Precise point mutation introduction is a key technical approach for studying gene function, dissecting disease mechanisms, and validating therapeutic targets. Alfa Cytology offers point mutation services based on CRISPR-mediated editing and Prime Editing technologies to enable precise single-nucleotide alterations in target genes. Leveraging our optimized editing strategies and rigorous clonal selection workflows, we deliver high-quality point mutation models to support a broad range of applications—from functional genomics to disease research.
Point mutations, single-nucleotide substitutions, are central molecular events driving a wide spectrum of genetic disorders and cancers. The precise introduction of specific point mutations enables researchers to model disease-associated genetic variants and dissect their impact on protein function and cellular behavior. Conventional approaches, such as CRISPR/Cas9 combined with ssODN donor templates via the homology-directed repair (HDR) pathway, have been widely adopted for point mutation introduction. However, HDR-mediated editing is often constrained by low efficiency, dependence on the cell cycle, and the requirement for donor templates. To overcome these limitations, the next-generation Prime Editing technology has emerged as a transformative tool. Prime Editing enables all 12 types of base-to-base conversions as well as small insertions and deletions, without requiring DNA double-strand breaks or donor templates. This unique mechanism significantly reduces the risk of unintended editing outcomes and offers distinct advantages for modeling complex mutations that are challenging to generate using conventional methods. By providing greater precision and flexibility, Prime Editing is expanding the possibilities for disease modeling, functional genomics, and therapeutic development.

HDR-mediated point mutation is a reliable strategy for introducing standard point mutations and generating disease-related mouse models. Prime Editing is particularly suited for complex mutation scenarios where HDR efficiency is limited, especially for small-fragment insertions, deletions, and precise edits that are difficult to achieve through HDR.
