Introduction to Adenovirus Packaging

Adenovirus is a non-enveloped, double-stranded DNA virus widely used in cancer research and gene therapy due to its high transduction efficiency, broad host range, and non-integrating nature. The virus enters cells via fiber protein–receptor (e.g., CAR) binding and receptor-mediated endocytosis, with its genome remaining episomal in the nucleus, enabling robust transient transgene expression without insertional mutagenesis risk. Adenoviral vectors accommodate large inserts (8–10 kb) and are readily propagated to high titers in HEK293 cells and derivatives. Most current systems are E1/E3-deleted, replication-defective Ad5-based vectors with reduced immunogenicity and cytotoxicity. Optimized MOI, harvest timing, and purification methods (e.g., CsCl gradients or chromatography) yield highly active, low-impurity preparations. In oncology, these vectors are broadly applied in transient overexpression, pathway analysis, recombinant protein production, and tumor vaccine development, being particularly advantageous for scenarios requiring rapid, high-level gene expression, such as oncogene validation, apoptosis assessment, and transient delivery of immunostimulatory molecules.

Quality Control Testing

Each batch of adenovirus undergoes comprehensive quality control testing:

  • Titer determination: Infectious titer (PFU/mL) measured by TCID50 or plaque assay; physical titer measured by qPCR.
  • Purity assessment: SDS-PAGE analysis and A260/A280 ratio determination.
  • Replication competence testing: Detection of replication-competent adenovirus (RCA) content.
  • Sterility testing: Bacterial, fungal, and mycoplasma detection.
  • Activity validation: Verification of transgene expression in target cells.

Applications of Adenoviral Vectors in Cancer Research

Transient Protein Overexpression

Adenoviral infection achieves high-level transient expression of target genes in host cells, suitable for protein function studies, protein-protein interaction analysis, and recombinant protein production.

Signaling Pathway Modulation

Constitutively active or dominant-negative mutants are delivered via adenovirus to specifically activate or inhibit key nodes in signaling pathways, enabling dissection of upstream activation mechanisms and downstream effector networks.

Transient Gene Function Studies

Adenovirus enables rapid and reversible assessment of gene function without permanent genomic alterations, making it suitable for short-term functional screening in primary cells and stem cells.

Tumor Vaccine Development

Adenovirus serves as a vaccine vector for efficient delivery of tumor-associated antigens or neoantigens, activating specific antitumor T-cell immune responses to support early-stage therapeutic vaccine development.

Recombinant Protein Production

Leveraging the high-efficiency protein expression capacity of adenovirus, recombinant proteins and virus-like particles can be rapidly produced in mammalian cells for antigen preparation, antibody screening, and structural biology studies.

Conditional Gene Expression

In combination with the Cre-LoxP system, adenovirus enables conditional gene regulation in specific cell or tissue types, supporting cell lineage tracing and spatially restricted gene function studies.

Service Highlights

  • High titer: Achieves up to 1010–1011 PFU/mL, ensuring high-efficiency infection.
  • Large packaging capacity: Supports insertion of exogenous genes up to 8–10 kb.
