Introduction to CRISPR/Cas Gene Editing Vector
The CRISPR/Cas system is currently the most powerful genome editing tool, utilizing a guide RNA (sgRNA) to direct Cas proteins to induce double-strand breaks (DSBs) at specific genomic loci, which are then repaired by endogenous non-homologous end joining (NHEJ) or homology-directed repair (HDR) pathways to achieve gene knockout, knock-in, or point mutation. The NHEJ pathway predominantly results in gene knockout via the introduction of frameshift mutations, whereas the HDR pathway enables precise knock-in or point mutation when a donor template is provided. Furthermore, by employing catalytically dead Cas9 (dCas9) fused to transcriptional activation or repression effector domains, CRISPR technology also enables transcriptional activation (CRISPRa) or interference (CRISPRi), offering reversible regulatory tools for gene function studies, particularly suited for essential gene functional analysis and the dissection of complex gene regulatory networks. In oncology research, CRISPR/Cas vectors are widely utilized in applications such as functional genetic screening, tumor driver gene validation, drug resistance model construction, and synthetic lethal target discovery. Depending on the research objectives, the CRISPR/Cas system can be employed to generate permanent gene knockout cell lines (via NHEJ-mediated frameshift mutations), precise gene knock-in models (via HDR-mediated site-specific integration), or reversible transcriptional regulation of endogenous genes through CRISPRi/a.

Applications of CRISPR/Cas Vectors in Cancer Research

Whole-Genome CRISPR Screening
Using lentiviral CRISPR library vectors for genome-wide functional screening to systematically identify tumor dependency genes and synthetic lethal targets. The construction and delivery of large-scale libraries are highly dependent on the lentiviral vector system.

Functional Validation of Gene Knockout
Employing SpCas9 or SaCas9 vector systems to knock out candidate genes in tumor cell lines, systematically assessing their impact on phenotypes such as proliferation, migration, invasion, and evasion of apoptosis. The vector determines knockout efficiency and cellular fitness.

Functional Study of Point Mutations
Utilizing vectors carrying Cas9 and sgRNA, in combination with HDR repair templates, to introduce clinically relevant point mutations and evaluate their effects on protein function and cellular phenotypes. This process relies on vector delivery efficiency and the design of HDR donor elements.

Transcriptional Regulatory Network Studies
Using dCas9 vectors fused with transcriptional activation or repression effector domains (CRISPRa/i) to achieve reversible transcriptional regulation of endogenous genes, thereby dissecting gene regulatory networks and compensatory mechanisms within signaling pathways.

Establishment of Drug-Resistant Models
Employing lentiviral CRISPR vectors to knock out or knock in drug-resistance-associated genes in tumor cells, establishing drug-resistant cell models with well-defined genetic backgrounds. These models provide reliable cellular tools for combination therapy strategies and the development of resistance countermeasures.

In Vivo Gene Editing
Utilizing AAV-CRISPR vectors to achieve tissue-specific gene editing in vivo, assessing the roles of target genes in tumor growth, metastasis, and remodeling of the immune microenvironment. The choice of AAV serotype (e.g., AAV8 for liver targeting, AAV9 for brain targeting) directly determines the in vivo editing efficacy.
