Inquiry

Lentiviral vectors combine stable genomic integration capacity with broad host cell tropism, making them ideal tools for achieving long-term transgene expression and establishing stable cell line models. As a preclinical CRO dedicated to the oncology field, Alfa Cytology provides high-quality lentivirus packaging services, committed to delivering efficient and stable gene delivery solutions for your research projects.

Introduction to Lentivirus Packaging

Lentivirus, a member of the Retroviridae family, has emerged as one of the most widely adopted gene delivery vehicles in cancer research, owing to its unique ability to infect both dividing and non-dividing cells and to stably integrate the transgene into the host genome. The lentiviral genome has been engineered into multiple separate plasmids, which preserves its potent transduction capacity while fundamentally eliminating the risk of autonomous viral replication. Lentiviral vector systems typically employ a three- or four-plasmid co-transfection strategy in packaging cells (e.g., HEK293T cells), wherein the gene of interest, packaging elements (gag/pol, rev), and envelope glycoprotein (commonly VSV-G) are distributed across distinct plasmids. This design substantially minimizes recombination events while enabling efficient and safe virus production. Through optimization of critical process parameters, including plasmid ratios, transfection reagents, and harvest timing, high-titer and high-purity viral stocks can be consistently obtained, meeting diverse requirements ranging from in vitro cellular assays to in vivo animal models. In oncology research, lentiviral vectors are extensively applied in gene overexpression, shRNA-mediated gene knockdown, CRISPR/Cas9-based gene editing, and genome-wide functional screening. Cells transduced with lentivirus achieve long-term, stable expression of the target gene or regulatory elements, providing an ideal tool for establishing stable cell lines, developing CDX models, and conducting in vivo engraftment studies.

Applications of Lentiviral Vectors in Cancer Research

Stable Cell Line Generation

Lentiviral vectors carrying the gene of interest or shRNA are transduced into target cells, followed by antibiotic selection (Puromycin, Blasticidin, Neomycin, etc.) to establish cell lines with stable overexpression or knockdown, providing a reliable platform for long-term in vitro functional studies.

CRISPR/Cas9 Gene Editing

Lentiviral vectors such as lentiCRISPR are used to deliver Cas9 and sgRNA for gene knockout or knock-in, enabling stable genomic modifications suitable for long-term gene function studies.

Genome-Wide Functional Screening

Lentiviral CRISPR knockout or shRNA libraries enable genome-wide functional screening to systematically identify tumor dependency genes, resistance-associated genes, and synthetic lethal targets, providing an unbiased tool for target discovery.

In Vivo Tumor Models

Tumor cell lines stably expressing luciferase reporters are established via lentiviral transduction and implanted into immunodeficient mice, enabling long-term real-time monitoring of tumor growth, metastasis, and therapeutic response through bioluminescence imaging.

In Vivo Gene Delivery

Lentivirus is directly injected into tumor tissues or specific anatomical sites to achieve localized expression modulation of target genes or RNAi elements in situ, supporting site-specific gene function studies.

Stem Cell Genetic Modification

Lentiviral-mediated stable genetic modification (overexpression, knockout, or knockdown) is achieved in stem cells such as iPSCs or MSCs, supporting differentiation studies, regenerative medicine, and cell therapy product development.
