CRISPR/Cas9 Gene Editing Platform
Inquiry
As a preclinical CRO deeply rooted in the oncology field, Alfa Cytology has established an efficient and reliable CRISPR/Cas9 gene-editing technology platform that provides core tools for cancer gene functional studies and drug target validation. The platform integrates advanced guide RNA design algorithms, flexible delivery systems (plasmid, lentivirus, AAV, and RNP), and an end-to-end quality control framework, supporting a broad range of applications from gene knockout, knock-in, and point mutation to transcriptional regulation.
Introduction to CRISPRCas9 Gene Editing Technology
The CRISPR/Cas9 system has revolutionized biomedical research by enabling precise, targeted modifications to the genome with unprecedented simplicity, efficiency, and scalability. The system consists of two essential components: the Cas9 nuclease, which functions as molecular scissors to introduce double-strand breaks (DSBs) at specific genomic loci, and a single guide RNA (sgRNA), which directs Cas9 to its target site through sequence complementarity. Cleavage occurs only in the presence of a protospacer-adjacent motif (PAM), a short consensus sequence (e.g., NGG for the widely used Streptococcus pyogenes Cas9) located immediately downstream of the target sequence. In cancer research, CRISPR technologies facilitate the generation of isogenic cell lines, the establishment of disease-relevant animal models, and the implementation of genome-wide functional screens, accelerating the drug discovery continuum from target identification through preclinical validation. The continued evolution of gene editing technologies—from CRISPR/Cas9 to base editing, prime editing, CRISPRi/a, and RNA-targeting systems—continually expands the application landscape from single-nucleotide correction to transcriptional regulation, providing increasingly powerful tools for functional genomics and translational research.

Leveraging our deep expertise in oncology and rigorous project management capabilities, we deliver end-to-end technical support for your drug development programs, from target discovery to IND filing, ensuring precision, robustness, and reliability at every step.
Based on the CRISPR/Cas9 system, our platform enables complete and stable disruption of target genes. We support a wide range of cell types, including conventional cell lines, primary cells, and stem cells, as well as mouse models. Through rigorous monoclonal selection strategies and dual validation at both the genomic and protein levels, we deliver homozygous, stable knockout models. This service is ideally suited for gene function analysis, disease model generation, and drug target validation.
Leveraging our proprietary HDR enhancement technology, we achieve precise integration of exogenous sequences at target gene loci. Applications include oncogenic mutation introduction, fluorescent/luciferase tagging, cell-based reporter system development, and mouse model knock-in generation. Our platform supports both cell line and mouse model applications, offering flexibility across diverse research needs.
Utilizing a dual-platform approach based on CRISPR-mediated editing and proprietary Prime Editing technology, we enable precise single-nucleotide alterations in target genes. Our system supports all 12 types of base conversions as well as small insertions or deletions, without the requirement for DNA double-strand breaks or donor templates. This offers distinct advantages in modeling complex mutations that are otherwise difficult to generate with conventional methods.
We provide custom CRISPR library design and construction services to support whole-genome knockout screening, pathway-focused studies, and targeted validation. Our offerings cover CRISPR knockout, CRISPRi, and CRISPRa applications, with production scales flexibly adaptable from small customized libraries to genome-wide libraries. Rigorous quality control ensures high representation, full coverage, and low bias across all library deliverables.
Core Capabilities of Our CRISPR/Cas9 Gene-Editing Platform
Advanced guide RNA Design
Our platform integrates specialized guide RNA design algorithms that comprehensively evaluate editing efficiency, targeting specificity, and off-target risks. For each target gene, multiple candidate sgRNA sequences are systematically assessed using advanced scoring models, such as CFD score, MIT score, and others, to predict editing activity and potential off-target sites, thereby identifying the optimal editing strategy. For off-target risk assessment, genome-wide unbiased detection methods such as GUIDE-seq are employed as the most rigorous framework for comprehensive identification and profiling of off-target effects.
Diverse Delivery Systems
Our platform supports a broad range of delivery systems, flexibly accommodating diverse cell types and application scenarios:
- Plasmid delivery: Suitable for conventional cell lines, enabling stable gene expression and long-term editing.
- Lentivirus delivery: Ideal for hard-to-transfect cells, achieving efficient and stable gene integration.
- AAV delivery: Applicable for primary cells and in vivo applications, offering low immunogenicity and targeted specificity.
- RNP delivery: Pre-assembled Cas9 protein–sgRNA complexes enable rapid editing with significantly reduced off-target effects, particularly suited for primary and sensitive cell lines
Rigorous Quality Control
Our platform incorporates an end-to-end quality control framework that covers the entire workflow, from sgRNA design validation to final edited clone identification:
- sgRNA sequence accuracy and effectiveness validation
- Delivery efficiency assessment
- Editing efficiency detection (T7EI assay, Sanger sequencing)
- Off-target effect analysis (GUIDE-seq and other orthogonal methods)
- Dual validation at both genomic and protein levels
Applications of CRISPR/Cas9 Gene Editing Technology
Target Discovery and Validation
CRISPR-based whole-genome screening enables systematic identification of genes associated with specific phenotypes. Combined with synthetic lethality screening strategies, this approach provides an unbiased functional genomics tool for target discovery. The functional relevance of candidate targets can be further validated through gene knockout or knock-in, delivering functional evidence to support drug development programs.
Disease Model Generation
By introducing disease-relevant mutations or genetic modifications, we generate cellular or animal models of human diseases, providing a robust platform for mechanistic studies and drug screening. Our capabilities support oncogenic mutation introduction, tumor suppressor gene knockout, and patient-specific mutation modeling to closely recapitulate disease pathology.
Mechanisms of Drug Resistance
Through CRISPR-based screening, we identify genes associated with drug resistance and further validate resistance mechanisms using gene-edited models. This approach uncovers novel escape mechanisms underlying acquired drug resistance in tumors, including those driven by genetic mutations or protein translocation.
