Introduction to Adeno-Associated Virus (AAV) Packaging
Adeno-Associated Virus (AAV) is currently one of the most widely used viral vectors in gene therapy and in vivo gene delivery. Belonging to the Parvoviridae family, AAV offers outstanding advantages including non-pathogenicity, low immunogenicity, non-integrating nature, and diverse tissue tropisms, establishing it as the gold standard for targeted in vivo gene delivery. The replication and packaging of AAV are strictly dependent on helper functions provided by co-infecting helper viruses, such as adenovirus or herpes simplex virus, a property that facilitates the production of high-purity viral preparations by effectively eliminating helper virus contamination during the manufacturing process. To date, over 12 naturally occurring AAV serotypes (AAV1–AAV12) and numerous engineered variants have been discovered or developed, each exhibiting distinct tissue tropisms—for instance, AAV9 can efficiently cross the blood–brain barrier to target the central nervous system, AAV8 displays high hepatic affinity, whereas AAV6 preferentially transduces muscle tissues. Depending on the specific tissue/organ targeting requirements, researchers can select AAV serotypes with tailored tropisms through systematic serotype screening and capsid engineering, thereby achieving efficient and precise gene delivery. By optimizing plasmid ratios in the triple-plasmid transfection system, cell culture conditions, and purification processes, robust production of high-titer, high-purity AAV preparations can be consistently attained, accommodating diverse in vivo application scenarios ranging from local injection to systemic administration.

Virus Packaging and Production
Under a GMP-like quality control system, we employ a triple-plasmid co-transfection system for AAV packaging and production:
- High-quality endotoxin-free maxiprep of plasmid DNA.
- PEI-mediated co-transfection of HEK293T/HEK293 cells.
- Serum-free culture system.
- Optimized harvest and purification processes to maximize infectious particle recovery.
Virus Purification and Concentration
We utilize optimized purification procedures to ensure high purity and high infectivity of AAV:
- Iodixanol density gradient ultracentrifugation or CsCl density gradient centrifugation.
- Ion-exchange chromatography for further purification (if required).
- Ultrafiltration concentration to the desired volume.
- Effective removal of empty capsids to enhance transduction efficiency.
Quality Control Testing
Each batch of AAV is subjected to comprehensive quality control testing:
- Physical titer: qPCR for genome titer (vg/mL).
- Infectious titer: Fluorescence-based assay for infectious units (IU/mL) or TCID50 method.
- Purity assessment: SDS-PAGE silver staining analysis and HPLC detection.
- Empty capsid rate: Electron microscopy or SEC-HPLC for assessment of full-to-empty particle ratio.

Applications of AAV Vectors in Cancer Research

In Vivo Gene Function Studies
Tissue-specific AAV serotypes (e.g., AAV9 for brain, AAV8 for liver, AAV2 for retina) are used to deliver overexpression or knockdown constructs, enabling precise study of gene function in specific tissues while avoiding physiological interference from systemic gene modulation.

Gene Therapy Candidate Development
AAV is the most widely used vector platform in gene therapy, with multiple AAV-based gene therapies already approved. AAV vectors are suitable for delivering therapeutic genes, RNAi, or CRISPR/Cas components to support gene replacement, gene silencing, and gene repair strategies.

Tumor Microenvironment Remodeling
Cytokines (e.g., IL-12, IFN-β), chemokines, or immunomodulatory molecules (e.g., anti-PD-1 antibodies, TGF-β traps) are delivered via AAV to locally remodel the tumor immune microenvironment, converting "cold" tumors to "hot" and enhancing immunotherapy efficacy.

Immunotherapy Research
AAV delivers tumor antigens or immune checkpoint modulators (e.g., PD-L1 shRNA, CTLA-4 antibodies) for sustained in vivo activation of antitumor immune responses, supporting the development and validation of novel immunotherapy strategies.

Genetically Engineered Mouse Models (GEMM)
AAV is used to deliver Cre recombinase to specific tissues in GEMMs, enabling conditional gene knockout or activation without the need for complex breeding strategies, significantly shortening model generation timelines.

In Vivo Imaging Tracking
AAV carrying luciferase reporter genes enables non-invasive in vivo imaging to track tumor progression, metastasis, and therapeutic response in real time.
