Introduction to In Vitro Gene Function Studies
In vitro gene function studies are a fundamental component of cancer biology and drug discovery. Through systematic gain-of-function and loss-of-function manipulations of target genes in a controlled cellular environment, these studies directly evaluate the effects of genes on tumor cell phenotypes, including proliferation, apoptosis, migration, invasion, drug resistance, metabolic reprogramming, and immune evasion. By excluding the confounding influences of complex systemic factors, these investigations enable precise dissection of the intrinsic functions of genes within tumor cells, providing direct evidence for understanding cell-autonomous roles of genes. Through the combined application of diverse gene modulation tools, in vitro studies can establish a comprehensive functional profile of gene regulation, systematically revealing the contributions of genes across multiple dimensions of tumor initiation and progression. Compared with in vivo studies, in vitro experiments offer significant advantages, including shorter turnaround times, higher throughput, lower costs, and clearer mechanistic resolution, making them the platform of choice for target screening, candidate drug evaluation, and signaling pathway investigation. The results obtained from in vitro studies provide critical evidence to guide further in vivo validation and clinical translation, serving as an indispensable starting point in the comprehensive chain of evidence for gene function research.

Our in vitro gene function study services cover the entire workflow from gene modulation tool construction to phenotypic detection and mechanistic dissection. We support a wide range of cell models, including conventional cell lines, primary cells, patient-derived cells (PDC), and patient-derived organoids (PDO), delivering high-quality, quantifiable functional data to support your target discovery, mechanistic elucidation, and pharmacodynamic evaluation.
Available Gene Modulation Strategies
Our in vitro gene function studies achieve activation or suppression of gene function through various strategies. Commonly employed approaches include:
Overexpression
The gene of interest is cloned into expression vectors (lentiviral/plasmid) and transduced into cells to achieve stable or transient high-level expression of the target gene.
CRISPR/Cas9 Knockout
CRISPR/Cas9 technology is utilized to achieve complete knockout of the target gene, suitable for studying the effects of complete gene loss on cellular phenotypes.
RNAi Knockdown
shRNA is employed to mediate post-transcriptional silencing of the target gene, suitable for partial knockdown scenarios and essential gene studies.
Point Mutation Introduction
Site-directed mutagenesis is used to introduce specific mutations, enabling the modeling of clinically relevant mutations or the study of key amino acid residue functions.
Epitope Tag Knock-in
CRISPR/Cas9-mediated knock-in of fluorescent or affinity tags into the target gene for real-time tracking of endogenous protein dynamics, immunoprecipitation, and interaction studies while preserving native regulatory elements.
CRISPRa/i Transcriptional Modulation
dCas9-VP64/KRAB system for targeted transcriptional activation or interference without genomic alteration, suitable for gene dosage, essential gene, and reversible regulation studies.

Applications of In Vitro Gene Function Studies in Drug Discovery

Target Discovery
Systematically identify tumor dependency genes at the cellular level. Employ genome-wide knockout screens to pinpoint genes critical for tumor proliferation and survival, thereby uncovering novel therapeutic targets.

Target Validation
Conduct systematic gain- and loss-of-function studies in a broad panel of tumor cell lines to validate the functional relevance of candidate genes and their driving roles in tumor biological behaviors.

Mechanistic Studies
Elucidate the downstream signaling networks and key effector molecules regulated by the target. Employ molecular and biochemical approaches to establish causal relationships between the target and tumor phenotypes, providing mechanistic rationale for targeted therapeutic strategies.

Drug Sensitivity Assessment
Evaluate the potential of the target as a therapeutic intervention node following gene modulation. Assess changes in drug sensitivity to validate target druggability and identify promising combination therapy regimens.

Resistance Mechanisms
Systematically investigate the functions of resistance-associated genes in drug-resistant cell models. Dissect alterations in gene regulatory networks driving resistance, identify key resistance drivers, and explore potential strategies to overcome resistance.

Combination Therapy Strategy Optimization
Evaluate the effects of various drug combinations on the survival, proliferation, and apoptosis of gene-modulated tumor cells, we identify combination regimens with synergistic efficacy, providing in vitro data to support subsequent in vivo combination validation and clinical translation.
