoint pathology paired with p53-pathway pharmacodynamics — all under one accountable preclinical operation.

Workflow of SJSA-1 / SJSA-1-luc Orthotopic Mouse Model Construction

Construction follows established intraosseous bioluminescent xenograft methodology as validated in Swiss nude and NSG mice: luciferase-labeled SJSA-1 cells are expanded under authenticated conditions, concentrated into a small-volume suspension, injected into the proximal tibial medullary cavity, and followed by serial imaging with bone-focused, mechanism-resolving endpoint analysis. The workflow comprises the following stages.

  1. Cell authentication and banking: SJSA-1 or SJSA-1-luc cells are verified by STR profiling, confirmed mycoplasma-free, and expanded into low-passage working banks under standardized RPMI-1640 culture conditions.
  2. Cell harvest and formulation: Exponentially growing cultures are trypsinized, washed, counted and viability-checked, then resuspended at approximately 1 × 106 cells in 10–20 μL of PBS or a Matrigel-supported suspension and kept on ice until implantation.
  3. Anesthesia and tibial access: Male immunodeficient mice are anesthetized with isoflurane, the proximal tibia is exposed through a small skin incision, and a fine-gauge needle enters the medullary cavity at the tibial plateau.
  4. Orthotopic implantation: The cell suspension is slowly injected into the proximal tibial cavity; the needle is held briefly after injection to limit reflux along the cortical tract and standardize take rates.
  5. Closure and perioperative care: The puncture site is sealed, the skin is closed, and animals receive analgesia with close post-operative monitoring, including gait observation for lameness and limb swelling.
  6. Engraftment confirmation and serial imaging: Luciferin is administered intraperitoneally and bioluminescence is acquired to confirm tibial tumor take and normalize group allocation; imaging repeats on a weekly cadence to track primary lesion growth and response.
  7. Intervention and endpoint analysis: Experimental arms — MDM2 antagonists, MAPK-pathway inhibitors, combination chemotherapy, and sequencing regimens — proceed per study design; endpoints integrate imaging with radiography and micro-CT (bone architecture), tumor-bearing limb dissection and weighing, and histopathology (H&E with Ki67, TRAP, p53/MDM2/p21 pharmacodynamic staining, and marker-specific immunohistochemistry).
